异养营养对丛生盔形珊瑚代谢及共生藻光合生理的影响

摘要:本文以三亚鹿回头近岸海域内的优势种造礁石珊瑚——丛生盔形珊瑚(Galaxeafascicularis)为研究对象,以丰年虾(Artemia salina)无节幼体为饵料,研究了异养营养对丛生盔形珊瑚光合作用与呼吸代谢的影响。结果表明:异养营养的输入能够增加丛生盔形珊瑚的共生虫黄藻密度以及色素含量,并能够显著提升其光合作用与呼吸速率,然而单个虫黄藻的光合速率降低,这与虫黄藻密度增加引起的遮蔽效应有关。从营养角度来看,异养营养的输入能够使丛生盔形珊瑚有机碳的净积累速率提升约44.5%。本研究结果显示异养营养的输入能够促进丛生盔形珊瑚的光合与呼吸代谢,且这种情况下丛生盔形珊瑚的营养模式更为健康。

Abstract: Galaxea facicularis, which is dominant in Luhuitou fringing reef, was selected to study the effects of Artemia salina nauplii supplementation on the photosynthesis and respiratory rate of coral holobiont. The results showed that feeding could significantly improve the photosynthesis and respiration rate of G. fascicularis simultaneously, and the zooxanthellae density and pigment concentration of holobiont were increased as well. However, the net photosynthesis rate (PNet) and pigment concentration per zooxanthellae were decreased when food was supplied, which relates to the shadowing effect caused by the increases of symbiont density after feeding. Despite that, feeding could increase the PNet/RD ratio of G. fascicularis by approximately 44.5%, which indicates a higher organic carbon accumulation rate of coral holobiont. In summary, heterotrophic nutrition input could improve the photosynthesis and respiration rate of G. fascicularis, and the feeding mode of G. fascicularis is healthier when heterotrophic sources of nutrition are available.

造礁石珊瑚通常生活在寡营养的热带、亚热带浅水海域, 其构成的珊瑚礁生态系统是海洋中生物多样性最丰富、生产力最高的生态系统之一[1]。造礁石珊瑚是混合营养生物, 其营养方式较为复杂。一方面, 造礁石珊瑚通过与其共生虫黄藻的光合作用获得基本生命活动所需的能量(可占90%以上); 另一方面, 造礁石珊瑚还能够通过捕食异养获得能量与碳、氮等多种元素的补充[2]。目前, 人们普遍认为石珊瑚-虫黄藻共生体系是造礁石珊瑚仍然能够在营养盐限制的热带浅海繁荣生长的最主要原因[3], 却往往忽略了异养营养对造礁石珊瑚的重要贡献。事实上, 造礁石珊瑚能够异养利用的物质十分广泛, 主要包括各类浮游生物、溶解有机物和颗粒有机物等[2]

近40年来, 国内外已经开展了大量研究, 探讨了虫黄藻的光合作用对珊瑚代谢方面的作用[4-7], 也已证实了异养摄食能够引起珊瑚本身及其共生虫黄藻的部分生理指标(包括光合、呼吸以及骨骼和组织的生长等)发生变化[8-11]。造礁石珊瑚与其共生虫黄藻紧密相连, 其光合自养和摄食异养过程也密不可分, 无论通过光合作用亦或摄食作用获得的营养物质, 都会在珊瑚与虫黄藻两者之间不断流通[12], 但却鲜有研究同时探讨异养摄食对珊瑚自身及其共生虫黄藻的生理影响。目前仅有Rodrigues等和Houlbrèque等探讨了异养摄食对虫黄藻生理以及珊瑚生长的作用[9-10, 13-14]; 以及Lyndby等和Borell等探讨了异养摄食对共生体呼吸作用以及虫黄藻光合作用的影响[15-16]。因此, 为更好地解释异养营养的输入对造礁石珊瑚的影响, 需对异养营养输入情况下的珊瑚代谢与虫黄藻生理开展更深入的研究。

