在全球变暖的背景下, 海水温度已呈逐渐上升趋势[1]。以东海为例, 1945—2006年, 该海域的表层水温升高了0.9℃, 平均每年升高0.015℃[2]。另一方面, 中国近海区域有越来越多的大型滨海热电厂和核电厂相继建成、投产, 电厂冷源系统多采用直流冷却方式处理废弃能量, 产生大量含有废热的冷却水体(即温排水)[3-5], 温排水从排水口排出后扩散进入周围海域, 导致受纳水体的水温迅速升高, 并可能造成一定程度的热污染, 对海洋动物及生态环境造成潜在影响[6-7]。
水温对海洋动物的繁殖存活、生长和洄游等生命活动具有关键限制性作用, 直接影响早期生活史、种群结构、分布和数量变动[8-9], 而海洋动物对温度的适应性决定了其热耐受能力[10]。变温性海洋动物(如鱼、虾、蟹类)缺乏自身体温调节机制, 对水温变化响应显著; 相对于逃避能力较强的游泳动物, 多数甲壳类动物属于狭域性动物, 自身迁移能力较低下, 更易受温排水等现象影响, 一旦温升超出其热耐受能力, 会通过促使胚胎过早孵化、抑制生理代谢活动和损伤免疫功能等对繁殖孵化、早期生长和存活等产生不利影响[11-13]。因此, 开展温排水海域常见甲壳类动物的热耐受性研究对于评价近海热污染状况及其生态环境效应具有重要科学意义。
海洋动物热耐受性研究主要包括不同海洋动物的热耐受性、不同驯化水温下温升速率对海洋动物热耐受性、种群数量和分布的影响等。常用的研究方法有动态实验法与静态实验法。前者以最大临界温度(Critical thermal maximum, CTM) [14-15]、后者以24 h高起始致死温度(24 hour upper incipient lethal temperature, 24 h UILT50)为主要表征指标[16-17]。静态实验法能较直观地反映实验动物的死亡率与温度之间的关系, 但不能研究温升速率对实验动物耐热受性的影响[18]。动态实验法可以开展相关研究, 但缺乏设置实验温升速率的统一标准(10~1℃/48 h)[19]。目前相关研究一般采用二者相结合的方式, 通过最大程度模拟自然状态下的温升变化, 以期更客观地反映实验动物的热耐受性。
三门湾位于浙江省中部沿海, 为半封闭海湾(图 1), 年平均表层水温为18.3℃, 月平均水温为8.2 (2月份)~ 29.6℃(8月份), 是许多经济生物的重要产卵场和育幼场。三门湾核电厂规划容量为6台百万千瓦级压水堆核电机组, 预计其投入运营后, 将有大量温排水进入三门湾海域[20], 可能产生热效应, 对生物正常生命活动及生态环境健康可能有潜在影响。
本实验以三门湾海域重要经济虾蟹类脊尾白虾(Exopalaemon carinicauda)、口虾蛄(Oratosquilla oratoria)和日本

实验依照不同季节分别在三门湾海域附近的水产苗种场进行。实验水槽容积为150 L, 实验用水为三门湾自然海水, 盐度为23.3~28.1, pH为8.00~8.10。实验过程中充气, 自然光照条件。各水槽水温由精密控温仪(温控范围:0~90℃, 感温灵敏度:0.1℃, 韩国A-MI 211H)和钛加热棒(100~2000 W, 德国Armaturenbau)控制。实验过程中定期用水银温度计测量校对水槽内水温。
1.2.2 动态实验本实验在持续升温状态下测试实验动物的热耐受能力, 获取其最大临界温度。基于三门核电一期工程的数模计算和物模试验结果, 本实验在8℃(冬季)、15℃(春季)、22℃(秋季)和29℃(夏季)驯化水温下设置9个温升速率:0.5、1.0、2.0、3.0、4.0、6.0、9.0、12.0和15.0℃/h。每个温升速率下设置3个平行组。在每个实验水槽内放置各物种个体4~8尾以排除个体差异导致的不确定性。以驯化温度为实验起始温度, 按照设定的温升速率对水槽进行持续升温。当观察到某一物种有个体出现行为异常如突然急速无序运动、失去平衡或对外界刺激无反应等行为时, 以此为该个体的实验终止点并记录当前水温, 以实验物种各个体临界水温均值为该物种CTM[21], 直至观察记录到所有实验物种的CTM后结束实验。
1.2.3 静态实验本实验在不同恒定水温状态下进行, 测定其24 h高起始致死温度。在8℃(冬季)、15℃(春季)、22℃(秋季)和29℃(夏季)驯化水温条件下(表 1), 按照1~2℃温差设置梯度值, 每个温度值下设置3个平行组。从驯化水槽中选取各物种个体4~8尾, 将其暴露于各温度梯度下, 观察其24 h内实验动物行为反应和死亡情况。实验过程中, 在自然光照条件下保持充气。以受试实验动物个体停止运动、对触碰无应激反应等行为定义为个体死亡[17]。实验过程中及时观察、取出、记录死亡个体。
1.3 数据处理统计分析前, 分别对各处理组的数据进行正态分布检验(Shapiro-Wilk normality test)及方差齐性检验(Levene test)。在满足正态分布和方差齐性前提下进行方差分析(ANOVA)和组间差异的多重比较(Duncan test)。
动态实验数据处理利用双因素方差分析法(two- way ANOVA)检验动态实验中的驯化水温和温升速率对各物种CTM的影响显著性, 并对组间差异性进行多重比较。
静态实验数据处理过程中, 先统计每个温度组中各物种的死亡率, 再利用Probit回归分析求得它们在不同驯化水温下的24 hUILT50。利用two-way ANOVA检验物种和驯化水温对其24 h UILT50影响的显著性, 并对组间差异性进行多重比较。
相关统计分析在SPSS 22.0上进行, 差异显著性水平设置为0.05。
2 结果 2.1 实验动物的最大临界温度双因素方差分析结果表明, 驯化水温和温升速率对3种实验动物的CTM均有显著影响(P < 0.05, 表 2)。

