以连云港市瀚涌水产养殖公司提供的条斑紫菜丝状体绿斑病样本为供试材料,自样本中分离得到病原优势菌株ZCX-1,通过ZCX-1的16SRNA序列比对、构建系统进化树分析,初步确定ZCX-1为 Ruegeria arenilitoris。滤纸扩散法对其进行抗生素药敏试验分析,并研究不同温度、pH条件对抗生素抑菌效果的影响。结果显示,抑菌效果最好的是氨苄西林,在pH 8.0,30℃条件下,对ZCX-1 抑菌效果最好,抑菌圈直径38.7 mm;抑菌效果最差的是依诺沙星,最低值出现在pH 7.2,40℃条件下,抑菌圈直径仅10 mm,头孢噻吩抑菌圈直径为0 mm;说明ZCX-1菌株对氨苄西林最敏感,其次为头孢曲松、新霉素、依诺沙星、链霉素,对头孢噻吩具抗药性。试验结果为条斑紫菜丝状体绿斑病的管控防治提供理论参考。 |

Isolation, identification and drug sensitivity analysis of dominant bacteria causing green spot disease of Pyropia yezoensis protonema |

Using samples of green spot disease of Porphyra yezoensis protonema provided by Lianyungang Hanyong Aquaculture Co., Ltd. as the test material. The dominant pathogenic strain ZCX-1 was isolated, and the system evolution tree was constructed through the 16SRNA sequence alignment analysis. ZCX-1 was initially identified as Ruegeria arenilitoris. The filter paper diffusion method was used to analyze the antibiotic susceptibility test, and the influence of different temperature and pH conditions on the antibacterial effect of antibiotics was studied. The results showed that ampicillin had the best antibacterial effect on ZCX-1 under the conditions of pH 8.0 and 30℃, with a diameter of 38.7 mm of inhibition zone; enoxacin had the worst antibacterial effect, the lowest value appeared at pH 7.2 and 40℃, the diameter of inhibition zone was only 10 mm, and the inhibition zone diameter of cefalotin was 0 mm. It shows that the ZCX-1 strain is the most sensitive to ampicillin, followed by ceftriaxone, neomycin, enoxacin, and streptomycin, and is resistant to cefalotin. The test results provide a theoretical reference for the management and control of green spot disease of Porphyra yezoensis protonema. |
