塑料是日常生活中最为常见的有机合成高分子聚合材料, 以其价格低廉、质量轻、使用方便等特点而被广泛应用。常见的不可降解塑料种类繁多, 主要成分包括聚丙烯(PP)、聚乙烯(PE)、聚氯乙烯(PVC)、聚苯乙烯(PS)以及丙烯腈─丁二烯─苯乙烯共聚合物(ABS)等。随着近几十年的使用消耗, 特别是不可降解塑料的消耗, 诸多不良效应也随之而来。据统计, 2013年全球消费塑料制品高达3亿吨[1]。中国是世界上较大的塑料制品生产国和消费国之一, 每年大约产生近200万吨废弃塑料[2]。塑料作为垃圾的重要组成部分, 造成的危害几乎无处不在, 不仅对生态环境造成污染, 甚至危及人类健康。
每年约有10%的塑料垃圾, 通过各种途径进入海洋, 给海洋生态系统带来严重威胁, 而且这一数量级还在不断增长[3]。剩余的塑料则通过回收、焚烧、填埋、丢弃等途径污染陆地环境[4]。不管是留在陆地还是进入海洋, 部分塑料在各种条件的作用下最终变成小型塑料, 直至微塑料。微塑料的概念最早是由Thompson在2004年提出, 是指直径小于5 mm的纤维、颗粒、碎片等[5-6]。微塑料污染无处不在, 除了我们所熟悉的内地河流、沿海, 微塑料还广泛存在于高原湖泊、大洋深海中, 甚至是与人类生活息息相关的食品、化妆品中, 微塑料污染已经演变为全球性问题[7-8]。微塑料污染所导致的海洋生态环境破坏及其对人类健康的威胁已引起了人们的高度重视, 海洋微塑料逐渐成为全球学者研究的热点, 针对微塑料在海洋环境中的分布、对海洋生物群落与生态环境影响及其降解治理等的研究迫在眉睫[9-10]。
本文系统地总结了国内外微塑料污染的研究历程与现状, 探讨了海洋微塑料的来源、分布、污染现状以及未来的研究方向, 归纳了微塑料的相关生物降解研究, 以期为海洋微塑料对海洋生态环境影响研究提供基础, 为海洋微塑料污染的治理提供启示。
1 国外微塑料的研究历程自20世纪20年代, 研究人员就开始了对海洋塑料的关注[11]。Thompson等[6]最早提出“微塑料”一词, 但研究人员对其定义有不同的解释:目前普遍采用的是粒径小于5mm的塑料碎片[6, 12-13]; 另一种则认为碎片小于1 mm的为微塑料[14]。早在20世纪70年代, 海洋微塑料的相关研究已经开展, 如进行了微塑料附着的微生物和硅藻群落种类等的研究。此后, 人们研究了微塑料颗粒的形成、分布、浓度以及存在形式, 探讨了微塑料吸附的持久性有机污染物(POPs)所引起的生态问题以及缓解问题的方法。直到2001年, 研究发现北太平洋海域水体中微塑料颗粒的浓度高达109粒/m3, 微塑料问题才开始受到各国的普遍关注[15]。此后研究人员将微塑料的研究重点转移到塑料垃圾附着的微生物群落以及潜在的生态学效应上[16]。
2001年-2004年大洋塑料垃圾带的发现让海洋塑料污染又重新引发热议。今天, 海洋塑料污染已经演变成了一个全球性环境问题[17]。迄今为止, 在太平洋[15, 18-19]、印度洋[20]、大西洋[21]、极地[22-23]和深海[24-25]都发现了塑料垃圾的存在。部分塑料经过物理、化学和生物过程造成分裂和体积减小形成显微塑料碎片。Browne等[14]研究发现世界各地的18个地点的海岸线都受到了海洋微塑料的污染, 涉及从赤道到极地的6个大陆。人们已经在非洲, 亚洲, 东南亚, 印度, 南非, 北美和欧洲发现了大量的微塑料颗粒物[26]。除海洋外, 人们在河口区[27]、河流[12, 28]、海峡[29]、内陆湖泊[30-31]、城市郊区[12]和其他地点[32]也都发现了微塑料的踪迹。由此可见, 微塑料的影响范围越来越广泛, 由此引发的环境问题也越来越突出。微塑料不仅影响藻类的光合作用[33], 引起水生生物消化系统[34]、呼吸系统[35]的物理损伤; 还会吸附环境中持久性有机污染物[36]和有机重金属[37]、释放自身携带或吸附有毒物质, 引起更严重的化学毒理伤害[5, 38-40]。