丛生盔形珊瑚(G. fascicularis)杯口大、触手长, 利于捕食多种浮游生物, 是三亚鹿回头海域的造礁石珊瑚优势种[17]。鹿回头海域靠近三亚河河口且处于人为活动较为密集的区域, 在陆源输入以及人为活动的影响下, 该海域夏季3 m水深处的光照强度仅有约100 μmol/(m2·s)[18], 远低于全球范围内珊瑚生活海域内的平均光强(约400 μmol/(m2·s))[19]。因此, 推测异养营养对于鹿回头海域内造礁石珊瑚的营养补充十分重要。丰年虾(A. salina)无节幼体富含多种营养成分, 作为生物饵料能够满足鱼类等许多物种的营养需求[20-21], 其个体大小约500 μm, 适合丛生盔形珊瑚摄食, 且已有研究证实了丛生盔形珊瑚对丰年虾无节幼体具有良好的摄食行为[22]

基于此, 本文以三亚鹿回头海域内的丛生盔形珊瑚为研究对象, 以丰年虾无节幼体为饵料, 研究了异养摄食对丛生盔形珊瑚光合作用与呼吸代谢及其共生藻光合生理的影响, 为探究环境胁迫下异养营养对造礁石珊瑚的影响提供基础。

1 材料与方法 1.1 珊瑚采集与暂养

2018年10月6日于鹿回头海域(18°12′N, 109°28′E) 2~3 m水深处, 随机采集三株丛生盔形珊瑚, 每株珊瑚均截取成直径约10 cm的块状。利用斜口钳, 将采集的珊瑚母体分离成单个珊瑚杯。在单珊瑚杯分离时, 通常选取珊瑚群体非边缘部分的成熟健康单杯, 单杯珊瑚的表面积大体相同(2.131±0.092 cm2)。使用阿隆发胶将单杯珊瑚固定于带有编号的陶瓷基座上, 并在实验站的室内珊瑚培育系统中暂养14 d, 以消除在分离过程中对珊瑚造成的生理损伤。

1.2 实验设计

实验设置摄食组(Fed)和未摄食组(Unfed)两个处理, 每个实验处理设置两个平行实验缸, 共计四个实验缸, 实验周期为28 d(以实验开始第一天为T0, 第7天为T1, 第14天为T2, 第21天为T3, 第28天为T4)。将暂养14 d的单杯丛生盔形珊瑚移入实验缸中, 每株珊瑚每缸随机选取3个单杯进行培养, 每个实验缸均含9个单杯丛生盔形珊瑚。实验于装有20 L沙滤海水的塑料缸中进行, 每隔一天更换50%的海水, 培养海水的盐度维持在约34‰。采用数显式温度控制器和钛合金加热棒(韦伯, 中国)共同控制水温(27±1℃), 各个实验缸中的海水通过一个小型潜水泵(350 L/h)混匀。实验体系由全光谱荧光水族灯(Giesemann)提供照明, 实验期间于每日7:00— 19:00提供光照, 19:00—次日7:00为黑暗模式, 控制实验缸底部的光合有效辐射约为100 μmol/(m2·s), 该光照强度接近鹿回头夏季3 m水深处的白天平均光强。

取1 g丰年虾卵置于1 L沙滤海水中曝气室温孵化36 h, 去壳, –20℃冷冻24 h后解冻, 去上清后得到浓缩的丰年虾无节幼体。以丰年虾无节幼体为饵料, 使用移液枪, 每日20:00对摄食组珊瑚进行投喂(每单杯约50个丰年虾无节幼体), 未摄食组珊瑚不做处理。每日喂食期间关闭全部潜水泵, 待所有珊瑚进食完毕后再开启潜水泵。

1.3 丛生盔形珊瑚光合与呼吸代谢的测定

本研究于T4进行了丛生盔形珊瑚净光合速率(PNet)、光增强呼吸(RL)以及暗呼吸速率(RD)的测定, 其中PNetRL的测量于白天进行, RD的测量于晚上熄灯一小时后进行。采用溶氧测量仪(Presens, OXY-4 mini)对上述参数进行测量:测量时温度为室温27℃, 且仪器于测量前经由两点经验值(27℃时的饱和空气海水和饱和Na2SO3海水)校准。其中在测量PNet时需提供一个100 μmol/(m2·s)的光照环境, PNet测量后去除光照并连续测量10 min, 此时计算得到的耗氧速率即为RL。上述参数均以单位时间内珊瑚产生或消耗的氧气量表示, 丛生盔形珊瑚每个单杯的光合和呼吸速率单位最终归一化到时间(nmol/min); 光合速率还可归一化到虫黄藻个数(pmol/(个·min)); 呼吸速率还可归一化到蛋白质量(mmol/(g·min))。总光合速率(PG)可通过PNet+RL计算得到。此外, 计算PNetRD之间的比值(PNet/RD), 即光合呼吸比, 以反映珊瑚共生体的自养能力[23]

