快速光响应曲线在底栖甲藻中的应用

摘要:
本研究为更准确的测定底栖甲藻对不同光背景的适应状况,选用了底栖甲藻Coolia tropicalis进行实验,比较了正序,反序和非序三种序列的快速光响应曲线(RLCs)测定6种不同光适应下的光合生理状态的结果,并对比了不同光化光时长对光合参数的影响。在正序RLCs中光曲线无法饱和,而反序和非序RLCs在高光化光下出现的饱和或光抑制。说明正序RLCs因序列内诱导的光适应导致其在高光部分的相对电子传递速率(rETR)偏高。光化光时长的增加加剧这种效应,正序的初始斜率(α)在30和60 s的与反序的无明显差异情况下,最大电子传递速率(rETRmax)显著高于反序的,使其半饱和光强(Ek)在这2个时长下出现与反序和非序的Ek相反的趋势。造成序列内光适应的主要原因是非光化学淬灭(NPQ)的影响,尤其在高光化光部分。使用反序RLC能够一定程度减少程序内光适应的干扰由于其降低了这部分的NPQ。考虑到反序的10 s时长相对正序的α存在高估,使用30 s时长的反序RLCs更能准确体现不同光背景下底栖甲藻的光适应情况。

Application of rapid light curves on benthic dinoflagellate

Abstract:
To better certify the photoacclimation of benthic dinoflagellate under different light history, we compared construction (up-, down- and non-sequence) and duration (10, 30 and 60 s) of rapid light curves to reflect the photosynthetic status of benthic dinoflagellate Coolia tropicalis under 6 gradients light history. RLCs with up-sequence failed to saturation whereas curves with down and non-sequence showed saturation or photoinhibition, which illustrates photoacclimation within up-sequence RLCs increased relative electron transport rate (rETR) under high actinic light. Prolonging duration would enhance the effect of photoacclimation. No significant different of maximal light use coefficient (α) and higher maximal rETR (rETRmax) of up-sequence RLCs were observed in in 30 and 60 s compared to of down-sequence RLCs, which caused its trends of light saturate coefficient (Ek) in these two durations totally different to other two RLCs. It is suggested that problems arose as a result of rapid fluorescence variations due to non-photochemical quench (NPQ), which was the dominant influence factor under parts of high actinic light. Thus, down-sequence RLCs reduced the photoacclimation effects within RLC due to the decreasing of NPQ in these parts. Considering α of down-sequence RLC with duration of 10 s overrated, using 30 s duration could better reflect the photoacclimation status of benthic dinoflagellate under different light history.
