从淡水到海洋, 多样化的水生生态系统, 居住着超过30 700种鱼类, 物种多样性极其丰富[1]。作为一种低等脊椎动物, 鱼类在动物系统进化过程中起着承上启下的作用, 其复杂的生殖策略几乎涵盖了脊椎动物王国的所有繁殖类型, 越来越多鱼类作为模型来研究生殖调控机制[2]。在全球范围内, 鱼肉为人类提供优质蛋白质, 是继谷类和牛奶之后人类食物蛋白质的第三大来源。
然而, 近年来, 一些鱼类已经灭绝或濒临灭绝, 部分鱼类存在种质资源退化和遗传多样性降低, 还有一些经济鱼类仍然存在生长周期长、占用空间大、繁殖困难等问题。例如, 根据《濒危物种红色名录》、《中国生物多样性红色名录》记载, 稀棘平鲉(Sebastes paucispinis)、冬鳐(Leucoraja ocellata)、北方蓝鳍金枪鱼(Thunnus thynnus)等鱼类由于生存环境遭到破坏以及过度捕捞, 已被列为濒危或极度濒危物种, 加之繁殖周期较长、体型较大、养殖困难, 面临着灭绝危险。鱼类“借腹生子”技术的出现, 结合生殖细胞冷冻保存技术, 为解决上述问题提供了新的替代途径。所谓的“借腹生子”技术, 即生殖细胞移植技术(germ cell transplantation, GCT), 该技术基于生殖干细胞具有多向分化潜能的特性发展而来[3]。把鱼类生殖干细胞移植到同种或异种受体中, 供体细胞然后在受体内定居、分化和成熟, 最终产生供体来源的配子。虽然GCT在哺乳动物中的应用已经相对成熟, 但这项技术在进入21世纪后才被应用于鱼类[4-7]。在过去的十几年中, 科学家们不仅在种内移植上获得成功, 还发现种间移植也能使供体细胞继续发育并形成有功能的配子或后代。
这是一种前景广阔的生物技术, 既可以保护濒危物种, 又可以通过移植冷冻保存的生殖细胞来长期保存种质资源[8-10]。此外, 通过易于饲养和繁殖的小型鱼类代理繁殖生殖周期长、养殖空间大、繁殖力低下、或者人工条件下性腺不能发育的经济物种, 节省了劳动力和养殖成本[11-12]。还可对已有的经济品种进行性别、性状控制, 例如, 在不使用外源性激素诱导条件下产生YY超雄品系[13]或通过转染供体生殖细胞产生转基因个体[1]。该技术的应用将为鱼类新型繁育模式, 遗传改良以及种质资源的保存和高效开发利用开辟新途径。
1 鱼类生殖细胞移植发展生殖细胞移植技术是Brinster及其同事在1994年[14]首创的一项功能强大的生殖技术。它包括将供体生殖干细胞移植到代孕动物的性腺中, 以便从受体中快速且理论上不受限制地产生配子。近年来, 该技术在生殖医学、濒危遗传资源的保护和动物繁殖方面已经获得成功。此外, GCT对于理解生殖细胞发育调控和干细胞生物学[15]也有一定的意义。
对鱼类进行GCT, 首次是在鲑科(Salmonidae)鱼类上尝试开展。Takeuchi等[4, 16]从虹鳟(Oncorhynchus mykiss)孵化胚胎生殖嵴分离荧光标记的原始生殖细胞(primordial germ cells, PGCs), 并移植到同种异体或相近物种大马哈鱼(Oncorhynchus masou)孵化胚胎腹腔内, 最终产生供体来源的配子, 经过受精产生虹鳟后代。通过不断探索, 供体细胞已从PGCs逐渐扩展至其他生殖干细胞, 主要包括精原细胞(spermatogonia, SG)和卵原细胞(oogonia, OG)。由于SG和OG具有发育可塑性和雌雄双向分化的能力, 相继作为供体细胞在鲑科鱼类上被成功移植, 产生功能性精子和卵子[17-20]。更有趣的是, 使用三倍体或敲降法(dnd-MO)等绝育受体, 因降低或消除内源性生殖细胞产生的免疫排斥反应而极大地提高了移植的效率[21-22]。例如, 以三倍体大马哈鱼为受体, 从冷冻保存的整个虹鳟精巢中分离SG作为供体细胞进行移植, 成功产生功能性配子, 且全部为供体来源[23]。作为鱼类生殖移植鼻祖, 虹鳟鱼研究较为详尽, 结合PGCs、SG乃至整个精巢、卵巢的冷冻保存, 为鲑科鱼类遗传资源保存提供新的思路, 同时为其他鱼类生殖移植提供参考。
