贝类毒素问题一直是海洋环境和水产品安全研究领域关注的热点。在2014年以前, 全球几乎每年有2 000人左右因为食用鱼类和贝类而中毒[1]。我国贝类毒素问题也非常严峻, 食用毒素污染贝类引起人员中毒的事件时有发生。据报道, 近30年来我国约有4 000多人次因食用各种螺、贝和鱼类中毒, 造成80多人死亡[2]。由于我国公共卫生监管体系尚不十分健全, 这个数据应远低于实际发生数。
氮杂螺环酸是人们认识较晚的一类贝类毒素。1995年11月, 荷兰发生第一起因食用爱尔兰港养殖的紫贻贝(Mytilus edulis)造成人类氮杂螺环酸贝类毒素中毒的事件[3]。一开始人们误认为是腹泻性贝毒中毒, 因为两类毒素的中毒症状极其相似, 都会表现出呕吐、腹泻、恶心等症状[4]。但后来的研究发现, 二者的化学结构差异很大, 作用模式完全不同[5], 氮杂螺环酸毒素的毒性远大于腹泻性贝毒[6]。研究发现, 氮杂螺环酸是聚醚类化合物, 含有一个独特的6, 5, 6-三螺环、一个环胺结构和羧基。根据其结构特征, 人们将其命名为Azaspir acids(AZAs)。因最初人们误认为该毒素是由原多甲藻(Protoperidinium)产生的, 故中文将其翻译为原多甲藻酸。目前发现, 该类毒素广泛存在于欧洲、北美、亚洲以及南美等地区[7, 8], 因此该毒素已经成为欧盟、美国、加拿大等组织和国家重点关注的对象。2004年, 在联合国粮农组织(FAO)、世界卫生组织(WHO)和政府间海洋委员会在都柏林举行的贝类毒素会议上, AZA毒素被正式列入八大贝类生物毒素之一[7]。
1 氮杂螺环酸的化学结构、来源与生成 1.1 氮杂螺环酸的化学结构氮杂螺环酸是一类含有螺环的聚醚类化合物, 末端含有羧基, 呈酸性。其碳骨架由40个碳原子组成, 分子中有20个立体异构中心和9个环(图 1)[9]。根据AZA毒素结构中C39位点上官能团的差别, 可将氮杂螺环酸分为348型(C39位点上脱甲基)和362型(C39连接甲基)。AZA-1是最常见的AZA毒素类型, AZA-2和AZA-3分别是AZA-1的8-甲基和22-脱甲基衍生物, 三者在有毒贝类中最为常见。现已发现, 自然界中存在至少59种氮杂螺环酸类似物, 而且这个数字还在不断增加。鉴于该类毒素对人类健康存在很大威胁, 欧盟规定每1kg贝肉中氮杂螺环酸的含量不得超过160 μg[10]。
1.2 氮杂螺环酸的来源与生成James等[4]从原多甲藻Protoperidinium crassipes中检测出AZAs, 因此认为其是AZAs的毒源种类。但后来的研究表明, 异养的原多甲藻并非AZAs直接的毒源种类, 只是AZA毒素的载体[8]。目前认为, 产生氮杂螺环酸毒素的甲藻主要在环胺藻属(Azadinium), 包括具刺螺环藻(Azadinium spinosum)、腹孔环胺藻(A. poporum)和A. dexteroporum以及Amphidoma languida等[11-13], 但并非所有的环胺藻都会产生AZA毒素[13, 14]。不同种类的AZAs产毒藻产生的毒素成分存在一定差异。研究发现, 具刺螺环藻中可产生AZA-1、AZA-2和AZA-33[11, 14-16]。腹孔环胺藻可产生AZA-2、AZA-11、AZA-36、AZA-37、AZA-40和AZA-41 [16-18]。A. dexteroporum中AZAs含量较低, 主要有AZA-3、AZA-7、AZA-35、AZA-54和AZA-58 [19-20]。Am. languida可产生AZA-2、AZA-43、AZA-38和AZA-39[15]。不过, 同一种藻不同株的AZA毒素成分也可能不同[16-20], 可能还存在非产毒藻。实际上, 有些A. dexteroporum、腹孔环胺藻的藻株并未检测出AZA毒素[15-16]。腹孔环胺藻产生的AZA种类最多, 株间差异最大[12, 16, 21]。
