近年来, 随着石油资源的日益枯竭以及人们赖以生存环境的每况愈下, 寻找可以代替或部分代替石油及其附属产品的新资源成为当务之急。将对环境有负面影响的废弃物转化成新的能源和材料是摆脱依赖石油工业品现状和改善生态环境的一种有效手段[1-2]。甲壳素(Chitin)是地球上仅次于纤维素产量第二大的生物资源, 广泛存在于海洋无脊椎动物、昆虫的外壳中。生物的种类不同甲壳素的含量也有所不同, 其中以海洋中蟹、虾类动物的甲壳素含量最高[3-4]。我国的海岸线长度为1.8万km, 是一个海洋大国, 盛产蟹、虾类等节肢动物, 年产量达200万t, 如果将食用后的甲壳废弃物收集并提取甲壳素, 那将是总量十分可观的生物质资源。壳聚糖(Chitosan)是由甲壳素经脱乙酰化处理而得到的一种脱乙酰化衍生物, 化学名是(1, 4)-2-氨基-2-脱氧-β-D-葡萄糖, 其结构中含有羟基和氨基以及一些N-乙酰氨基等极性基团, 可方便的对其进行化学改性制备功能型壳聚糖基材料。由于壳聚糖具有良好的生物相容性及生物降解性, 并且可与许多生物大分子产生亲合性, 使其在环境保护、生物医药、农业等领域中具有广泛的应用前景, 尤其在水处理方面的应用研究更受到了极大的关注[5-8]。
废水中的铅[Pb(Ⅱ)]、镉[Cd (Ⅱ)]、汞[Hg(Ⅱ)]、镍[Ni(Ⅱ)]和铜[Cu(Ⅱ)]等重金属离子在较低浓度下就有很强的毒性, 并可以直接或间接通过食物链富集到人体内, 造成肝脏和肾脏损伤以及新陈代谢的紊乱[9-10]。因此, 水体中的重金属离子是必须进行处理的水体污染物。壳聚糖的比表面积(2~30 m2/g)远低于活性炭的比表面积(800~1 500 m2/g), 然而壳聚糖对重金属离子呈现了卓越的吸附性能, 这主要是因为壳聚糖分子中具有氨基和羟基等活性基团, 壳聚糖分子结构如图 1所示, 这些基团可通过络合作用, 离子交换或形成离子对等方式与金属离子结合, 进而达到吸附金属离子的目的[11-13]。本文从物理改性壳聚糖和化学改性壳聚糖两类方向出发, 重点介绍壳聚糖及其衍生物对水体中金属离子移除的研究进展以及壳聚糖基吸附材料发展现状。
1 物理改性壳聚糖吸附剂的吸附性能主要取决于其自身的尺寸大小、比表面积以及孔隙率。一般来讲, 壳聚糖基吸附材料的粒径越小, 比表面积就越大, 对金属离子的吸附量也相对较大[14-16]。Shafaei等[17]通过研磨和筛分制备了大小分别为0.177、0.5和1.19 mm三种不同粒径的球状壳聚糖吸附剂, 并对水体中的Hg(Ⅱ)进行吸附行为研究, 结果表明, 在pH=6, Hg(Ⅱ)浓度为3×10–3 mol/L时, 粒径大小为0.177 mm的样品对Hg(Ⅱ)的吸附量最大, 可达到1127.1 mg/g。Feorence和Battery指出, 有毒金属对水生生物的毒性顺序为: Hg > Ag > Cu > Cd > Zn > Pd > Cr > Ni > Co, 由此可见, Hg(Ⅱ)是在众多重金属污染物中最危险的一类, 这主要是因为水体中的微生物可将无机汞转化为毒性更强的甲基汞, 并在鱼类等生物体内富集并通过食物链进入人体, 引起各种疾病[11, 18]。