微生物在全球海洋中是生物量和生产力的主要贡献者, 是生命和非生命系统联系的关键环节, 是生源要素循环的重要驱动力[1]。浮游细菌作为海洋微生物的重要组成部分, 在微生物食物环中发挥至关重要的作用, 其生产力相当于整个海洋初级生产力的10%~80%[2], 并承担海洋中75%~95%有机质的分解任务[3], 其中变形菌门作为海洋环境中最主要的浮游细菌类群, 在碳、氮等物质循环中发挥着重要作用。
SAR86是一类与γ-变形菌纲系统发育地位相近, 但生理和代谢功能极其多样的细菌类群, 1991年在马尾藻海首次发现其16S rRNA序列, 根据文库序列比对将其划归到γ-变形菌纲[4], 2000年被证明在全球海洋中广泛存在[5], 但至今还未得到纯培养菌株。海洋细菌SAR86在丰度上也占据重要地位, 部分海域中SAR86占到微生物细胞总数的10%, 是海洋环境中的优势微生物类群[6-7]。本文综述了海洋细菌SAR86遗传多样性、环境生态适应和地球化学功能等方面的研究进展, 并对未来研究趋势进行了展望。
1 海洋细菌SAR86系统发育多样性近年来, 随着对微生物多样性研究的深入, 系统发育多样性的研究逐渐占据了主导地位, 它不仅克服了传统微生物分离培养方法的限制, 而且提供了一种科学且简便的定量研究方法[8]。海洋细菌SAR86最早也是通过rRNA基因同源性分析方法被发现, 并证实其在海洋中普遍存在[5-6], 然而关于SAR86的系统发育分析, 相关研究报道仍然较少。Sabehi等[9]为了识别新的含有视紫红质(PRs)的基因片段, 构建了BAC文库, 将PCR扩增后得到的16S rDNA, ITS和23SrDNA的序列比对后构建系统进化树, 将SAR86类群分为Ⅰ型、Ⅱ型、Ⅲ型3个亚支, 其中Ⅲ型亚支又分为2个亚群(Ⅲa和Ⅲb), 从序列相似性及种的判定标准来看, SAR86类群可以划分为3~4个种, 或8~10个亚种级分类单元; 同时基于PRs的氨基酸序列构建了新的系统发育进化树, 发现SAR86Ⅰ型和Ⅱ型的亲缘性较远。Dupont等[10]利用海洋环境宏基因组测序信息及收集到的2个单细胞测序结果, 组合成SAR86的全基因组(包括A、B、C、D四个组分), 通过系统进化分析发现SAR86A、B、D属于Ⅰ型亚支, 而SAR86C属于Ⅱa亚支; 另外利用PR蛋白的保守序列重新构建系统进化树, 证实SAR86与γ-变形杆菌(特别是假单胞菌、海洋螺菌等)的同源性较高。
2 海洋细菌SAR86的生态分布世界海洋微生物普查计划(ICoMM)对全球海洋微生物焦磷酸测序数据和脂质数据的分析发现, 在所有环境中, 排名前20位的最丰富微生物类群包括γ-变形杆菌纲的10个类群[11], 其中就有SAR86类群。Giovannoni和Stingl的研究发现, 海洋中广泛分布的浮游细菌至少有20个主要类群, 包括SAR11和SAR86, 栖息的环境有表层海水、海洋真光层、近岸生态系统等[12], 因此海洋细菌SAR86可以占据海洋环境中的多种生态位[13], 并通过广泛参与海洋地球化学循环来影响海洋生态环境。
从大尺度水平分布上来看, SAR86类群在全球五大洋中均有分布(见表 1), 其中, 太平洋[14-22]、大西洋[7, 23-34]相关研究报道最多, 并多为优势物种[35]。从垂直空间分布来看, SAR86多分布在海水表层[15-18, 36-37], 偶有报道在深海混合水团中发现[26]; SAR86的垂直分布也随着海流等因素影响而发生季节性变化, 研究表明SAR86通常分布在水深100 m以上海域, 但在春季因水体对流混合作用, SAR86会分布至150 m水深处, 夏季又上升至50 m左右[38]。从全年季节变化来看, SAR86在夏季(6~8月)相对丰度最高, 春季(4~5月)与秋季(9~10月)的相对丰度均比冬季有所增加[38]。从栖息环境类型上来看, SAR86主要在表层海水中广泛分布[24, 29, 39], 在海山环境、厌氧沉积环境等特殊环境中未见相关报道; SAR86在近海丰度较远洋区高[40], 尤以内湾和河口区更高, 这也符合海洋细菌普遍的分布规律。

