海洋中的一些生物在受到周围流场的扰动时, 可以通过体内的一系列化学反应将化学能转化为光能, 并发出可见光, 这些生物被称为海洋发光生物[1]。海洋发光生物分布广泛, 从近岸生态系统到外海生态系统, 从海洋上层到海洋深层都有分布[2]。海洋中发光生物种类繁多, 从低等的细菌, 到甲藻、腔肠动物、环节动物、软体动物、节肢动物, 乃至高等的脊椎动物, 都有发光物种[3-4]。在众多的海洋发光生物中, 发光甲藻是一类非常重要的类群。在世界范围内的很多海域, 甲藻是主要的发光生物类群[5-6]; 同时, 甲藻也是海洋浮游植物中唯一具有发光能力的类群[2, 7]。
甲藻的发光功能有很多可以被人类应用的地方, 比如甲藻发光强度的变化可以作为指示一个海区浮游生物生物量变化的指标, 甲藻发光强度的增强往往预示着该海区浮游生物生物量的升高[8]; 另外, 一些发光甲藻物种可以用来检测海水中重金属等污染物的生物毒性, 是快速、有效的环境评价载体[9]; 同时, 甲藻的发光特性还可以应用于海洋军事探测方面[10]。因此, 近年来发光甲藻得到了人们越来越多的关注[2, 7]。
1 已知的发光甲藻的种类甲藻在海洋中分布广泛, 是海洋中重要的浮游生物类群, 总物种数超过1 500种, 隶属于117个属[11]。尽管海洋中甲藻种类很多, 其中只有少数的甲藻物种是具有发光能力的。发光甲藻的确认是比较困难的。首先, 对于自然环境中的甲藻群落, 需要将各甲藻物种单独分离出来进行纯化培养; 其次, 即使是发光甲藻, 由于昼夜的演替或者甲藻本身生理状态的不良, 也会导致其在一段时期内不发光, 从而形成误判; 第三, 对于同一种甲藻物种, 不同株系之间也存在发光功能的差异, 即有的株系发光, 有的不发光[12]。因此, 尽管甲藻种类繁多, 目前已经确认的具有发光能力的甲藻只占少数。经过前人的研究, 目前已确认的具有发光能力的甲藻总计67种(表 1), 多数物种集中于亚历山大藻属(Alexandrium)、角藻属(Ceratium)、膝沟藻属(Gonyaulax)、原多甲藻属(Protoperidinium)和梨甲藻属(Pyrocystis)等。

甲藻是海洋上层水体的主要发光生物类群[7]。在北大西洋海域, 仅角藻属(Ceratium)和原多甲藻属(Protoperidinium)两个属的发光物种就可以贡献该海域总发光强度的90%左右[25]; 在挪威的维斯特福德海域附近, 这两个属的发光物种可以占到该海域总发光强度的96%[43]; 在阿拉伯海, 原多甲藻属(Protoperidinium)和梨甲藻属(Pyrocystis)两个属的发光物种, 贡献了该海域总发光强度的50%左右[44]; 在马尾藻海, 夜光梨甲藻(Pyrocystis noctiluca)一种物种就可以贡献该海域总发光强度的64%[45]。因此, 一个海域发光甲藻的发光强度, 很大程度上可以影响和决定该海域所有发光生物的总发光强度的大小。
发光甲藻在海洋中分布广泛, 不过一般在光照充分、营养盐充足的海域生物量较高, 如近岸海域、上升流海域或者真光层等[7], 这跟浮游植物在海洋中的分布趋势类似。一般来说, 如果海水环境适合浮游植物的生长, 其中的自养型甲藻也会迅速繁殖; 同时浮游植物中的一些饵料物种可以作为异养型甲藻的营养来源, 促进其生长。因此, 浮游植物生物量的升高往往会伴随着发光甲藻生物量的升高, 叶绿素a浓度高的地方, 发光甲藻也会比较集中[44, 46]。另外, 在一些浮游植物的藻华过程中, 一些发光甲藻的生物量也会迅速升高[25, 47]。发光甲藻与叶绿素a浓度的相关性在一些情况下也会不显著:一些异养型发光甲藻细胞内是不含叶绿素的, 如果这类甲藻物种在自然群落中占据优势地位, 即使发光强度较大, 海水中的叶绿素a浓度仍然会比较低[33]。
