于1998年7月从大连碧龙海珍品有限公司购得人工养殖的皱纹盘鲍,运用化学发光法研究在体外条件下利用6种活性氧清除剂SOD(超氧化物歧化酶)、CAT(过氧化氢酶)、苯甲酸钠和DMF(二甲基呋喃)、NBT(硝基四唑蓝)及金属螯合剂EDTA(乙二胺四乙酸)等试剂对皱纹盘鲍血细胞吞噬活动中化学发光的产生和影响。结果表明,活性氧清除剂对皱纹盘鲍血细胞的吞噬发光都有抑制作用,说明皱纹盘鲍血细胞在吞噬活动中能够释放出活性氧、行使杀菌功能;皱纹盘鲍血细胞释放的活性氧种类包括超氧阴离子(O2-)、过氧化氢(H2O2)、羟自由基(OH.)和单线态氧(1O2)。 |

Effects of scavengers superoxide dismutase (SOD), catalase (CAT), sodium benzoate, DMF of active oxygen species and some reagents, NBT, EDTA, on chemiluminescence of Haliotis discus Hannai (collected from Dalian in July 1998) hemocytes during phagocytosis were measured in vitro. In a manner resembling the respiratory burst of activated mammalian polymorphonuclear leukocytes, hemocytes of Haliotis discus Hannai generated active oxygen species during in vitro phagocytosis of yeast cells. The result that SOD can inhibit the chemiluminescence of Haliotis discus Hannai hemocytes during phagocytosis (P<0.01) suggests that Haliotis discus Hannai hemocytes can generate superoxide anion (O2-), and NBT can decrease the chemiluminescence because the intracellular blue formazan deposition was reduced by O2- released by Haliotis discus Hannai hemocytes (P<0.01). Catalase (CAT) can inhibit the chemiluminescence of Haliotis discus Hannai hemocytes (P<0.01) during phagocytosis (P<0.01), which suggests that Haliotis discus Hannai hemocytes can generate hydrogen peroxide (H2O2). Sodium benzoate can inhibit the chemiluminescence of Haliotis discus Hannai hemocytes during phagocytosis (P<0.01, P<0.05), which suggests that Haliotis discus Hannai hemocytes can generate Hydroxyl radicals (OH.). 2,5-dimethylfuran (DMF) can inhibit the chemiluminescence of Haliotis discus Hannai hemocytes during phagocytosis (P<0.01),which shows that Haliotis discus Hannai hemocytes can generate singlet oxygen (1O2), and EDTA can inhibit the chemiluminescence of Haliotis discus Hannai hemocytes during phagocytosis (P<0.01), which indicates that the divalent cation Ca2+ are required to initiate phagocytic and oxidative activity in Haliotis discus Hannai hemocytes. |