海洋细菌SAR86的分布同样也受环境因素影响, 研究表明盐度对SAR86影响不大, 无论在盐度低的淡水水域, 还是盐度高的海域, 以及盐度变化大的河口区域, SAR86都能很好的生存[41]; 然而SAR86受温度的影响比较明显[42], 研究发现SAR86B生活在温暖水域, 而SAR86C和SAR86D生活在较冷的沿海水域[10]; Crump等[43]对切萨皮克湾季节性缺氧水体的研究发现, SAR86丰度在缺氧水体的表层占据优势地位, 类似结果在南太平洋最小含氧区(OMZ)也被发现[17]。另外Sjöstedt等对马尾藻海的研究发现, SAR86丰度与亚硝酸盐浓度呈负相关[44]。Lanfranconi和Bosch的研究表明, 在短期柴油污染实验后, SAR86的种系数量减少[45]。
SAR86的分布和丰度与海洋生态系统中其他生物体也有着复杂的相关性, 报道最多的是浮游植物。早期对俄勒冈海岸硅藻爆发期间浮游细菌的研究发现, 优势种群SAR86的丰度并没有发生显著变化[46], 但后续研究表明SAR86会受到浮游植物爆发的影响; Thiele等[47]在南大西洋的铁施肥实验(简称LOHAFEX)中发现, 浮游植物爆发过程中检测到SAR86的序列数量和细胞总数都有所增加; El-Swais等[48]对西北大西洋浮游细菌季节性变化的研究表明, SAR86种群在秋季赤潮发生时期会出现; Korlević等[49]在亚得里亚海的研究也发现, SAR86类群在浮游植物爆发期被检测到; Yang等[50]在厦门海域研究发现, 赤潮能够刺激海水中SAR86种群丰度的增加; Li等[51]通过宏蛋白质组学技术发现, 甲藻赤潮爆发期间, SAR86是细菌群落的主要类群之一。
SAR86细菌的分布还与SAR11等相关微生物有着生态位互补或竞争关系, Tinta等[52]对亚得里亚海、地中海的近岸生态系统研究发现, 浮游植物爆发会刺激海水中SAR86细菌丰度增加, 并使SAR11种群的优势地位发生改变; SAR86的某些功能与SAR11类群具有明显的互补性而非竞争性, 这可能是SAR86能够在海洋环境中广泛存在并处于优势生态位的一个原因[53]; Aylward等[54]对转录模式的研究也证实无论在沿岸还是大洋生态系统, SAR86和SAR11之间都表现出特定的生态作用关系。另外, 也有研究表明海洋古菌MGII类群的丰度与SAR86有显著相关性[55]。
3 海洋细菌SAR86的基因组学研究基因组是指一个生物体中所有携带遗传信息的遗传物质总和, 开展海洋微生物基因组学研究不仅有助于了解海洋微生物生理生化、多样性特征及其在海洋生态系统中的地位, 而且对微生物功能基因鉴定和生物进化有着积极的促进作用。由于SAR86至今还无法进行纯培养, 所以Dupont等[10]利用宏基因组学技术进行测序和拼接, 装配了两个几乎完整的基因组SAR86A和B, 其中SAR86A基因组由41个重叠群组成, 序列总长为1.25Mbp; SAR86B基因组由31个拼接片段组成, 序列总长为1.70Mbp; 两个基因组分别含有1 316和1 712个开放阅读框。鉴于宏基因组组装可能产生的偏差, Dupont等还采取了单细胞测序方法对两个鉴定为SAR86的单细胞重新扩增测序, 得到了SAR86C和D, 总长度分别为750和925Kbp, 分别位于142和194个重叠群之间, 具体基因组信息如表 2所示。

Dupont等对组装后的SAR86基因组进行代谢预测和分析, 在维生素与辅助因子合成方面, SAR86A, C和D中缺少部分维生素生物合成途径中的相关基因, 包括B12、B6、生物素、泛酸、硫胺素和视黄醇; 在糖利用方面, SAR86A和B都可以进行糖酵解, SAR86B可以进行完全氧化的磷酸戊糖途径, 但产生的脂肪酶和酰基辅酶α合成酶含量较少; 在脂质β氧化方面, SAR86在β氧化的产物中, 酰基辅酶α脱氢酶、烯酰辅酶A水合酶含量比Ehhadh抗体和酮脂酰CoA硫解酶要高; 在抗生素抗性方面, SAR86中含有大环内酯类抗生素, 可以快速产生抑菌作用。另外Teira等[59]利用放射自显影和原位杂交技术, 研究了SAR86对碳酸氢盐、亮氨酸和淀粉三种物质的利用情况, 结果显示SAR86优先选择利用淀粉。Mou等[60]利用荧光免疫和16S rDNA技术研究了浮游海洋细菌在溶解有机碳(DOC)转移和利用中的作用, 发现SAR86可以吸收二甲巯基丙酸(DMSP)、甜菜碱(GlyB)、羟基苯甲酸(pHBA)和香草酸(VanA)等溶解有机碳的典型化学物组分。
综上所述, SAR86可能是需氧的化能异养型微生物, 基于蛋白质生成ATP, 可以通过一种特殊的扩大自身基因组的能力来降解脂质和碳水化合物, 随着更多的SAR86基因组被测序, 将有助于进一步探究其生物学和生态学特征。