1.4 虫黄藻生理状态及其密度与色素含量的测定

使用Diving-PAM (Walz), 于T0-T4测定单杯丛生盔形珊瑚的共生虫黄藻叶绿素荧光指数(Fv/Fm), 即虫黄藻的最大光化学量子产量[24]。此外, 于实验结束后对每个单杯丛生盔形珊瑚进行虫黄藻密度和色素含量的测定。

使用气枪将单杯珊瑚的组织与骨骼分离, 然后利用手持式匀浆器将组织进行充分研磨, 以分离出虫黄藻组分和宿主蛋白组分。采用血球计数板计数虫黄藻, BCA法试剂盒测量蛋白含量(南京建成), 最终虫黄藻单位归一化于蛋白质量(个/mg)。另取100 μL虫黄藻组分, 加入500 μL甲醇, 4℃过夜提取色素。之后吸取320 μl(光程1 cm), 用酶标仪测定480、510、630、664和750 nm的吸光值。色素含量最终单位归一到单杯或单个虫黄藻(μg或pg/个)。甲藻叶绿素a(Chl a)、叶绿素c(Chl c)和类胡萝卜素(Carotenoids)含量的计算参考Ritchie[25]和Strickland and Parsons[26]

1.5 数据处理与分析

本实验利用独立样本T检验(Independent-Samples T Test), 采用软件SPSS(version 21.0)对数据进行处理分析。本文中实验数据均以平均值±标准误差(mean±SE)表示。

2 实验结果 2.1 摄食对丛生盔形珊瑚光合与呼吸代谢的影响

摄食能够促进丛生盔形珊瑚的光合与呼吸代谢(图 1)并提升其光合呼吸比(图 2)。首先, 摄食能够显著提升丛生盔形珊瑚的PNet(未摄食组: 40.573± 3.288 nmol/min; 摄食组: 78.420±5.728 nmol/min, P < 0.001), 摄食后PNet提升约93.3%(图 1a); 当PNet归一到虫黄藻数目时, 摄食组珊瑚单个虫黄藻的产氧能力显著低于未摄食组, 降低约84.6%(未摄食组: 0.048± 0.007 pmol/(个·min); 摄食组: 0.026±0.002 pmol/(个·min), P=0.004)(图 1b)。其次, 摄食能够显著促进丛生盔形珊瑚的RD(未摄食组: 48.575±2.112 nmol/min; 摄食组: 65.26±2.325 nmol/min, P < 0.001), 摄食后RD提升约34.3%(图 1c); 当RD归一到蛋白含量时, 摄食组珊瑚每毫克蛋白的耗氧能力显著低于未摄食组, 降低约26.5%(未摄食组: 22.852±1.276 mmol/(g·min); 摄食组: 18.065±1.310 mmol/(g·min), P=0.013)(图 1d)。同时, 摄食也显著提升了其RL(未摄食组: 53.368± 3.125 nmol/min; 摄食组: 74.757±3.678 nmol/min, P < 0.001), 摄食后RL提升约40.1%(图 1e)。此外, 摄食能够显著提升丛生盔形珊瑚的PG(未摄食组: 93.941±3.220 nmol/min; 摄食组: 153.177±6.673 nmol O2 nmol/min, P < 0.001), 摄食后PG提升约63.1%(图 1f)。而通过对光合呼吸比(PNet/RD)的计算, 发现摄食能够显著促进丛生盔形珊瑚的光合呼吸比(未摄食组: 0.852±0.074;摄食组: 1.232±0.100, P=0.005), 摄食后PNet/RD比值提升约44.5%(图 2)。