微塑料已经成为生物群体的一种新型栖息地。Zettler等[41]根据北大西洋塑料垃圾的研究, 提出了“塑料圈”的概念。“塑料圈”指塑料基质和塑料垃圾存在异养生物、自养生物和共生生物组成的生物群落的组合[41]。在风浪和海流的作用下, 塑料圈携带的入侵物种、有害藻类、有毒病原微生物和吸附的持久性有机污染物、有机重金属离子迁移并释放到新的环境中。不同的塑料圈对其所处的生态系统有着不同的影响[42], 塑料圈的提出让人们认识到微塑料污染对海洋生态系统影响的复杂性。
2 我国微塑料污染现状 2.1 微塑料污染形势虽然塑料材料的使用产生了巨大的社会效益, 但“塑性时代”却带来了新问题:塑料在环境中的积累[11]。在海洋环境中广泛存在、积累的纳米尺度微塑料碎屑最大浓度可达105粒/m3[43]。近年来微塑料已从近海走向远洋, 甚至进入深海[24]。塑料污染无处不在, 大到海洋的哺乳动物[44-47], 甚至是作为食物链顶端消费者的人[43, 48-50], 小到显微镜下的单胞藻类[51-52]、桡足类[53]; 从经济发达、人口众多的近海沿岸[24], 到位置偏远、人迹罕至的高原湖泊[7]、极地地区[22-23]; 再到与人们日常生活息息相关的食品[54-55]、化妆品[8]中, 都发现了塑料的踪迹。
同样, 中国也面临严峻的微塑料污染形势。2016年, 国家海洋局发布的《中国海洋环境质量公报》指出我国41个海域的海面漂浮垃圾和海滩垃圾中, 塑料垃圾的比例在70%以上, 约80%来自陆地[56]。这些海域的时空跨度非常大, 从2011年到2015年, 涵盖从北到南的中国四大海。除海洋污染外, 内陆水域的微塑料污染也越来越突出。我国研究人员发现南方水系均不同程度地受到了微塑料的污染。甚至连素有“世界第三极”之称的人类活动非常有限的青藏高原, 其7个湖泊采样点中有6个检测到了微塑料[7]。环境中积累的微塑料进入食物链, 危及食品安全, 目前已在啤酒、蜂蜜、食盐和贝类等多种食品中检测到了微塑料的存在[54-55, 57]。
可见, 微塑料不仅影响了海洋和陆地生态系统的健康和可持续发展; 还通过食物链传递和富集, 危及人类安全健康。越来越多微塑料污染的报道表明, 全面系统地了解和研究微塑料污染在我国海洋和淡水生态系统的时空分布规律已迫在眉睫。
2.2 中国沿海地区微塑料污染状况微塑料是积累在环境中的塑料碎屑的小碎片[58]。世界各地几乎每个海洋栖息地都发现了微塑料[10]。虽然中国关于海洋微塑料污染的研究刚刚起步, 但是取得了较为全面的数据。目前, 微塑料的污染调查范围涵盖了我国四大领海[59-64]以及香港[65-67]、台湾地区[68-69]; 调查种类包括海洋漂浮垃圾、海洋海底垃圾和海洋海岸垃圾以及塑料颗粒吸附POPs的污染状况[61, 65, 70]。调查表明我国从北到南的邻近海域已普遍受到微塑料污染(表 1):北达秦皇岛渤海边和大连黄海边[61], 中到东海海域[64], 南抵南海北部沿海[59]; 从海岸滩涂到地表海水再到海底沉积物[60-61, 63, 70]也均发现了微塑料的污染。同时, 孙承君等[71]根据相关文献整理了我国2011年-2015年我国近海海底垃圾中塑料垃圾的比例, 发现海底垃圾中塑料垃圾的比例在逐年升高, 由2011年的57%上升到2015年的87%。海底塑料垃圾的不断增长, 将加剧塑料污染对海洋生态的影响。另外, 研究人员分别对2008年与2017年香港地区海边的泳滩和后海湾、吐露港、青衣、维多利亚港的地表水及沉积物中塑料污染情况和塑料颗粒吸附POPs的污染状况进行了调查, 发现香港地区是塑料污染的热点地区, 高于国际平均值[65-67]。台湾地区的微塑料污染主要集中在台湾北部区域, 研究表明微塑料颗粒尺寸与数量之间呈负相关[68-69]。