3 甲藻的发光机理甲藻的发光是由细胞内的闪光体(Scintillons)产生的[48]。闪光体是甲藻细胞内的一种嵌套于细胞液泡膜上的细胞器, 直径约0.5 μm[49], 内部含有荧光素、荧光素酶以及荧光素结合蛋白[50-51]。当甲藻生存的环境发生物理扰动时, 扰动会穿越甲藻的细胞壁并作用于液泡膜上, 使液泡膜和闪光体膜产生动作电位; 动作电位的产生导致闪光体内的pH降低, 从而激活了荧光素酶[52]; 在荧光素酶的作用下, 闪光体内的荧光素和氧气分子结合并氧化, 最终产生波长为474~476 nm的可见光[15, 53]。
甲藻闪光体的发光反应主要受闪光体内环境中pH变化的触发和调控, 这主要是由两方面原因决定的:首先, 荧光素酶独特的三级结构导致了pH的变化会改变荧光素酶的构象, 使其活性位点可以和荧光素结合[54]; 其次, 通常情况下荧光素会被荧光素结合蛋白绑定, 后者只有在pH降低情况下才会释放荧光素, 使其自由并可以与荧光素酶结合[55], 从而可以进行随后的氧化反应。甲藻的荧光素是一种四吡咯化合物(图 1), 化学分子结构与叶绿素相似, 不同之处在于后者所含的金属离子为镁离子[56]。有学者推断, 甲藻的荧光素和叶绿素之间可能存在互相转换行为, 即白天以叶绿素形态存在进行光合作用, 夜间则转换为荧光素形态进行发光反应[2], 不过这个观点目前尚未得到验证。
4 甲藻发光的影响因素甲藻的发光主要受三方面的影响, 分别是水体的物理扰动、甲藻自身的营养和生理状况以及昼夜的规律演替。
4.1 水体的物理扰动对甲藻发光的影响海水的物理扰动是触发甲藻细胞发光的最重要的外部因素[22]。一般来说, 海水的物理扰动越强, 甲藻的发光反应就越强烈, 发光强度也越大[57]。海水的剪切应力是表征海水扰动强度的一个重要参数。近年来, 很多研究致力于探索海水剪切应力强度和甲藻发光强度之间的关系[16, 57]。Maldonado等[57]发现, 当把多边舌甲藻(Lingulodinium polyedrum)暴露于库爱特流所产生的剪切应力作用下时, 剪切应力的数值越大, 多边舌甲藻(L. polyedrum)的发光强度越大; Latz等[20, 58-59]的研究也得到了类似的结果。除了扰动强度的大小, 海水扰动强度的变化速率也对甲藻发光有影响。Von Dassow等[60]发现, 当剪切应力变化范围相同时, 多边舌甲藻(L. polyedrum)在剪切应力增加速率快的实验组的发光强度明显高于增加速率缓慢的一组; Latz等[20]也有类似的发现。他们认为, 在扰动强度增加缓慢的海水环境中, 甲藻细胞会慢慢适应这种变化而不发光或发光不明显。在海洋中, 海水比较稳定状态下的运动所产生的扰动较小, 剪切应力往往低于甲藻发光反应的阈值, 这种情况下甲藻较少发光[20, 61]; 风浪、舰艇或游动生物所产生的扰动相对较大且强度增加迅速, 这种情况下甲藻才会有发光反应发生[62-63]。
不同甲藻物种对海水物理扰动的敏感程度存在差异, 具体表现为触发其发光的海水剪切应力的阈值不同[58]。Latz等[20]通过对4种发光甲藻梭状角藻(Ceratium fusus)、多刺角甲藻(Ceratocorys horrid)、多边舌甲藻(L. polyedrum)和纺锤梨甲藻(Pyrocystis fusiformis)的研究发现, 能够触发这四种甲藻发光的剪切应力阈值不同, 范围介于0.02到0.3 N m–2之间, 其中多刺角甲藻(C. horrida)最低, 其次为纺锤梨甲藻(P. fusiformis)和梭状角藻(C. fusus), 多边舌甲藻(L. polyedrum)的阈值最高。