2.2 摄食对丛生盔形珊瑚共生藻生理状态及其密度与色素含量的影响

摄食能够提高丛生盔形珊瑚的共生虫黄藻密度及其色素含量, 但对其最大光化学量子产量(Fv/Fm)无影响(图 3, 图 4)。首先, 摄食能够显著提升丛生盔形珊瑚的虫黄藻密度(未摄食组: 435 568.542± 36 860.591个/mg; 摄食组: 843 783.412±74 370.563个/mg, P=0.001), 摄食后虫黄藻密度升高约93.7%(图 3a), 而Fv/Fm并不会在摄食条件下发生显著改变(图 3b)。此外, 摄食能够提升丛生盔形珊瑚的色素含量, 其中叶绿素c和类胡萝卜素的结果显著(图 4a); 而相比于未摄食组, 摄食后每个虫黄藻的色素含量均出现了不同程度的下降(除类胡萝卜素外结果不显著) (图 4b), 这与图 1b中摄食造成每个虫黄藻的PNet显著下降相符合。

3 讨论

异养营养能够增加造礁石珊瑚体内的虫黄藻密度以及虫黄藻的叶绿素含量, 从而使其光合速率上升[27-31]。以柱状珊瑚(Stylophora pistillata)、鹿角杯形珊瑚(Pocillopora damicornis)和一种陀螺珊瑚(Turbinaria reniformis)为例, 异养营养能够提升这三种珊瑚的光合能力、虫黄藻密度和色素含量, 其最大净光合速率(PNetmax)和达到PNetmax的光照强度也有所增加[15, 31-33]。就珊瑚个体水平研究而言, 本文与之前研究的结果类似, 证实了异养营养能够提升丛生盔形珊瑚的光合速率、虫黄藻密度和叶绿素含量。这也表明了丛生盔形珊瑚能够充分利用异养摄食获取的能量, 供虫黄藻快速繁殖, 进而促进其光合速率。本文的结果也证实了造礁石珊瑚摄食异养对光合自养的正反馈效应(Positive feedback effect)[33-34]。此外, 本研究结果表明, 异养摄食不会影响丛生盔形珊瑚的Fv/Fm。而Borell等的研究显示在高温胁迫下, 异养营养的输入能够显著提升柱状珊瑚(S. pistillata)的Fv/Fm[35]Fv/Fm是指光系统Ⅱ(PSⅡ)的最大量子产量, 它反映了虫黄藻的潜在最大光合能力, 能够指示造礁石珊瑚共生藻的生理状态。本研究中Fv/Fm不发生变化只能说明在正常环境条件下, 异养营养的输入与否不会对丛生盔形珊瑚的PSⅡ产生影响。而对于环境胁迫条件下异养营养的输入对丛生盔形珊瑚Fv/Fm的影响, 则需开展进一步实验以证实。

结合三亚鹿回头海域光照条件较差的背景[18], 可推测当丛生盔形珊瑚在弱光条件下生活时, 必须通过异养摄食获得额外的能量补充, 一方面直接用于自身生长, 另一方面可被虫黄藻利用, 使之快速增殖, 从而在有限的光照条件下尽可能地提升光合速率。与本研究类似, Anthony等和Mie等研究证实了造礁石珊瑚能够通过两种营养方式之间的转化与互相弥补, 抵御悬浮颗粒物和高温等胁迫[36-37]。因此, 造礁石珊瑚在受到环境胁迫时, 可通过异养弥补自养和异养促进自养的营养模式维持珊瑚个体的正常生长。

从虫黄藻个体水平分析, 异养摄食反而抑制了单个虫黄藻的光合作用, 并且使得单个虫黄藻的色素含量下降, 即异养营养的输入降低了丛生盔形珊瑚单个虫黄藻的光合产氧速率。究其原因, 一方面, 异养摄食提供的额外能量促进了珊瑚体内虫黄藻的增殖, 使得虫黄藻的数目和密度大幅上升, 进而形成了遮蔽效应, 导致珊瑚组织深层虫黄藻获得的光照强度和CO2减少, 因此使单个虫黄藻的光合效率出现下降; 另一方面, 由于饥饿使未摄食组珊瑚无法提供多余能量以供虫黄藻增殖, 此时宿主珊瑚需要将虫黄藻控制在合适密度, 通过有限的虫黄藻充分利用光照, 以满足宿主珊瑚的正常生长。这种宿主珊瑚对虫黄藻数目的控制作用, 也体现在不同季节尺度和不同深度尺度上[38-39]。事实上, 造礁石珊瑚确实能够通过控制共生虫黄藻的密度及其色素含量以调节自身的能量利用与代谢模式, 从而应对环境变化。例如高温胁迫下的珊瑚白化可造成虫黄藻或色素的稀释, 从而使得珊瑚内部所接受的光强显著上升, 进而提高虫黄藻的生产力[40]。本研究发现未摄食组珊瑚在异养营养输入受到限制时, 可将虫黄藻的数目控制在较低水平, 说明丛生盔形珊瑚在异养营养输入或限制情况下, 可通过不同能量利用方式, 改变能量分配策略, 适应环境变化。