可见, 中国沿海地区普遍受到海洋微塑料的污染, 总结为以下特征: (1)空间分布:微塑料空间分布广泛, 遍布中国所有海域及沿海区域; 垂直分布范围广, 从海滩、表层海水到海底沉积物都有涉及, 遍布整个海洋栖息地。(2)丰度:中国近海地区的微塑料丰度普遍较高, 部分海域超出了国际平均水平。(3)组成:微塑料组成多样, 其中聚乙烯与聚丙烯的比重最大。
2.3 中国陆地微塑料污染状况相对于海洋微塑料的研究, 淡水系统中微塑料的研究较少[31]。中国淡水微塑料的水平几乎未知[64]。由于河流和污水已被确定为海洋微塑料来源的主要途径, 内陆水域的微塑料污染开始受到越来越多的关注[30]。Zhao等[60, 64, 72]研究了长江口水域、椒江、瓯江、闽江水体以及陆地区域生态系统中微塑料的污染状况(表 1); Zhang等[7, 73-74]调查了三峡水库、其支流湘西河回水区以及西藏北部西陵盆地四个湖泊的湖岸沉积物中微塑料的发生情况和分布特征; 王元元等[75]研究了青岛胶州湾河口沉积物中的微塑料污染情况, 从墨水河、白沙河和大沽河三个站点沉积物样品中提取和分析了典型微塑料颗粒。

陆地水域微塑料污染主要特点总结为: (1)陆地水域生态系统的微塑料污染形势严峻, 从东部沿海河口到西部高原, 微塑料广泛分布; (2)河口区微塑料研究相对较多, 水库成为微塑料污染的主要区域; (3)陆地水域微塑料组成与海洋微塑料组成基本相同, 主要包括聚乙烯、聚丙烯、聚苯乙烯、聚氯乙烯等。但是, 相对海洋微塑料污染研究而言, 陆地水域微塑料研究仍比较少; 且陆地水域微塑料研究主要集中在调查方面, 评价微塑料对陆地生态系统的影响方面近乎空白。
2.4 微塑料的来源和去向微塑料的来源主要有两种: (1)初生微塑料:直接进入环境中的粒径小于5 mm的塑料产品; (2)次生微塑料:源自较大的塑料碎片, 经过物理、化学和生物过程造成分裂和体积减小而成[26]。
目前普遍认为海洋微塑料的来源主要归于次生微塑料, 但并未意识到微塑料潜在来源-初生微塑料正广泛的存在于人们的日常生活中。从家用洗衣机洗涤纤维衣服产生的废水, 到日用护理化妆品, 再到以微塑料作为载体的药物, 以及工业生产所涉及的钻井液和空气爆破介质, 甚至是某些农药产品如驱虫剂等, 都涉及到了微塑料[14, 76-82]。总的来说, 初生微塑料主要是指特定工厂或者家用产品直接产生的微观尺寸的塑料(直径通常为2~5 mm)。
次生微塑料的产生可以看成是在多种环境因素与聚合物性质综合作用下, 降低大塑料碎片结构的完整性, 形成微塑料的过程。暴露在阳光下的大塑料碎片, 由于强紫外线, 强氧化以及波浪、湍流引起的物理磨损, 变脆、变黄, 形成裂纹, 变成了较小尺寸的微塑料[13, 36, 76, 83]。随着时间的推移, 这些碎片淹没在地表水或深层环境中, 低温和弱紫外线导致分解缓慢; 但分解仍在持续, 直到微塑料的尺寸变得更小[76]。微塑料的增加, 导致了塑料在海洋环境中的丰度增加, 增加了其对生态环境的潜在影响。