4.2 甲藻自身的营养和生理状况甲藻的营养和生理状况会显著影响其发光能力。对于异养型甲藻, 当环境中缺少饵料或饵料不存在的情况下, 甲藻的发光能力会显著降低:在硅藻饵料供给缺乏时, 赫氏原多甲藻(Protoperidinium huberi)的发光强度明显降低; 如果停止硅藻饵料的供给, 该物种则完全失去发光能力[36]。在对叉分原多甲藻(Protoperidinium divergens)和厚甲原多甲藻(Protoperidinium crassipes)的研究中, 也发现了类似的现象[35]。对于自养型甲藻, 多边舌甲藻(L. polyedrum)在夜间的发光强度与其白天接受日照辐射的时长存在正相关性, 这表明其夜间的发光能力受白天光合作用的影响, 白天光合作用越充分, 夜间则会有越多的能量可以供甲藻发光使用[64]。甲藻的发光是一种非常消耗能量的行为, 足够的食物来源和能量贮存才可以保证其发光功能的正常运转[7]。
4.3 昼夜的规律演替很多甲藻物种存在发光能力的昼夜变化, 即白天发光能力很弱或完全消失, 夜间发光能力很强[36, 65]。Lapota等[33]发现梭状角藻(C. fusus)和钝角原多甲藻(Protoperidinium curtipes)两种物种在夜间的发光强度分别是白天的29倍和137倍; Tortorec等[17]在芬兰群岛海的一次亚历山大藻物种(Alexandrium ostenfeldii)水华中监测了甲藻发光强度的变化, 发现甲藻的发光强度在日落后开始增强, 午夜达到最大, 到早晨则完全消失。
关于甲藻发光能力的昼夜变化, 有学者认为跟甲藻细胞的内源性调节机制有关, 即经过长期的适应和进化, 甲藻的发光功能存在一个24 h的“生物钟”似的开启和关闭的机制[7]。Kelly等[66]将从自然海区采集的甲藻置于完全黑暗环境中培养数日, 发光能力仍然存在昼夜变化的规律, 并且变化周期为24 h; Biggley等对巴哈马麦甲藻(Pyrodinium bahamense)的研究[41]以及一些室内培养实验的研究[50, 67]都得到类似的结果。以上研究证实了甲藻细胞自身内源性调节的观点。另外, 也有学者认为甲藻发光能力的昼夜变化跟光抑制作用有关。这种现象最早是在室内对自养甲藻的培养实验中被发现的[68]; 随后, Sullivan等发现在梭状角藻(C. fusus)[69]、Li等发现在扁平原多甲藻(Protoperidinium depressum)[34]、Hamman等[70]发现在新月梨甲藻(Pyrocystis lunula)中都存在光抑制现象。
5 甲藻发光的生态学功能甲藻的发光是一种非常消耗能量的行为[7], 即便如此, 甲藻细胞对发光的能量分配依然优先于细胞自身的生长和繁殖[2]。从进化角度来讲, 这一行为的保留和存在必然具有生态学意义, 会给甲藻自身的生存和种群的延续带来优势。目前, 关于甲藻发光的生态学功能主要有两种假说:报警(Burglar alarm)假说和恐吓(Startle effect)假说。
5.1 报警(Burglar alarm)假说甲藻在遭遇浮游动物的摄食行为时, 感受到物理扰动后通过发光吸引可以摄食浮游动物的其他高营养级生物, 后者通过甲藻发出的可见光发现并捕捉浮游动物, 从而使甲藻免于被浮游动物摄食[7]。甲藻的发光行为确实提高了其他高营养级生物对甲藻摄食者的摄食效率: Abrahams等[71]发现棘鱼(Sticklebacks)在发光甲藻存在时对桡足类的摄食率明显高于在不发光甲藻存在时的摄食率; Mesinger等[72]发现当环境中存在发光甲藻时, 相对于仅有不发光甲藻的情况, 硬骨鱼对糠虾的摄食效率大幅提升; Fleisher等[39]的研究获得了类似的结果, 他们发现头足类可以利用甲藻的发光来寻找和捕获猎物。
5.2 恐吓(Startle effect)假说甲藻的发光可以对桡足类的摄食行为产生直接的影响。Buskey等[73]发现在遇到甲藻发光时, 桡足类Acartia hudsonica的游动速率明显增快, 远远快于正常情况下的游动速率。这种快速的游动可以被理解为桡足类受到甲藻发光的“恐吓”而产生的应激逃跑行为。由于在这种快速的游动过程中, 桡足类不能产生稳定的摄食流, 因此对甲藻的摄食将会减弱或暂停。Esaias等[74]研究了太平洋哲镖溞(Calanus pacificus)对4种发光能力不同的甲藻物种摄食的研究, 发现太平洋哲镖溞(C. pacificus)的摄食效率会随着甲藻物种发光能力的增强而降低; 其他的研究也得到了类似的结论[26, 75]。这些研究表明, 甲藻的发光行为可以通过“恐吓”而减小摄食者对它们的摄食效率。