AZA毒素在贝体内会经历复杂的代谢过程, 有些AZA衍生物就是AZA在贝体中代谢产生的, 有些AZA衍生物则在产毒藻和贝体内均有存在[22]。有研究显示, AZA-3至-12以及AZA-17, -19, -21, -23都是AZA-1和2代谢的产物, 其中AZA-17和AZA -19是主要代谢产物。但有些产毒藻可产生AZA-3和AZA-11等毒素[16, 19, 22]。另外, 加工过程也会造成毒素结构的变化, 可能会产生新的毒素。如贝体加热后, AZA-17、AZA-19、AZA-21、AZA-23分别会转化成AZA-3、AZA-6、AZA-4和AZA-9[9]。
关于AZA产毒藻的毒素生成机制的研究不多。对于具刺螺环藻而言, 其产生的毒素成分(AZA-1、AZA-2和AZA-33)非常稳定, 可能受基因调控, 但环境因素会显著影响其生成[23]。Jauffrais等[23]观察了不同环境因子(温度、盐度、光照、通风)和营养盐条件下具刺螺环藻的生长和产毒情况, 发现不同氮源(尿素、硝酸盐和铵盐)条件下, 具刺螺环藻的产毒无明显不同; 低于最适生长温度和通风的情况下, 具刺螺环藻的产毒量显著增加。Li等[24]以腹孔环胺藻为材料, 研究了氮磷营养盐变化对产毒藻生长及AZA生成的影响, 发现对数生长期单位藻细胞毒素的含量显著低于稳定期, 氮的浓度对腹孔环胺藻产AZA的影响不大。但磷的浓度变化可明显影响毒素的生成, 磷限制可促进毒素的生成[24]。
2 氮杂螺环酸的毒性与致毒机理 2.1 氮杂螺环酸的毒性氮杂螺环酸与腹泻性贝毒的中毒症状非常相似, 表现为恶心、呕吐、严重腹泻、胃痉挛等[25]。但不同AZA毒素的毒性不同, 目前欧盟等国际组织和地区仅对AZA-1、AZA-2和AZA-3有限量规定, 其毒性研究主要来自AZA-1。AZA-1、AZA-2和AZA-3对小鼠的口服LD50分别为443 μg/kg (95% CL: 350~ 561 μg/kg)、626 μg/kg (95% CL: 430~911 μg/kg)和875 μg/kg (95% CL: 757~1010 μg/kg) [26], 对小鼠的腹腔注射最小致死量分别为150 μg/kg、110 μg/kg和140 μg/kg[27]。
AZA的主要作用部位是胃肠道, 也可作用于淋巴结、肝脏、脾脏、胸腺和肺等器官。进入小鼠体内后24 h时, 毒素在胃中含量最高, 而在肠道、肝、小肠、肾、肺、大肠和心脏及脑组织中, 毒素呈含量降序分布[26]。Caco-2细胞(human colon adenocarcinoma cell line)实验证实, AZA-1可通过肠道屏障[28]。
AZA-1可引起小鼠肺、胃肠道、肝脏、淋巴组织(胸腺和脾)等多个器官组织的损伤, 表现出肝毒性、免疫毒性、心血管毒性和神经毒性等, 有导致肺部肿瘤发生的危险[26, 27, 29, 30], 但并未发现有明显的遗传毒性[31]。口服中等剂量的AZA-1(100~300 μg/kg)一般不会引起明显的中毒症状, 但口服高剂量的AZA-1(420~780 μg/kg)会导致小鼠剂量和时间依赖性的中毒症状, 如萎靡不振、运动减少等[30]。口服AZA-1最常见的病理变化是小肠上段衬里(lining surrounding the upper small intestine)的降解, 可看到小肠绒毛变短、隐窝变长、上皮层剥落等变化[30]。口服高浓度的AZA(500~700 μg/kg)可导致小鼠胸腺、脾脏和淋巴集结中T细胞和B细胞出现不同程度的坏死[3]。小鼠连续口服1或5 μg/kg的AZA-1, 可出现明显的胃肠道效应, 表明AZA-1对小鼠的最小可见损害作用水平(lowest observed adverse effect level, LOAEL)是1 μg/kg[30]。AZA-1中毒后, 小鼠的恢复非常缓慢, 3个月后胃和小肠糜烂和绒毛变短的情况仍然存在; 肺泡壁水肿、出血的症状甚至会持续56 d[29]。不同组织的恢复时间为:肝脏7 d, 淋巴10 d, 肺56 d, 胃12周[29]。