无论是初生微塑料还是次生微塑料, 均可通过海上活动或陆上活动等不同途径进入到海洋环境中。如人们日常使用的化妆品及洗涤纤维服装产生的微塑料, 可以通过工业或生活排水系统进入水生环境, 即使含有微塑料的水体经过污水处理厂, 但由于其密度、尺寸和丰度等原因, 仍会大量存在于水体中, 最终通过径流进入海洋环境[76, 84]。微塑料还会通过暴风雨、下水道、风和水流等直接进入海洋环境[84-85]。污泥也可能是微塑料污染的另一个来源, 因为它比输送到生态系统的水流含有更多的微塑料[86-87]。此外, 微塑料还可以通过浮游动物的粪便进入海洋[88], 暴露在微塑料环境中的生物体容易摄食微塑料, 通过肠道的微塑料被包裹在粪便中随后排出, 排泄后粪便可能被更大的桡足类摄入。而且, 微塑料的尺寸(< 5 mm)和相关的低密度有助于通过水流进行远距离的传输和广泛分布。综上所述, 微塑料最终的去向是海洋环境:或在海洋水体环境中积累, 或沉积在海床沉积物中, 或被生物体摄取以及在食物链中传递富集。
2.5 微塑料的影响水体环境中大量积累的微塑料可能导致复杂的生态问题。由于微塑料颗粒的颜色、密度、形状、大小、电荷和丰度等因素与环境及生物的作用, 加大了人们对微塑料在海洋环境中的动态和影响的认知困难[43, 89]。传统的塑料污染包括占用土地空间、视觉污染、污染空气、污染水体、造成火灾隐患、滋生细菌和引起疾病; 在水体环境中还会影响航运安全以及缠绕生物、限制运动和摄食等。但是淡水和海洋环境中的微塑料的有害影响远不止于此。水体环境中微塑料丰度的增加必然导致生物利用度的增加, 随着微塑料摄入率的增加, 生物易将微塑料颗粒误认为食物摄入体内。微塑料被海洋生物摄入时会造成化学和物理伤害, 导致机械效应:如作为聚合物附着在外表面, 从而阻碍流动性和堵塞消化道; 也能导致化学效应:如炎症、肝脏应激、生长下降等[90]。当微塑料被食物链中的低营养级生物摄取后, 会沿着食物链富集并传递到较高营养级生物, 甚至影响到人类安全健康。此外, 一些微塑料颗粒从海水中吸收的或者自身含有的持久性有机污染物(如二氯二苯基三氯乙烷、多环芳烃、多氯联苯、多溴代二苯醚等)随着其在食物链中的传递释放到生物体内[91-92]。已有研究发现微塑料具有从水环境中吸收痕量重金属的能力, 这些物质可能会沿食物链运输, 被海洋生物体摄取并侵入到组织中[48]。
微塑料除了对海洋或淡水系统的生物造成物理化学伤害外, 还会直接和间接的影响到人类的生存发展。人类利用海洋获得食物或其他制品已有上千年的历史, 作为食物的来源, 需警惕微塑料可能造成的危害。研究发现, 进入餐桌的某些鱼类含有不同数量级的塑料, 地中海地区约有18%的顶级食肉动物, 如剑鱼(Xiphias gladius), 蓝鳍金枪鱼(Thunnus thymus)和长鳍金枪鱼(Thunus alalunga)受到不同程度的微塑料污染[91]。非生物海产品中存在的微塑料也可能导致海产品的污染并潜在转移给人类。Yang等[54]证明了食盐也受到微塑料的污染。而且, 随着微塑料携带的有毒物质的富集和转移, 最终会影响到人类的健康和生存[90, 92]。
3 微塑料的生物降解全球每年塑料产量标准已超过3亿吨[1]。世界各地的塑料生产、使用和丢弃, 使它正以每年4 000万吨的速度在环境中大量积累[93]。塑料废弃物已成为目前最严重的固体废物污染问题。目前针对“白色污染”带来的生态环境问题的解决措施仅停留在传统方法, 如堆积、填埋、焚烧以及回收利用等, 但这几种处理方式对环境造成了不可避免的二次污染, 尤其是塑料垃圾的不可降解性, 可对生态环境产生难以估量的影响。如何处理好塑料垃圾已成为环保的最大课题[4], 对聚烯烃类塑料生物降解的研究可为解决白色污染问题提供了启示和参考。