Benavente等[19]从虾壳中提取出壳聚糖, 制备不同粒径的壳聚糖颗粒, 并对水体中的Hg(Ⅱ)进行了移除, 研究结果表明, 在pH=4时, 壳聚糖颗粒对Hg(Ⅱ)的最大吸附量为106.3 mg/g, 并通过氯化钠溶液将壳聚糖颗粒上吸附的Hg(Ⅱ)进行解吸以此达到Hg(Ⅱ)溶液的浓集和壳聚糖颗粒回收再利用的目的。氯离子浓度较高时可与金属离子形成稳定的络合物, 即HgCl4–、HgCl3–和HgCl2, 由于这些汞络合物与壳聚糖的氨基具有排斥作用, 进而使Hg(Ⅱ)从壳聚糖表面解吸。值的一提的是, 当制备的壳聚糖基材料的粒径过小时, 微粒在溶液中会聚集形成粒径较大的团簇, 使材料的吸附效率降低。由此可见, 在保证壳聚糖颗粒比表面积足够大的同时也应该充分考虑颗粒的分散性, 否则壳聚糖颗粒的吸附效率会有所下降。虽然有较好的吸附性能, 然而机械强度较低且酸性条件下不稳定的缺点使颗粒状壳聚糖在实际应用中受到极大的限制。Lopes等[20]通过溶剂蒸发的方法制备超薄的壳聚糖薄膜, 并研究了该薄膜在不同温度下对Hg(Ⅱ)的吸附能力。其研究结果表明, 随着温度的升高, Hg(Ⅱ)更易吸附到壳聚糖薄膜表面。
吸附剂对金属离子的吸附可分为以下三个过程: (1)金属离子扩散到吸附剂表面; (2)金属离子扩散到吸附剂内部孔隙; (3)金属离子与吸附位点发生相互作用。对于天然壳聚糖来说, 金属离子只能在其表面进行附着, 很难进入壳聚糖结构内部的孔隙当中, 导致壳聚糖的吸附效率大大降低。为了提高壳聚糖对金属离子的吸附效率, 可将其结构内部的孔隙数量和孔隙度增大, 实现金属离子在壳聚糖骨架内部的充分扩散[21-23]。利用物理和化学的方法可构筑壳聚糖内部多孔结构, 增加内部孔隙数量, 进而增加比表面积, 提高壳聚糖吸附效率。冷冻干燥技术是制备多孔材料, 增加材料比表面积的有效手段, 利用该技术所制备材料的形貌和多孔结构可通过原料壳聚糖分子质量、浓度以及冷冻温度和冷冻速率进行调控。Allouchea等[16]利用低温冷冻干燥技术, 制备了类似于三维海绵结构的多孔壳聚糖材料, 该材料具有大量孔径小于100 μmol/L的孔道(图 2), 对Hg(Ⅱ)最大吸附量可达350 mg/g。
壳聚糖基天然复合材料是两种以上的天然高分子材料进行物理混合所制备的具有特殊性能的新材料[16]。壳聚糖的酸性溶液可与其他天然高分子共混, 通过高分子链之间的氢键作用形成稳定的复合材料。Rahmi等[24]将壳聚糖酸性溶液和颗粒状纤维素混合制备壳聚糖基/纤维素复合物薄膜, 并应用到水体中Cd(Ⅱ)的移除。当pH=4时, 薄膜对Cd(Ⅱ)最大吸附量可达204.082 mg/g。此外, 复合物薄膜吸附的Cd(Ⅱ)可再次溶解于0.1 mol/L的硝酸溶液中, 同时实现壳聚糖/纤维素复合物薄膜的循环使用。壳聚糖与其他天然高分子共混后, 壳聚糖的结晶度明显下降, 机械强度大大提高, 这都为材料吸附效率的提高和材料自身的回收再利用提供了保障[25]。