小鼠腹腔注射AZA-1后, 可出现缓慢的进行性麻痹、呼吸困难和痉挛等症状, 表现出一定的神经毒性[32], 可导致胃和肝脏肿胀并伴随胸腺和脾脏重量减小, 引致肝细胞空泡形成和脂肪酸的积累、胰脏实质细胞固缩(parenchymal cell pyknosis)、胃糜烂触血等病理变化[30]。连续给大鼠注射低剂量的AZA-1, 在未观察其他器官损伤的情况下, 就可发现大鼠血压变化, 同时降低大鼠血浆中金属蛋白酶-1的水平, 抑制心脏胶原沉积, 并改变心肌的超微结构[33]。注射AZA-2毒素, 可致大鼠心律不齐、PR平台期延长, 并出现二级和三级心传导阻滞[34], 说明AZAs毒素存在明显的心血管毒性。连续给小鼠注射50和20 μg/kg AZA-1, 会导致肺部肿瘤出现[30]。
2.2 氮杂螺环酸的致毒机理氮杂螺环酸的致毒机制和分子靶点尚不清楚, 涉及细胞信号转导、细胞骨架、物质和能量代谢以及离子通道等。研究发现, AZA-1、AZA-2和AZA-3可调节人类淋巴细胞胞质Ca2+的水平, 增加胞内cAMP浓度[35]。AZA-1可增加小鼠皮层神经元乳酸脱氢酶的释放, 诱导核固缩, 刺激caspase-3的活性[36], 提示AZA-1可通过诱导细胞凋亡和坏死两种途径导致皮层神经元死亡。AZA-1可导致淋巴细胞、成神经母细胞瘤等细胞中F-肌动蛋白紊乱和细胞黏附减少, 诱导神经母细胞瘤和Caco-2肌动蛋白丝束的重组和黏着点的缺失, 上调神经母细胞瘤肌动蛋白相关蛋白3、膜联蛋白A2、桥粒联结蛋白、蛋白S100-A10和钙调素结合蛋白的表达, 提示细胞骨架可能是AZA的重要靶点[36, 37]。
氮杂螺环酸可显著影响涉及胆固醇和脂肪酸合成、金属硫蛋白和糖酵解的基因的表达[5]。研究发现, 在Caco-2细胞中, AZA-1可显著上调糖酵解中关键酶磷酸果糖激酶(phosphofructokinase, PFKP)、编码参与细胞摄取胆固醇的低密度脂蛋白受体基因的表达, 诱导AP-1应答, 上调缺氧诱导因子Egl nine homolog 1(EGLN1)和EGLN3表达[5]。AZA-1可显著增强神经母细胞瘤线粒体电子传递链、糖酵解和phosphofruktokinase-1关键酶的表达, 下调神经母细胞瘤低聚高尔基复合体5(oligomeric Golgi complex 5)、ras相关蛋白、mRNA剪接与加工以及囊泡运输蛋白的表达, 引起高尔基复合体的损毁和囊泡的积累, 并导致明显的、暂时性的ATP耗竭[37]。电镜观察显示, AZA-1可使Caco-2细胞内自噬体增加, 闭合蛋白不均一分布; 引起细胞核损伤, 使异染色质更加收缩[28]。加大AZA-1浓度, 还可导致线粒体损伤, 使其变黑, 嵴分化明显, 代偿性增加ATP合酶的活性[28]。这些结果提示AZA可对机体物质和能量代谢产生很大影响[37]。
最近有人提出, 氮杂螺环酸是hERG通道开放状态的阻断剂[38]。氮杂螺环酸可能会抑制其他类型的丝氨酸/苏氨酸PPs(PP2B, PP2C, PP4, PP5), 或者是另一种蛋白磷酸酶亚型, 如酪氨酸特异性磷酸酶、脂磷酸酶等[39], 但不会抑制激酶、G蛋白偶联受体(protein-coupled receptors, GPCRs)和肌动蛋白的聚合和解聚。Twiner等测定了40多种激酶和76种GPCRs, 并未发现AZAs对其产生抑制作用[30]。
3 氮杂螺环酸的检测方法 3.1 小鼠检测法小鼠生物试验法(mouse bioassay, MBA)和大鼠生物试验法(rat bioassay, RBA)检测氮杂螺环酸毒素曾在全球范围普遍使用[40]。通过给大鼠投喂疑似有问题的海产品, 观察16 h内大鼠身体是否出现腹泻等状况; 或者将疑似有问题海产品的提取物注射到小鼠腹腔内, 记录24 h或者48 h小鼠的死亡率。由于存在大量的假阳性和假阴性结果, 可信度不高, 同时又违背动物保护公约的相关规定, 开发新的AZAs检测方法便成为人们关注的焦点。
