本文以脂肪酶ADM47601为目标,使用多种材料对其进行了固定化处理。实验表明,海藻酸钠固定化脂肪酶最佳条件为3%(w/v)海藻酸钠,1%CaCl2,每毫升海藻酸钠添加600U脂肪酶,最大固定化酶活力回收率为17.11%。海藻酸钠-明胶固定化脂肪酶最佳条件为1%海藻酸钠,1%明胶,0.5%CaCl2,0.25%戊二醛,每毫升海藻酸钠-明胶混合液添加600U脂肪酶,最大固定化酶活力回收率为54.45%。壳聚糖固定化脂肪酶最佳固定化条件为2%壳聚糖,10% NaOH,1%乙酸,0.25%戊二醛,每克壳聚糖微球添加840U脂肪酶,最大固定化酶活力回收率为87.06%。聚乙烯醇固定化脂肪酶最佳条件为7.5%聚乙烯醇,2%海藻酸钠,1.5%CaCl2,每毫升聚乙烯醇添加360U脂肪酶,最大固定化酶活力回收率为33.22%。对比四种不同固定化酶方法,脂肪酶活力回收率高度高低顺序为:壳聚糖吸附交联法>海藻酸钠明胶协同包埋法>聚乙烯醇-硼酸法>海藻酸钠包埋法。 |
