两种玉蟹超科蟹类线粒体基因组比较分析及其系统发育位置研究

摘要:
短尾类蟹系统分类研究日益依赖线粒体基因组证据,但玉蟹超科属级层面的分子资料仍较不足。为补充该类群基础数据,本研究对长形栗壳蟹和迅速长臂蟹完整线粒体基因组进行了测序、组装与注释,并比较基因组成、碱基组成、tRNA二级结构、氨基酸组成和同义密码子使用特征,同时基于16个物种13个蛋白编码基因采用贝叶斯推断和最大似然方法重建系统发育关系。结果显示,两物种线粒体基因组长度分别为15,717 bp和15,333 bp,均含37个标准线粒体基因,整体呈明显AT偏倚,AT含量分别为71.0%和70.3%。共识别出3种基因排列模式,两目标物种共享同一种衍生重排类型。系统发育结果支持长形栗壳蟹和迅速长臂蟹分别嵌套于各自所属支系,其系统位置与传统分类基本一致。本研究为玉蟹科补充了新的线粒体基因组资源,并为细化玉蟹超科内部系统关系提供了分子证据。

Comparative Analysis of the Mitochondrial Genomes of Two Leucosioid Crabs and Their Phylogenetic Position

Abstract:
Studies on the systematics of Brachyuran crabs increasingly rely on mitochondrial genome evidence; however, molecular data at the generic level within the superfamily Leucosioidea remain insufficient. To enrich the basic data for this group, the complete mitochondrial genomes of Arcania elongata and Myra celeris were sequenced, assembled, and annotated in this study. Gene composition, base composition, tRNA secondary structure, amino acid composition, and synonymous codon usage were compared, and phylogenetic relationships were reconstructed based on 13 protein-coding genes from 16 species using Bayesian inference and maximum likelihood methods. The results showed that the mitochondrial genomes of A. elongata and M. celeris were 15,717 bp and 15,333 bp in length, respectively, and both contained the standard set of 37 mitochondrial genes. Both mitogenomes exhibited a pronounced AT bias, with AT contents of 71.0% and 70.3%, respectively. Three gene arrangement patterns were identified, and the two target species shared the same derived rearrangement type. Phylogenetic analyses supported that A. elongata and M. celeris were nested within their respective lineages, and their systematic positions were generally consistent with traditional classification. This study provides new mitochondrial genome resources for Leucosiidae and offers molecular evidence for refining the internal phylogenetic relationships within Leucosioidea.