3.1 微生物的降解生物降解是指在一定的条件下, 一定的时间内环境中的高分子塑料在物理、化学或者生物的共同作用下, 高分子的主链发生断裂, 分子质量逐渐变小, 通过细菌、霉菌、藻类等微生物发生酶解、水解或者两者协同作用等, 最终成为微生物能够利用的单体或代谢为CO2和H2O[4]。
3.1.1 聚烯烃类塑料的降解聚烯烃类的塑料, 由于其分子质量大、疏水性强以及表面能低等因素, 导致其降解过程复杂且降解进度非常缓慢[94]。根据聚烯烃类塑料的分子特性、结构特征以及微生物降解机制, 需要对塑料进行预处理。预处理方法主要包括: UV辐射(光氧化)、热氧化和化学氧化[95-99]。预处理后塑料长链变成短小的支链、分子质量减小、亲水性强、表面能增强, 易于被微生物吸附、生长和分解利用。
目前对聚烯烃类塑料研究最多、机理比较清晰的是聚乙烯。生物降解聚乙烯过程中主要包含两种降解方式:氧化式生物降解(Oxo-biodegradation)和水合式生物降解(Hydro-biodegradation)[100]。两种聚乙烯的生物降解均可分为4个过程: 1、生物吸附、侵蚀塑料基质; 2、经过生物氧化或酶水解的作用, 长链变成短链; 3、短链的聚合键断裂, 形成脂肪酸; 4、微生物消耗脂肪酸, 产生CO2和H2O[101-102]。此外, 生物降解聚乙烯过程中发现了四种关键的氧化步骤, 即末端氧化、两端氧化、次末端氧化、末端过氧化[103-108]。
3.1.2 聚酯类塑料的降解常见的可生物降解的聚酯类塑料主要来源有两种:天然生物质和石油基。天然生物质一般是对淀粉、纤维素、甲壳素等原料进行改良或者由微生物分泌、植物贮存后合成的高分子化合物, 如构成可再生合成塑料的聚羟基脂肪酸酯(PHA)、聚乳酸(PLA)等。石油基是指由不可再生的石油原料合成的脂肪族聚酯塑料, 主要包括聚己内酯(PCL)、聚琥珀酸丁二醇酯(PBS)、聚乙烯醇(PVA)等[108]。聚酯类塑料的生物降解过程与聚烯烃类塑料的生物降解大体相同, 均是有机体被微生物侵蚀、破坏和吸收利用的过程。由于可降解聚酯类塑料本身的独特性质, 微生物更易于吸附在有机体上, 随后微生物分泌的降解酶附着于塑料底物上催化其水解成为寡聚体二聚物和单体, 最终水解产物由微生物转化为CO2和H2O[109-110]。
3.1.3 可降解微塑料的微生物种类微塑料的生物降解主要包括两种方式:生物物理沉降和生物化学降解。生物物理沉降是指浮游生物物吞噬或者吸附海水中的微塑料, 通过重力作用沉降到海底中, 或迁移到其他区域。例如在海洋食物网中扮演重要生态角色的箭虫、桡足类中的哲水蚤(Neocalanus cristatus)和磷虾(Euphausia pacifia)[111], 误将海水中的微塑料作为食物吞噬; 澳大利亚大堡礁附近的近海珊瑚每小时可消耗高达~50 μg/m2的微塑料[32]; 浮游硅藻群聚集分泌二氧化硅, 可吸附微塑料颗粒沉降到海底[112]; Bathochordaeus stygius幼虫从周围的水中过滤微塑料颗粒, 然后将微塑料包裹其粪便颗粒中, 或者粘贴在其由黏液形成的“房屋”上, 快速下沉到海底[113]。
生物化学降解是指附着在微塑料上的微生物与其发生生物化学作用, 将微塑料基质分解成可被利用的单体或者CO2和H2O。目前已筛选出来降解聚烯烃类和聚酯类塑料的菌株有几十种。以真菌为主, 真菌中又以霉菌居多。如降解聚烯烃类微塑料(主要为聚乙烯)的真菌有黑曲霉(Aspergillus niger)与腐皮镰孢霉菌(Fusarium solani), 细菌有蜡样芽孢杆菌(Bacillus cereus)与玫瑰红球菌(Rhodococcus rhodochrous)等; 来自土壤、海洋、堆肥、活性污泥等环境中的降解聚酯类微塑料的真菌有草酸青霉菌(Penicillium oxalicum)与拟层孔菌(Fomitopsis pinicola), 细菌有嗜麦芽窄食单胞菌YB-6(Stenotrophomonas maltophilia YB- 6)与假单胞菌DS04-T(Pseudomonas sp.DS04-T)等[101]。