3.2 体外细胞分析法Sérandour等发现, 氮杂螺环酸毒素对人肝细胞系(HepG2)和小鼠成神经细胞瘤细胞系(neuro-2a)具有细胞毒性, 因此提出可根据亲脂类毒素对这两种细胞系的毒性作用检测氮杂螺环酸毒素[41, 42]。
另外, 有研究者尝试采用特制的基因芯片检测海产品中的氮杂螺环酸毒素。研究发现, 接触氮杂螺环酸毒素后Caco-2细胞中发生差异调节的17个基因中, 有5个基因对氮杂螺环酸毒素产生明显的信号, 其指纹有望用于AZA毒素的检测[43]。
3.3 液相色谱-串联质谱联用(Liquid Chromatograph-Mass Spectrometer, LC-MS)LC-MS技术在贝类毒素的分析检测方面得到了广泛应用。早在上世纪末, LC-MS/MS技术就已用于氮杂螺环酸毒素的分析[44-47]。LC-MS分析方法非常灵敏, 检出限为5~40 pg (S/N=3)[7], 最重要的是它所需要的样品量少, 能够分析鉴定毒素之间细微的差别以及一些新出现的结构。Lehane等[48]利用独特的母体离子和产物[M+H−H2O−C9H10O2R1R3]+建立了LC-MS3方法用于检测AZA, 这一方法不需要完整的色谱分辨, 一次检测仅需7 min左右; 利用优化的等度反相法(optimised isocratic reversed phase), 使用含0.05%三氟乙酸和0.004%乙酸铵的乙腈/水(46︰54, V/V)从贝体中将毒素提取出来, 使用离子阱质谱、API 3000三重四极杆串联质谱以及Agilent 1100系列仪器进行检测; MS3对每种AZA的[M+H]+和[M+H-H2O] +离子进行捕获和断裂, 达到检测目的。韩深等[49]建立了QuEChERS(Quick、Easy、Cheap、Effective、Rugged、Safe)净化技术结合超高效液相色谱-串联质谱法, 为食用贝类中AZA-1、AZA-2和AZA-3毒素的筛查提供了有效手段。
值得指出的是, 有研究者尝试用9-anthryldiazomethane (ADAM)将AZA毒素衍生化建立LC-FLD检测方法, 并取得成功, 为AZA毒素的检测提供了一种新的选择[50]。
3.4 免疫测定技术作为初筛方法, 免疫分析法因其简单、快速、稳定, 受到人们的广泛青睐[51]。目前已通过多克隆和/或单克隆抗体技术得到了AZA-1的相应抗体, 并建立了氮杂螺环酸酶联免疫吸附测定方法、免疫磁珠法和免疫微球测定法(microsphere-based immunoassay)[51-52]等。酶联免疫检测法的工作范围为0.45~8.6 ng/mL, 检出限为57 μg/kg组织。磁珠法的工作范围为120~ 2875 μg AZA-1 equiv./kg组织, 检出限为63 μg AZA-1 equiv./kg组织[51], 免疫微球测定法的检测范围为15~280 μg/kg组织[52], 均明显低于欧盟限量标准。
4 氮杂螺环酸在双壳贝类中的分布 4.1 氮杂螺环酸在不同贝类中的分布最初氮杂螺环酸是在紫贻贝(Mytilus edulis)中发现的, 后来在其他的双壳贝类中也陆续检出氮杂螺环酸, 如钝顶贻贝(Mytilus chilensis)、长牡蛎(Crassostrea gigas)、欧洲牡蛎(Ostrea edulis)、大扇贝(Pecten maximus)、紫扇贝(Argopecten purpuratus)、菲律宾蛤仔(Ruditapes phillipinarium)、鸡帘蛤(Chamelea gallina)、鸟尾蛤(Cerastoderma edule)、斧形尖峰蛤(Mesodesma donacium)、Mulinia edulis、鸟蛤(包括Cardium edule、Glycymeris glymeris)、斧蛤Donax trunculus、竹蛏(包括Solen vagina、Ensis arcuatus和Ensis siliqua)等[7, 15, 27, 53]。目前发现, AZA在贝体中的积累和代谢存在明显的种间差异[54, 55]。在所有贝类中, 贻贝积累的AZA毒素最多[56], 其代谢产生的AZA类似物也多于其他贝类[56]。事实上, 目前仅发现贻贝与人类氮杂螺环酸中毒有关[56]。不同贝类中AZA毒素的成分也不尽相同。有调查指出, AZA-1可能是贻贝中的主要毒素, AZA-2是扇贝中的主要毒素[55], 但不同海域贝类中毒素的成分存在较大差异[55]。