6 发光甲藻的应用 6.1 军事追踪和探潜水中目标追踪和探测一直是海战中常用作战手段。早期探测以声学手段为主, 随着各国海军隐身技术的发展, 单纯依靠声学手段不能满足探测需求。随后, 更先进的光学探测技术如激光水下探测技术等得到开发, 同样的, 各国激光隐身材料的应用可以影响激光探测的精度和灵敏度。因此, 研究并发展新的水下目标感知方法, 使各种探测手段优势互补, 是适应未来高科技海战的需要[76]。
通过海洋中发光甲藻受水体扰动后的发光现象, 发现并定位海洋中的移动军事目标, 在海军的军事追踪和探潜中应用较大[77]。当水面舰船或水下潜艇等经过含有发光甲藻的海域时, 其所造成的流场扰动会刺激发光甲藻产生一段时间的持续发光, 并在航行区域留下明显的发光轨迹[78]。军方通过遥感探测等手段可以监测并发现可见光轨迹, 从而定位敌方军事目标[10]。该技术具体可应用于舰艇遥感探测、反潜作战、近海岸反蛙人预警等军事领域。
6.2 有害藻华的预警和监测近年来, 全球范围内近岸海域有害藻华频发, 其预警和监测工作日益受到重视。当前有害藻华的监测主要依靠航次现场调查和实验室显微镜镜检分析, 比较耗时耗力, 且有延迟性的缺陷; 卫星遥感技术可以实时对海区浮游植物生物量进行监测, 不过该技术受天气状况影响较大, 并且监测精准度不高[79]。2006年, Kim等[80]在韩国沿海成功建立了甲藻发光的实时监测系统来监测藻华的发生, 取得了良好的效果。如果海区出现富营养化状况, 各类浮游植物类群如硅藻、甲藻生物量增加的同时, 往往也伴随着一些发光甲藻物种生物量的增加。所以, 通过对这些发光甲藻发光强度变化的监测, 可以反映当地海域甲藻生物量和浮游植物总生物量的变化趋势[8]。另外, 目前已知的发光甲藻物种中, 很多是可以产生毒素的[2], 可以形成有害藻华[81-83]。所以, 通过发光甲藻发光强度的监测来对近海有害藻华进行预警和监测也是一种有效的手段[17]。
6.3 环境污染物毒性检测利用海洋浮游植物细胞来检测环境污染物的毒性程度是一种常用的技术手段。传统的浮游植物检测方法多以浮游植物细胞密度为指标来评价污染物的生物毒性, 该方法比较耗时费力, 并且不经济。当海水中存在重金属、多环芳烃和氨等有毒物质成分时, 发光甲藻的生理活性会降低, 其发光功能会受到抑制甚至消失[42, 84]。因此, 可以利用发光甲藻发光强度的变化, 来评测海水中各种污染物成分的毒性。这种方法具有操作简单、灵敏度高、快速经济且可实现现场检测等优点, 目前, 国外已有该检测方法相关标准的建立, 并得到了广泛应用[9, 40]。
7 展望发光甲藻的研究在国外开展较早, 目前已取得了比较多的进展[7]。国内该方面的研究起步较晚, 且研究很少。未来国内的相关研究可以从如下几方面展开:
(1) 在各海区进行生态学本底调查, 摸清我国各海域发光甲藻的物种组成及生物量的时空分布和变化规律; 结合物理、化学等调查手段, 掌握发光甲藻的物种组成和生物量的时空变化与环境因子之间的关系。
(2) 研究发光甲藻发光强度与环境因子之间的关系。针对特定海域的发光甲藻进行人工培养并开展受控实验, 基于环境因子、流场剪切应力强度、发光甲藻细胞丰度等要素, 研究其对发光甲藻发光强度的影响, 同时明确各发光甲藻物种的剪切应力的发光阈值。
(3) 发光甲藻的原位观测。通过浮标、潜标外加光学原位探测装置, 原位监测目标海域发光甲藻发光强度的动态变化, 结合现场调查手段对该海域有害藻华的发生进行预测。设置安全阈值, 研发预警模型, 最终实现有害藻华的自动化实时监测。