4 海洋细菌SAR86在生物地球化学循环中的作用关于SAR86在海洋中的生态作用, 其细胞中含有的变形菌视紫质(PR)是研究热点。PR是一种质子泵型的视紫红质, 是迄今为止除叶绿体外, 人类在自然界生物体中发现的另一种能将光能转化为化学能的功能单位[1]。SAR86可利用PR将光能转化为细胞膜内外质子梯度化学势能, 直接产生ATP和能量[34, 39, 56], SAR86也利用PR的光合作用直接获取光能, 从而减少呼吸作用对有机碳的消耗, 并通过充氮降低氧气供应的方法对细胞呼吸作用进行抑制, 从而对海洋固碳及其他碳循环具有间接影响, 这种利用PR的新型光合作用使SAR86在海洋生态系统的物质能量循环中具有关键作用。Beja等[14]利用激光光解技术计算出环境原位上每一个SAR86细菌细胞内, 含有2.4×104个PR分子, 如此高的分子含量, 说明SAR86对于海洋碳循环和能量流动的作用不容忽视。另外Dupont等[61]发现, 高丰度的浮游细菌如SAR86、SAR11等在叶绿素含量最高的亚表层水体中, 表现出低水平的转录活性, 并且与硝酸盐代谢速率无关。
SAR86在微食物环中同样具有重要作用。微食物环是指海水中的溶解有机碳被细菌利用, 细菌被原生动物(鞭毛虫、纤毛虫)摄食, 原生动物被浮游动物摄食, 上述生物又产生溶解有机碳, 碳在这个环形回路中循环利用的过程[62], 在海洋生态系统的能量流动和物质循环中起着重要作用[63]。海水中的溶解有机物(DOM)主要来源是浮游生物, 部分DOM虽然不能被浮游动物吸收, 但却可以与颗粒性溶解有机物(POM)一起被细菌吸收并增加细菌生物量。由于SAR86基因组中缺少有关鞭毛形成、趋化和运动、胞外聚合物(EPS)生产、已知介导颗粒粘附等其它途径所需的基因, 这意味着SAR86是自由生活的浮游生物[10]。由于SAR86可被鞭毛虫、纤毛虫等原生动物所摄食, 而后者又被小型浮游动物所摄食[64], 因此说明SAR86不仅是分解者, 也是生产者, 并且在有机碳转换过程中起到重要作用。
SAR86等海洋微生物还能够降解海洋有机污染物[65], 对海洋环境保护和污染物治理有积极作用; 另有研究表明, SAR86可以利用藻类产生的有机物质进行生长和繁殖, 一定程度上可以缓解赤潮危害[66], 如加以利用可作为调控赤潮的有效手段[67]。
5 海洋细菌SAR86未来研究的重点方向和领域对于不可培养微生物的基础和应用研究是海洋微生物研究的核心方向, 鉴于海洋细菌SAR86的特点与重要性, 借助分子生态学的研究手段从基因、细胞、个体和群落水平上对SAR86进行基础研究, 并进一步优化改良培养技术和手段, 将会为人类利用海洋微生物以及保护海洋环境有至关重要的作用。未来对于SAR86的研究可从以下四个方面开展工作:
(1) 纯培养技术突破:随着SAR86宏基因组学和蛋白组学研究的深入, 更多的序列信息将有助于指导我们优化培养基组分(如利用KOMODO网站预测), 同时利用高通量稀释培养、流式细胞分选以及其他新技术, 用于SAR86纯培养菌株的分离。
(2) 生态分布模式解析:为深入理解SAR86的全球地理分布, 还需利用Python或Perl等工具, 对全球所有海洋和微生物的分子序列数据库和环境指标数据库进行整理和分析, 并构建科学的数学模型, 解析SAR86细菌不同生态型与分子序列及环境的关系, 重点关注缺氧、酸化等特殊海域的SAR86分布规律。
(3) 加强遗传进化研究:目前对于SAR86系统发育多样性的研究还非常有限, 对于遗传多样性、代谢多样性等的了解更为欠缺, 下一阶段不仅可以加强对SAR86与其他SAR家族细菌以及γ-变形菌的进化分析, 还可以利用KEEG、PICRUST等新的功能预测软件, 进一步加强遗传和代谢等功能多样性研究。
(4) 开展功能应用研究:鉴于SAR86在生物地球化学循环中的显著作用, 建议重点关注与污染物降解、重金属生物转化与修复、相关的功能基因研究, 也可深入研究SAR86的抗生素抗性基因合成及作用机制, 并将上述SAR86的特殊功能基因克隆转入工程菌, 推进小试、中试乃至工业化生产。
6 小结SAR86因其生物量丰度高、分布广泛, 逐渐成为海洋细菌的研究热点, 近年来借助分子生态学技术发展, 在系统发育和多样性、基因组功能和生物化学循环等方面取得了积极进展, 丰富了人类对于SAR细菌类群的认识, 但其生物学特性、代谢途径和生态功能等仍有诸多未知之处, 相信通过可培养技术发展和分子序列数据库完善, 对SAR86的基础研究会更加深入, 并有助于其资源开发和利用。