壳聚糖也可与合成型聚合物进行物理共混, 利用壳聚糖和聚合物分子链间的相互作用力制备机械强度高、吸附能力强的壳聚糖/聚合物复合吸附材料。聚乙烯醇(PVA)不仅是一种亲水性较强、机械强度较高、无毒, 生物相容性良好的合成型高分子, 更为重要的是聚乙烯醇分子链上的孤对电子可与重金属离子的价电子层相结合, 所以PVA对水体中的金属离子具有良好的吸附性能, 因此, PVA常用来与壳聚糖共混制备新型吸附材料并应用到水体中Pb(Ⅱ)、Fe(Ⅲ)、Ni(Ⅱ)、Cu(Ⅱ)等金属离子的移除[16, 22-23, 26-27]。
2 化学改性壳聚糖在不改变壳聚糖分子骨架的前提下, 通过对壳聚糖进行化学改性可显著提高壳聚糖的吸附效率并赋予新的性能。在壳聚糖分子骨架上接枝不同类型的官能团可增加材料吸附点的数量, 扩大吸附剂应用范围, 甚至可以使壳聚糖基吸附材料对特定金属离子的吸附具有选择性。因此, 化学改性壳聚糖在相关应用领域受到了研究者们的广泛重视。壳聚糖进行化学改性的目的主要有两点: (1)提高提金属离子的吸附性性能; (2)改善壳聚糖在水中或酸性介质中的溶解性。壳聚糖分子发生取代反应的位置包括按C-2位置上的氨基和C-3和C-6位置上的羟基, 其中氨基的活性较高。因此, 壳聚糖更易通过氨基基团进行化学改性[4, 6, 10]。化学改性壳聚糖主要分为两大类, 第一类是交联型壳聚糖, 第二类是接枝型壳聚糖, 其他类型的化学改性壳聚糖大都是由这两类衍生而来。
2.1 交联壳聚糖壳聚糖在酸性溶液中易发生质子化, 形成的游离态氨基会失去对重金属的配位能力, 使壳聚糖对重金属的移除效率降低。为了提高壳聚糖在酸性环境中对金属离子的移除效率, 研究人员利用双官能团或多官能团的化合物对壳聚糖进行分子内或分子间交联改性以此提高壳聚糖交联产物对重金属离子的吸附能力并改善其自身的机械强度。目前制备交联壳聚糖吸附材料所用的交联剂主要为戊二醛(GLA)[28-29]、环氧氯丙烷(EPI)[30-31]以及乙二醇二缩水甘油(EDGE)[32-34], 其中, GLA和EDGE主要通过壳聚糖分子链段上的氨基进行交联, 而EPI是通过羟基进行交联, 交联方式如图 3所示。
在水产养殖, 特别是池塘养殖生产, 为控制有毒有害藻类, 常把CuSO4用作杀虫剂和控制藻类生长, 但铜离子浓度过高时, 对养殖生物有很大的毒害性, 并容易在生物体内积累[35]。Ngah等[36]利用交联壳聚糖和原壳聚糖对水体中的Cu(Ⅱ)进行了吸附。研究结果表明, 当溶液pH=6时, 壳聚糖-GLA、壳聚糖-EPI、壳聚糖-EDGE和原壳聚糖对Cu(Ⅱ)的最大吸附量分别为59.67、62.47、45.94和80.71 mg/g。在弱酸性和中性条件下, 原壳聚糖的吸附量可能略大于交联壳聚糖的吸附量; 在酸性较强的溶液中, 原壳聚糖溶解于水中, 吸附量大大降低, 而交联壳聚糖可稳定存在于酸性介质中, 并保持吸附量不变甚至有所提高。Vieira和Beppu以EPI和GLA为交联剂制备交联壳聚糖材料, 并对水体中的Hg(Ⅱ)进行了吸附和解吸附行为研究, 同时与未改性天然壳聚糖比较。