另外, 某些菌类对聚烯烃类和聚酯类塑料均可降解。例如, 烟曲霉菌(Aspergillus fumigatus)既可以降解聚乙烯, 又可以降解PHA; 假单胞菌属菌株(Pseudomonas sp.)可降解聚乙烯与PLA、PHA等。
3.2 降解塑料的生物酶生物降解塑料的关键步骤之一是细菌、霉菌、藻类等微生物的酶解、水解或两者协同作用。常见的降解聚乙烯的酶有漆酶(laccase)[114]、锰过氧化物酶(MnP)[115]、单/双加氧酶[105]、P450细胞色素氧化还原酶[107]、脂肪酸氧化酶类(酰基肉碱转移酶等)[116]。可降解聚酯类塑料的酶比较单一, 主要是解聚酶、几丁质酶、酯酶等[109, 117]。Shirakura等[118]研究了解聚酶的作用机制, 发现解聚酶具有使环性聚合物内部键水解的活性, 而且解聚是从羟基端开始的, 如PLA的解聚酶。迄今为止已报道的分离成功的具有降解PCL的酶大多数是酯酶。嗜热菌株Streptomyces thermoviolaceus sub sp. 76T-2产生的降解PCL的酶则为几丁质酶[119]。此外, 刘玲绯等[120]认为蛋白酶K具有降解PLA的能力。
4 未来的研究与展望微塑料污染已经成为一个全球性环境问题, 引起各国政府的高度关注, 均纷纷采取措施进行治理缓解。例如2014年, 美国等若干国家禁止在化妆品配方中使用微塑料[8]。中国早在2007年就下达了“限塑令”, 虽然在一定程度上缓解了塑料污染的蔓延, 但未能彻底的解决塑料污染问题。2016年, 国家海洋局发布的中国海洋环境质量公报也提出了对我国海洋及陆地水域环境中微塑料污染的监管需求[56]。章海波等[121]在中国科学院海岸科学与可持续发展专题中指出加强我国海洋及海岸环境中微塑料污染的监管是保护我国渔业资源和滨海生态环境, 服务国家海洋环境外交的必然要求。可见, 微塑料污染的解决仍需建立相关海洋政策和完善管理和技术体系, 加强环境保护和监控力度, 控制和减少微塑料向环境的输入; 还应增强人们的社会环保意识, 鼓励和支持可降解塑料的生产应用等。
海洋、淡水、沉积物中均发现了相同塑料聚合物组成的颗粒, 说明微塑料污染问题在时间空间上的分布相当广泛。但目前关于微塑料调查采样与测定方法多种多样, 没有统一的标准。对于海水样品、沉积物和水体中不同粒径大小的微塑料分离方法以及微塑料颗粒的鉴别, 应根据微塑料的化学组分、颜色、形状、大小等建立更加完善科学的采样和分析方法, 发展新的方法[122]。调查过程中, 应建立优化合理的微塑料监测站位, 为海域水质的时空变化趋势评价提供有力支撑[123], 还应充分考虑到塑料用途和丢弃模式, 提出微塑料调查工作的新主题和新途径。另外, 相对于海洋微塑料污染研究, 淡水系统微塑料的分布、组成、丰度以及影响研究仍较少, 应该加强对水库、偏远地区湖泊的微塑料调查。
海洋中微塑料的生态风险评估仍处于起步阶段, 就微塑料的研究仅仅停留在微塑料的分布、浓度、影响, 微塑料对海洋生物与海洋生态环境的影响评价仍较少[122]。因此, 应该深入探讨“塑料圈”在水体中的时空分布、迁移规律、附着生物的时空变化特点以及海洋微塑料的生物降解等。还应该重视微塑料及其复合物在食物网中的作用, 分析微塑料在分子、细胞、组织、器官以及整体的毒害作用, 阐明在个体、种群、群落以及整个生态系统的作用机制[124]。