异养营养的输入不仅提升了丛生盔形珊瑚的光合能力, 也对其呼吸代谢产生了促进作用。相似地, 以柱状珊瑚(S. pistillata)和鹿角杯形珊瑚(P. damicornis)为对象的两项研究, 也发现了异养营养对造礁石珊瑚呼吸速率的促进作用[15-16]。异养营养的输入能够提升造礁石珊瑚自身蛋白质和脂类的含量[11, 41], 这可直接导致宿主代谢速率的提升; 此外, 虫黄藻数目的上升也意味着虫黄藻的总呼吸量上升, 尽管虫黄藻的呼吸占比不足共生体系的30%[42-43], 但也可导致造礁石珊瑚呼吸速率的整体上升。从另一角度看, 本实验中未摄食组丛生盔形珊瑚可通过降低自身代谢速率, 以弥补异养营养输入的限制以及整体光合产出的下降。但这种降低自身代谢速率的行为, 也可能伴随着蛋白质和脂质含量的下降等负面影响[16]。此外, 本研究中发现摄食组珊瑚单位蛋白的暗呼吸速率下降, 即异养营养的输入使丛生盔形珊瑚的平均暗呼吸速率下降, 可推测异养营养输入情况下, 丛生盔形珊瑚的平均能量利用效率更高、有机碳净积累速率更快。

目前大多数学者认为异养营养能够促进造礁石珊瑚的光合作用与生长速率。但有研究发现, 虽然异养营养可以刺激共生态海葵(Aiptasia pallida)的光合作用, 却并不能够促进其本身的生长, 这主要是由于共生藻自身新陈代谢需求的增加, 导致了营养物质从共生藻传输到宿主珊瑚的过程受到了抑制[44]。本研究通过计算光合呼吸比(PNet/RD)对这一问题进行了探讨。当PNet/RD≥1时, 说明共生体具备完全的自养能力, 即光照条件下, 共生藻光合作用固定的净有机碳, 完全可以支撑黑暗条件下呼吸作用的消耗; 当PNet/RD < 1时, 则说明珊瑚夜晚暗呼吸消耗的有机碳大于光照下共生藻光合作用净累积的量, 即珊瑚的内源能量物质出现净损耗[23]。本研究发现异养营养促进的光合作用能够使丛生盔形珊瑚个体获得更多的净有机碳积累, 这可能是由于种间差异造成的对异养营养利用能力的差异。事实上, 虫黄藻光合作用产生的碳大部分被用于维持珊瑚正常的生理代谢[5], 但其光合产物通常缺乏氮、磷等珊瑚生长所需的必要元素[45], 而通过对浮游生物、颗粒有机物和溶解有机物等的异养摄食, 造礁石珊瑚能够获得碳、氮、磷等多种元素的补充[36, 46-47]。即使本实验中由于虫黄藻密度的上升可导致虫黄藻自身耗能的增加, 但通过对丰年虫的异养摄食, 丛生盔形珊瑚能够获得充足的氮源补充, 并以此促进更多光合产物的合成, 进而使摄食后的丛生盔形珊瑚表现出更高的净有机碳积累能力[48]。因此, 推测异养营养的输入可能导致虫黄藻向宿主输送光合产物效率的降低[44], 但却能够提升被输送光合产物的质量(即光合产物碳氮比上升)。

4 结论

本研究以三亚鹿回头海域优势种造礁石珊瑚——丛生盔形珊瑚为研究对象, 探究了异养营养的输入对其代谢以及共生藻的生理学影响, 得到以下结论:异养营养的输入能够显著提升丛生盔形珊瑚的代谢速率(光合速率和呼吸速率), 并显著提升虫黄藻密度及色素含量。在异养营养输入的情况下, 虫黄藻的快速增殖导致的遮蔽效应, 使单个虫黄藻的光合产氧速率和色素含量出现不同程度的下降。整体而论, 异养营养的输入能够促进丛生盔形珊瑚有机碳的净积累, 即异养营养输入情况下丛生盔形珊瑚的营养模式更为健康。
