基于鲎试剂生产的中国鲎血液质量监测方法研究

摘要:
中国鲎是我国生产鲎试剂的海洋药物动物,由于种群数量下降,2019年3月在世界自然保护联盟(IUCN)红色名录里的等级更新为濒危(EN)。为了研究中国鲎血液质量监测方法,有效保护、科学利用中国鲎资源,本实验选择9只中国鲎检测其血液中铜含量、血细胞数量、血浆蛋白含量、血淋巴蛋白含量等指标,并进行相关性分析。结果显示,中国鲎血液中铜含量与血细胞数相关性r=0.917,与血浆蛋白含量相关性r=0.995,与血淋巴蛋白含量相关性r=0.983,均呈现极显著相关性。中国鲎血液中铜含量与血细胞数的相关关系为Y1=6.14+0.86X(R2=0.840),与血浆蛋白含量的相关关系为Y2=2.08+0.78X(R2=0.989),与血淋巴蛋白含量的相关关系为Y3=1.87+0.62X(R2=0.966)。因此,中国鲎血液中铜含量不但能够直接反映血蓝蛋白的含量,而且可以反映血细胞数、血浆蛋白含量、血淋巴蛋白含量。因此,以中国鲎血液中铜含量作为监测血液质量的指标,具有简便、高效、快捷、省时、省力、省钱的特点。在鲎试剂生产中对中国鲎血液质量进行监测可以减少不当采血对中国鲎健康造成的影响,对中国鲎的利用与保护具有重要意义。

Study on blood quality monitoring method of Tachypleus tridentatus for TAL production

Abstract:
Tachypleus tridentatus is a Marine medicinal animal that produces Tachypleus Amebocyte Lysate in China. Due to the decrease of population, its classification on the IUCN Red List was updated to endangered (EN) in March 2019. In order to study the monitoring method of Tachypleus tridentatus blood quality, and to effectively protect and scientifically utilize the Tachypleus tridentatus resources, 9 Tachypleus tridentatus were selected to detect the blood copper content, blood cell number, plasma protein content, lymphatic protein content, and the correlation analysis was carried out. The results showed that the correlation of the copper content with blood cell number r = 0.917, the correlation with plasma protein content r = 0.995, the correlation with lymphatic protein content r = 0.983. There were obviously correlation. The relationship between Tachypleus tridentatus copper content and blood cell number is Y1 = 6.14 + 0.86 X (R2 = 0.840), The relationship between Tachypleus tridentatus copper content and plasma protein content is Y2 = 2.08 + 0.78 X (R2 = 0.989), The relationship between Tachypleus tridentatus copper content and lymphatic protein content is Y3 = 1.87 + 0.62 X (R2 = 0.966). Therefore, Tachypleus tridentatus blood copper content can not only directly reflect the hemocyanin content, but also reflect the blood cell number, plasma protein content, lymphatic protein content. Therefore, it is simple, efficient, fast, economical and saves time and manpower to use the copper content in the blood of Tachypleus tridentatus as the index to monitor the blood quality. Monitoring the blood quality of Tachypleus tridentatus during the production of Tachypleus Amebocyte Lysate can reduce the impact of improper blood collection on the health of Tachypleus tridentatus, and is of great significance to the utilization and protection of Tachypleus tridentatus.