4.2 氮杂螺环酸在贝类不同组织中的分布与代谢氮杂螺环酸在贝类体内积累的时间可长达6~8个月以上, 自然净化速率非常慢。除了可在肝胰腺中积累外, AZA可遍及贝类全部组织。氮杂螺环酸在贝类不同组织中的积累不同, 且受染毒方式的影响[57]。有研究指出, 投喂AZA产毒藻, 毒素主要积累在贻贝的肝胰腺中[56, 58], 水体中溶解的AZA则主要积累在鳃组织[57]。不同组织中积累的AZA的成分也不同, 消化腺中的毒素主要为AZA-1和AZA-2, 鳃和其他组织则主要为AZA-1和AZA-17[57]。毒素在贝体内会经历复杂的代谢和生物转化过程, 包括氧化、羟基化、羧基化和脱羧等[59]。研究发现, 贻贝体内AZA-1和AZA-2的生物转化速率非常快, 在接触到毒藻后3 h, 就可发现AZA-1和AZA-2转化为其他的AZA类似物[60]。通常鳃和其它组织中AZA-1和AZA-2代谢产物的比例高于消化腺[57], 提示不同组织中AZA的代谢转化速率和途经可能不同。
氮杂螺环酸可能会和贝体中某些蛋白(相对分子质量为21.8 ku和45.3 ku)结合, 这些蛋白与组织蛋白酶D、超氧化物歧化酶、谷胱甘肽S-转移酶Pi以及细菌鞭毛蛋白具有同源性。这可能是贝类中氮杂螺环酸排出缓慢的原因[61], 也是氮杂螺环酸提纯比较困难的原因[62]。
需要指出的是, 对贝体加热可因水分的丢失使贝体内的毒素的浓度增加, 同时造成毒素结构的变化, 但目前有关AZA在海产贝类中的限量标准仅针对生鲜贝类, 显然这是不够的[63], 限量标准应充分考虑到加热对贝体内AZA浓度及种类的影响, 加大对毒素种类的监管范围, 降低限值浓度。
4.3 氮杂螺环酸对贝类生存的影响氮杂螺环酸可严重影响贝类的摄食。研究发现, 喂食具刺螺环藻后, 贻贝清除率、滤水率和吸收率显著下降, 假粪生成显著增多, 同时可导致贻贝的消化腺小管变薄[60]。推测贻贝可能会通过减少滤水率和增加假粪生成减少毒素在体内的积累。不过, 喂食AZA产毒藻腹孔环胺藻似乎并不会造成贝类如贻贝和扇贝的氧化损伤[54]。
AZA是否会对贝类的繁殖造成影响, 目前尚未见报道。通过给日本青鳉(Oryzias latipes)胚胎注射AZA发现, AZA对胚胎具有致畸作用[64], 推测AZA对贝类胚胎也可能存在类似作用。
5 问题与展望综上, 尽管氮杂螺环酸是一类新的贝毒素, 但从我国沿海孢囊中分离培养得到了多种/株AZA产毒藻[13, 21], 同时姚建华等[65]从广州和大连的市售贝类样品中检测到AZA毒素。因此, 全面调查了解我国沿海贝类海产品中氮杂螺环酸的污染状况十分必要。由于AZA毒素的中毒症状与腹泻性贝毒非常相似, 因此极易被混淆为腹泻性贝毒[4], 这需要引起有关部门的警惕和关注。
另一方面, 氮杂螺环酸的致毒机理和在贝体内的代谢解毒机制尚不清楚, 弄清其代谢机制无疑对研究氮杂螺环酸的脱除具有重要意义, 而致毒机制的研究则有助于建立针对性的毒素中毒治疗方案, 因此组合利用转录组学、蛋白组学和代谢组学方法解析AZAs在贝体内的代谢机理, 采用同位素示踪结合现代分子生物学手段探讨产毒藻中氮杂螺环酸的生物合成机制应该是未来努力的方向。最后, 考虑到我国沿海已发现多种AZAs产毒藻, 也在多种贝类中检出这种毒素, 但贝类AZAs的来源与产毒藻的关系并不十分明确, 因此全面调查我国沿海AZAs产毒藻的分布状况, 明确贝类AZAs主要来源, 建立从产毒藻到染毒贝类的系统的氮杂螺环酸监控体系十分必要。