研究结果表明, 随着溶液pH的增大, 交联壳聚糖以及天然壳聚糖对Hg(Ⅱ)的最大吸附量增加。在相对较低的pH条件下, 壳聚糖分子上的氨基发生质子化, 与金属离子产生静电排斥, 进而降低壳聚糖对金属离子的吸附能力。上述材料对Hg(Ⅱ)吸附能力的大小为: GLA交联壳聚糖 > EPI交联壳聚糖≈壳聚糖[20, 37]。交联反应发生后, 壳聚糖结晶度减少, 分子链间的空间增大, 更多的氨基暴露在分子链表面, 使Hg(Ⅱ)与氨基接触的几率大大增加。此外, GLA交联壳聚糖中的亚胺键对水体中的Hg(Ⅱ)同样具有吸附作用, 综合上述两种原因, GLA交联壳聚糖对Hg(Ⅱ)的吸附能力大于其他两种壳聚糖材料。Sudhakar等[38]分别以戊二醛(G)和巴比妥-戊二醛(BG)为交联剂制备了两种交联壳聚糖材料, 即G-交联壳聚糖和BG-交联壳聚糖(图 4), 并应用到水体中Hg(Ⅱ)、CH3Hg、C6H5Hg的移除。交联剂戊二醛一般可连接两个壳聚糖分子, 而巴比妥-戊二醛交联剂理论上可连接三个壳聚糖分子, 因此以BG为交联剂制备的壳聚糖材料的孔隙率(50 µm)相比于G-交联壳聚糖的孔隙率(100 µm)较小, 在相同单位体积条件下比表面积相对较大。另外, 巴比妥也是金属离子很好的络合剂, 常用来检测水体中是否存在金属离子。综上两种原因, BG-交联壳聚糖对汞污染物的移除效率高于G-交联壳聚糖材料的移除效率。交联壳聚糖的交联度对其吸附性能有重要的影响。Sadek等[39]以谷氨酸为交联剂制备交联壳聚糖(GCs), 并利用该材料对废水中Cu(Ⅱ)和Ni(Ⅱ)进行了移除。谷氨酸的用量影响交联壳聚糖对金属离子的移除效率, 当谷氨酸用量从2.5 mmol增加到20 mmol, 金属离子的移除率从18.8 mg/g下降到17.4 mg/g, 交联剂用量增加引起交联度增大, 使交联壳聚糖的孔隙数量减少, 金属离子移除率下降。粉末状壳聚糖可与其他天然高分子颗粒交联制备纳米复合材料, 相比于单一天然高分子基吸附材料, 这类纳米复合材料的孔隙数量、孔隙度以及对金属离子的吸附量大大提高, 例如交联壳聚糖-海藻酸钠纳米复合材料(CS-AL-Nanocomposites)对水体中Hg(Ⅱ)的吸附量为666.0 mg/g, 而海藻酸钠对Hg(Ⅱ)的吸附量仅为32.0 mg/g。复合材料的吸附量同时也依赖于颗粒粒径, 即随着颗粒粒径的降低, 材料的吸附量迅速增大[21, 40-42]。
为了提高交联壳聚糖材料的吸附量, 相关研究者们设计研发了多重功能型交联剂, 其中最具代表性的是冠醚型交联剂。与传统的交联剂不同, 冠醚具有富电子中央空穴可通过配位键与金属离子相互作用, 进而提高交联壳聚糖的吸附量, 此外, 通过控制冠醚的空穴尺寸, 可选择性的络合金属离子[43-46]。Alsarra等[47]以4, 4x-二甲酰基二苯基-18-冠醚-6为交联剂制备交联壳聚糖(CCdBE)并用来吸附水体中的Hg(Ⅱ)和Pb(Ⅱ)(图 5)。CCdBE对金属离子的吸附量远远高于未交联壳聚糖。另外, 在Hg(Ⅱ)和Pb(Ⅱ)的混合体系中, CCdBE对Hg(Ⅱ)具有较高的选择性, 体系pH越高CCdBE对Hg(Ⅱ)的选择性就越强。