利用组织切片及超薄切片电镜技术对患“乳化病”三疣梭子蟹的鳃、肝胰腺、步足肌肉、心肌和性腺5种组织进行病理学分析。组织病理显示:乳化病的病变性质为变质性炎症,主要病变部位在肝胰腺,其病灶表现为水变性、脂肪变性和细胞坏死等症状,步足肌和心肌表现出肌纤维断裂、浑浊等症状。超微病理显示:在肝胰腺和步足肌组织中有大量细菌,肝胰腺细胞结构损伤主要表现为微绒毛损伤脱落、线粒体坏死、溶酶体增多及脂肪滴变性,肌纤维断裂、溶解,血隙中血淋巴细胞受严重侵染。 |

ON HISTOPATHOLOGY AND ULTRASTRUCTURE OF PORTUNUS TRITUBERCULATUS SUFFERED FROM “EMULSION DISEASE” |

Portunus trituberculatus is an economically valued marine crab species. An acute and fatal disease spread in Zhejiang Province since 2002 in the species that its internal tissues are emulsified. In order to find out the pathogens and nosogenesis, this study was made. Its syndrome, biochemical pathology, histopathology and ultrapathology were observed and analyzed in our experiment.
Two strains of pathogenic bacteria were isolated from the suffered P. trituberculatus in local farms from 2002 to 2004 identified as Vibrio alginolyticus and Candida lusitaniae.
The infected Portunus trituberculatu samples were collected using cultural net in Zhoushan, Zhejiang Province. 5 parts of P. trituberculatus tissues (gill, hepatopancreas, muscle, heart & gonad) were observed under regular microscope and SEM. The histopathological changes were characterized by alternative inflammation. The pathological changes were serious in the hepatopancreas. The center of hepatopancreas show water-denaturation, fat-denaturation and cell decrease, as well as the muscle fiber damage. The findings in ultrastructure showed that a kind of bacterium was observed in hepatopancreas and muscle cells. The changes of organelle of the hepatopancreas cells were characterized by fallen-away and decreased microvilli, condensated mitochondria, increasing number of lysosome and necrosis, and hyperplasia of lipid drop lets. The pathogen could damage the fibre of muscle cell and lymphocyte. The result proved that Vibrio alginolyticus and Candida lusitaniae were the pathogenic bacteria of the disease in P. trituberculatus. Candida lusitaniae was responsible for the secondary infection, and trauma position was the primary transmission of the pathogentic bacteria. According to pathology changes, we analysed the causing mechanism and suggested the solution. |