近年来, 鱼类GCT从鲑科扩展到其他种类, 包括鲤科[24-25](Cyprinid)、脂鲤科[26](Characidae)、怪颌鳉科[27-28] (Adrianichthyidae)、丽鱼科[29-30] (Cichlidae)、鲶科[31](Siluridae)、鲈科[32](Percoidea)、银汉鱼科[33-34](Atherinidae)、鲟科[8, 35-36] (Acipenseridae)等。对于受体, 其中使用性成熟的成鱼作为受体进行移植, 因节约了受体成长时间, 而具有迅速形成生殖系, 传播后代的优势。例如, Lacerda等[30]用经过白消安处理, 去除内源性生殖细胞的罗非鱼(Oreochromis niloticus)成鱼作为受体进行生殖移植, 在短的时间内成功产生了供体来源配子。从一个胚胎中获得的PGCs数量有限(20~30个), 而且很难捕获具有重要经济价值的新孵化的胚胎。与PGCs相比, 从未成熟或已成熟的鱼性腺中获得大量的SG相对容易, 且获得SG并进行移植的操作也更简单。因此, 近年的研究中更多的使用SG进行移植, 尤其是一些海洋鱼类, 包括石首鱼科[7, 11, 37](Sciaenidae), 鲭科[12, 38](Scombridae), 以及鲹科(Carangidae)[39-40]。比如, 将五条鰤(Seriola quinqueradiata)的SG移植到竹荚鱼(Trachurus japonicus)体内, 成功获得供体来源的配子和后代[40]。
2 鱼类生殖细胞移植类型鱼类生殖细胞移植有不同方式。根据供体细胞不同, 可分为PGCs移植, 囊胚细胞移植(内含PGCs), 精原细胞移植, 卵原细胞移植; 根据移植受体时期不同, 可分为囊胚期胚胎移植、孵化胚胎移植, 成鱼阶段移植; 根据移植部位不同, 可分为囊胚移植, 腹腔内移植, 成鱼生殖孔移植(图 1); 根据供受体亲缘关系, 可分为同种移植(表 1)和异种移植(表 2), 其中异种移植又可细分为属内移植, 属间移植, 科间移植, 目间移植等等。下面就供受体亲缘关系远近, 详述近十几年来鱼类GCT情况。

同种移植是鱼类GCT成功概率最高的一种类型。由于同一物种生殖细胞发生发育、生殖调控、生存环境高度一致, 使得供体细胞在受体内迁移、定居、增殖、分化、产生两性配子整个过程十分顺利。近年来, 由于近亲交配, 环境污染导致某些鱼类存在遗传资源退化, 遗传多样性降低问题。借助生殖细胞冷冻保存, 通过同种移植产生新的后代, 进行增殖放流, 可以维持群体遗传多样性。还有些经济鱼类繁殖周期较长, 通过产生转基因品系来改造鱼类性状需要几代筛选, 如果移植转染的供体细胞, 大大缩短了形成品系的时间。在水产养殖中, 某些鱼类如黄颡鱼(Pelteobagrus fulvidraco)、条石鲷(Oplegnathus fasciatus)、孔雀鱼(Poecilia reticulata)雄鱼比雌鱼具有生长优势或观赏价值。可通过同种移植进行性别控制, 在不使用外源性激素诱导性逆转情况下, 产生遗传上YY超雄群体。Okutsu等[13]把虹鳟的精原细胞移植到同种雌性受体, 使其产生XY卵子, 通过与野生XY精子杂交, 产生YY超雄个体。
2.2 生殖干细胞异种移植异种移植在代理养殖及种质资源保存方面有更广阔应用空间, 可以作为一种新的方式解决养殖过程中一些难题[41]。以太平洋蓝鳍金枪鱼(Thunnus orientalis)为例, 3~5年达到性成熟, 成熟的鱼可能超过100公斤[42-43]。如果把蓝鳍金枪鱼生殖干细胞移植到亲缘关系相近、体型更小、成熟更快、易于饲养物种如鲐鱼(Scomber japonicus), 其配子可能更容易、更迅速地产生。
对于异种移植, 属内移植、属间移植成功率最大, 其次科间移植、目间移植。Saito[24]等分离珍珠斑马鱼(Danio albolineatus)、金鱼(Carassius auratus)、泥鳅(Misgurnus anguillicaudatus)单个PGC, 并移植到吗啡啉敲降(dnd-MO)斑马鱼囊胚, 探索亲缘关系由近及远异种移植情况。结果发现移植珍珠斑马鱼的受体(属内移植)可获得功能性两性配子, 而移植金鱼(属间移植)和泥鳅(科间移植)只能获得功能性精子。泥鳅移植斑马鱼是目前唯一能产生供体来源配子及后代的科间移植报道, 虽然只获得了单一性别。