含有氮、氧、磷、硫等原子的杂氮冠醚也被用于制备交联壳聚糖。依据软硬酸碱理论, 氮、磷、硫等原子的给电子能力均大于氧原子的给电子能力。因此, 相比于冠醚型交联壳聚糖, 杂氮冠醚型交联壳聚糖具有较优异的金属离子络合能力和对特定金属选择性。在天然高分子骨架上引入活性基团是最常见的制备功能型天然高分子的方法[48-49]。Wang等[46]将N-烯丙基苯并15-冠-5冠醚接枝到壳聚糖骨架上得到杂氮冠醚壳聚糖(CTSC), 而后以环氧氯丙烷为交联剂制备交联杂氮冠醚壳聚糖(CCTSC)。由活性基团行化学修饰后的CCTSC对水体中的Pd(Ⅱ)和Ag(Ⅰ)呈现了高度的选择性(表 1)。

交联壳聚糖衍生物在酸性和碱性溶液中具有较好的稳定性, 其热稳定性也强于壳聚糖。另外, 交联壳聚糖可快速的吸附水体中的金属离子, 并可通过一些方法对其进行回收再利用[46]。尽管如此, 交联壳聚糖无法在工业领域中广泛应用, 究其原因主要有三点: (1)壳聚糖交联后分子链的柔性下降, 使应用条件变高而范围变窄; (2)交联壳聚糖的吸附量降低。壳聚糖的交联反应主要发生在氨基官能团上, 导致对金属离子的络合能力有所降低; (3)交联壳聚糖的交联密度难于调控。交联密度过高, 容易引起吸附速率变慢, 吸附量下降; 交联密度低, 交联壳聚糖的耐酸性和机械强度差。(4)由于大部分交联剂具有一定毒性, 使交联壳聚糖的应用范围受到了一定的限制。
2.2 接枝壳聚糖接枝壳聚糖分为两大类型, 第一类是小分子基团修饰壳聚糖, 即将小分子活性官能团接枝到壳聚糖骨架上[50]。第二类是接枝共聚壳聚糖。无论是小分子基团修饰壳聚糖, 还是高分子接枝共聚, 其最终目的是使壳聚糖基材料同时具有小分子化合物和聚合物相关的物理、化学特性以及良好的生物降解性与生物相容性。
在第一类接枝壳聚糖材料中, 接枝到壳聚糖骨架上的官能团通常含有氮, 氧, 硫等元素, 例如天冬氨酸[51]、氮杂冠醚[43, 45, 52]、甘氨酸[53]、甲基丙烯酸酯[54], 丙烯酸丁酯[55], 乙二胺四乙酸[56]、羧甲基[57]、黄酸盐[18, 58], 硫脲[59]等。小分子基团修饰后, 壳聚糖材料的吸附点密度大大增加, 适用的pH范围扩大, 并且通过改变吸附机理或吸附点位置实现对特定金属离子的选择性吸附。关于小分子基团修饰壳聚糖的相关研究已有多篇综述对其进行过详尽论述, 在此不再做过多介绍。
接枝共聚法制备壳聚糖基吸附材料:所谓接枝共聚法是指在壳聚糖骨架上接枝功能型合成高分子(图 6), 提高壳聚糖复合材料的相关物理化学性质并扩大应用范围。在众多合成型高分子当中, 聚丙烯酸(PAA)常作为支链接枝到壳聚糖分子骨架上, 这主要是因为水溶性聚合物PAA不仅可以提高壳聚糖接枝共聚物溶解性, 并且PAA重复结构单元中的羧基可吸附金属离子, 增加壳聚糖接枝共聚物对金属离子的吸附量。通常PAA的分子链越长, 接枝率越大, 接枝共聚物对金属离子的吸附能力越强, 吸附量也越大[60]。