属内移植大多数都能产生供体来源的功能性精子, 如长鳍叉尾鮰(Ictalurus furcatus)精原细胞移植斑点叉尾鮰(Ictalurus punctatus)[44]; 有些鱼类还可以产生两性配子, 得到供体来源后代, 如银汉鱼(Odontesthes bonariensis)精原细胞移植牙汉鱼(Odontesthes hatcheri) [33], 还有些濒危鲟科鱼类, 性成熟周期时间8年以上, 利用精原细胞进行属内移植, 在受体内性腺内检测到供体细胞的定居, 如中华鲟(Acipenser sinensis)移植达氏鲟(Acipenser dabryanus)[8], 西伯利亚鲟(Acipenser baerii)移植小体鲟(Acipenser Ruthenus)[36]。根据目前研究, 发现属间移植在部分鱼种里面也获得成功, 如大西洋鲑(Salmo salar)精原细胞移植三倍体虹鳟孵化幼苗, 产生供体来源的雌雄配子, 通过受精, 得到供体来源后代[21]; 箕作黄姑鱼精巢生殖细胞通过成鱼生殖孔移植入雌性箕作黄姑鱼与雄性银姑鱼(Pennahia argentata)杂交不育体成鱼, 成功产生供体来源精子, 并得到供体来源后代[5]。
然而, 如果供受体亲缘关系较远, 如科间及科间以上水平, 除了在泥鳅和斑马鱼间移植获得供体来源的精子及后代外, 在其他物种上尚未突破。供体来源的生殖细胞常常在发育和成熟过程中被排异或吸收。Bar等[12]尝试通过五条鰤代理产生南方蓝鳍金枪鱼(Thunnus maccoyii)配子, 将分离金枪鱼精原细胞移植入五条鰤孵化幼苗的腹腔, 可检测到供体细胞迁移至生殖嵴, 但在移植后165天, 供体生殖细胞消失。同样, 在五条鰤移植箕作黄姑鱼的科间移植研究中, 移植的五条鰤精原细胞也未能发育成配子[11]。在亲缘关系更远的目间移植中, 分离日本鳗鲡(Anguilla japonica)的PGCs, 移植到斑马鱼胚胎, 只能观察到受体PGCs迁移至生殖嵴[45]。
在目前报道的异种移植的研究中, 无论亲缘关系较近的属内移植, 还是亲缘关系较远的科间移植、目间移植, 供体细胞都能成功迁移至受体生殖嵴, 并在发育中的性腺定居、增殖。这暗示了不同鱼类生殖细胞迁移机制和早期增殖调控方式较为保守, 或许与迁移过程中生殖细胞受化学趋化因子SDF-1a及G蛋白偶联受体CXCR4b共同作用有关[46]。对于科间移植、目间移植, 供体细胞在受体内诱导分化, 只能产生单一性别配子, 或者不能保持到配子产生现象。这可能是由于供受体亲缘关系较远, 性别分化, 配子成熟调控机制不同造成, 也可能与供受体整个生长环境差异较大有关, 包括温度、光照、饲养条件等。
3 问题与展望生殖细胞移植作为一种辅助生殖技术, 已经在多种鱼上进行研究, 与其他相关技术方法相结合, 可以有效地从濒危物种和有重要经济价值动物中产生功能性配子和后代。然而, 关于鱼类生殖干细胞的基本生物学认知仍然不完整, 这对GCT在鱼类中的应用造成了相当大的限制。进一步加深我们对生殖干细胞的了解, 将对鱼类GCT方面的进展做出重大贡献。
如何选择一个合适受体也是目前鱼类GCT面临的一个问题。为防止受体对外源细胞产生免疫排斥, 提高移植效率, 科学家们常会选择不育体作为受体, 包括三倍体、杂交不育体、白消安处理不育体、生殖基因敲除或敲降不育体。但是这些受体自身也各有不足之处, 例如, 通过生殖基因敲除或敲降方法获得不育体, 因其操作问题, 只适合模式鱼类, 不适合大型、生长周期长的经济鱼类; 利用化学药物白消安处理获得不育体, 因破坏了精巢的微环境, 使得移植效率降低; 三倍体雌鱼有些可以恢复生育能力, 不能达到完全不育, 雄性虽然不含有成熟精子, 但含有精原细胞、精母细胞、精子细胞, 充斥生殖上皮, 与移植供体细胞争夺空间; 杂交不育体性腺也存在未成熟早期生殖细胞, 获得杂交不育体同时受物种亲缘关系的限制。未来发展一种适合的不育体受体至关重要, 既要保证操作简便, 适用性强, 还要提高移植效率。通过不断探索, 突破远缘移植造成生殖壁垒, GCT技术将在代理养殖、资源保护上发挥更大作用。