为了降低接枝共聚物的成本并提高材料的重复使用率, Wang等[61]利用凹凸棒粘土替代部分PAA, 制备壳聚糖-g-聚丙烯酸/凹凸棒粘土(CTS-g- PAA/APT)复合材料, 该复合材料在进行五次吸附-解吸附过程后, 对Cu(Ⅱ)的吸附量无明显下降。聚丙烯酰胺[50, 62-63]、聚氮异丙基丙酰胺[64]、聚马来酸酐[65-66]、聚衣康酸[67]等其他亲水性聚合物也可接枝到壳聚糖分子链上, 所得共聚物对Cu(Ⅱ)、Cr(Ⅱ)、Fe(Ⅱ)等金属离子表现出良好的吸附能力。在壳聚糖骨架上接枝疏水性高分子可提高壳聚糖基共聚物的机械强度, 同时由于疏水性侧链的存在, 壳聚糖的吸水性下降, 而对重金属离子吸附能力提高。Rajesh等[68]通过微波辐射法制备壳聚糖-g-聚丙烯酸丁酯共聚物并对废水中的Cr(Ⅵ)进行了吸附, 共聚物对Cr(Ⅵ)最大吸附量为17.15 mg/g。聚乙烯亚胺[54, 69-70]、聚丙烯腈[71-73]、聚醋酸乙烯酯[74-75]、聚苯胺[76]、聚苯乙烯[77]等其他的疏水性聚合物也可接枝到壳聚糖分子链上, 所得共聚物对Cd (Ⅱ)、Pb(Ⅱ)、Cr(Ⅵ)等金属离子具有良好的吸附性。
3 结语和展望壳聚糖基吸附材料在水体中重金属污染治理领域得到了长足的发展, 其原料充足易得、无毒、廉价并可生物降解等优势使壳聚糖基功能吸附材料的研发工作充满活力。但是, 很大一部分研究成果只局限于实验室内的基础研究, 只针对某一单一低浓度金属溶液体系为研究对象, 而在实际工业重金属废水体系中, 物质的组成十分复杂, 存在众多因素需要考虑。换句话说, 基础研究成果未必适用于实际应用领域, 这使看似理想的研究结果变得索然无味。因此, 若想实现壳聚糖基吸附材料大规模的实际应用, 还有许多科学问题有待解决。
针对物理改性壳聚糖:物理改性是一种绿色、经济、有效的制备壳聚糖基材料的方法, 然而该方法制备的壳聚糖基吸附材料的机械强度低, 在实际应用中易发生材料的断裂, 破碎等问题, 并且材料本身的重复使用性差。另外, 壳聚糖基吸附剂应用条件较为苛刻, 在酸性较强的体系中吸附能力大大减弱。
针对交联壳聚糖:交联壳聚糖吸附剂的机械强度有所提高, 在酸性环境中也可保持较高的吸附量, 然而材料制备过程中所使用的大多数交联剂毒性较强, 很容易造成环境的二次污染。此外, 由于交联反应消耗了部分氨基使交联壳聚糖的吸附量有所下降。为了克服交联壳聚糖的上述缺陷, 研发廉价、无毒、多功能化的交联剂势在必行。
针对接枝壳聚糖:壳聚糖代替部分合成型高分子制备的接枝壳聚糖材料, 在保障材料吸附性能的同时还可以提高其自身的生物降解性, 生物相容性, 降低材料的制备成本, 然而壳聚糖接枝共聚物的热稳定性较差, 高分子链很容易从壳聚糖表面脱离。此外, 残留单体毒性、引发剂残留、应用范围较窄等问题有待急需解决。
此外, 针对实际废水中金属离子浓度高、种类繁多以及含有其他的无机和有机污染物等特点, 研究壳聚糖及其衍生物对高浓度重金属离子废水的吸附处理方法及处理复合污染体系的相关方法, 同时, 设计合成对特定金属离子具有选择性的壳聚糖衍生物, 在保护环境的同时对金属进行回收利用, 避对环境造成二次污染。