驯化水温和温升速率均对脊尾白虾的CTM有显著影响, 但在不同驯化水温下表现出不同的变化趋势; 同时在同一温升速率下, CTM随驯化水温的升高显著增大(P < 0.05, 表 2, 图 2)。驯化水温8℃时, 9℃/h(33.3℃)的温升速率组的CTM显著低于其他组(P < 0.05), 而其他组别之间差异不显著(P > 0.05)。驯化水温15℃时, CTM呈先升高后降低的趋势, 即4℃/h(34.5℃)温升速率组的CTM显著高于0.5~2℃/h (31.9~32.8℃)的低温升速率组和12~15℃/h(32.8~ 33℃)的高温升速率组(P < 0.05)。驯化水温22℃时, CTM随温升速率增大而显著升高(P < 0.05), ≥9℃/h (34.4~34.7℃)的温升速率组的CTM显著高于≤1℃/h (33.0~33.3℃)的温升速率组(P < 0.05)。驯化水温为29℃时, CTM随温升速率增大而显著升高(P < 0.05), ≥12℃/h(38.9~39.0℃)的温升速率组的CTM显著高于≤1℃/h(37.7~38.1℃)的温升速率组(P < 0.05)。
日本
驯化水温和温升速率对口虾蛄的CTM均有显著影响; 在同一温升速率下, CTM随驯化温度的增大而显著上升(P < 0.05;表 2, 图 4)。驯化水温8℃时, CTM随温升速率增大而显著上升(P < 0.05), 在≥9℃/h (26~26.2℃)的高温升速率组显著低于≤2/h(27.4~ 27.8℃)的低温升速率组(P < 0.05)。驯化水温15℃时, CTM随温升速率增大而显著降低(P < 0.05), 在≤1℃/h (27.1~27.2℃)的低温升速率组显著低于≥12℃/h (29.1~29.4)的高温升速率组。驯化水温22℃时, CTM在0.5℃/h到9℃/h(31.5~32.5℃)的温升速率组中差异不显著(P < 0.05), 显著低于≥12℃/h(33.3~33.4℃)的温升速率组(P < 0.05)。驯化水温29℃时, CTM随温升速率增大而显著升高(P < 0.05), 在≥9℃/h(35.3~35.7℃)的高温升速率组显著高于≤2℃/h(33.0~33.5℃)的温升速率组。
2.2 实验动物的24 h高起始致死温度(24 h UILT50)驯化水温对3种实验动物的24 h UILT50影响显著(P < 0.05;图 5, 表 3)。

本实验中, 不同的甲壳类动物的热耐受性表现出明显的种间差异, 日本
甲壳类生物的热耐受性研究在海洋热污染风险评估中具有高代表性和应用性。但相对于鱼类等游泳生物, 对甲壳动物的相关研究较少。游泳动物具备较强的迁移活动能力和应急逃避能力, 在自然状态下温排水对其影响多限于其分布范围的改变, 非极端条件下致死现象鲜有发生[27]。而甲壳类动物迁移能力较差, 当栖息环境出现热胁迫, 易受到温排水的热冲击, 甚至出现致死现象。另外, 鱼类的热耐受性一般高于虾类, 但低于蟹类[19], 这表明在对温排水潜在热污染评估中, 除了鱼类外, 选择以蟹虾类为代表的甲壳类动物开展热耐受性研究能够更为全面地量化热污染程度。
本实验中的3种实验动物的热耐受指标(CTM和24h UILT50)总体上均随驯化水温升高而呈上升趋势。海洋动物多缺乏稳定的自身体温调节能力[22], 在其他环境因素稳定的前提下, 基础水温越高, 海洋动物的热耐受性越强。温度的微小变化都可能对海洋动物的各种生命活动造成显著影响, 特别是尚未具备自我调节能力的早期生活史阶段, 即幼体阶段[28]。因此在海洋动物热耐性研究中多采用幼体为研究对象以期更好地评估水温变化对动物本身及生态环境的影响。研究表明, 大部分甲壳类动物的热耐受性都与驯化水温呈正相关关系, 例如, 切脊熟若蟹(Lophozozymus incisus)、弓蟹(Hemigrapsus crenulatus)、短沟对虾(Penaeus semisulcatus)、褐虾(Crangon crangon)、大虎虾(Penaeus monodon)和欧洲龙虾(Astacus astacus)等的热耐受能力均随驯化水温升高而显著上升[29-32]。与此同时, 由于不同季节气候变化引起的动物机体代谢水平和生理活动状态不同, 同种海洋动物的热耐受性在不同驯化水温下也存在较大差异。例如, 云斑厚纹蟹(Pachygrapsus marmoratus)的热耐受指标都随驯化水温从20℃升高到26℃而显著增大; 褐美对虾(Farfantepenaeus aztecus)的CTM值在不同驯化水温下约比驯化水温高3~5℃不等[24, 33]。另外, 即使是同一物种, 在不同的季节也表现出不同的热耐受性, 以日本
在本实验中, 各物种的热耐受性在不同驯化水温下对温升速率的反应表现出显著差异性。其显著特征之一是种间特异性, 即不同海洋动物对温升速率的响应程度不同。例如, 在相同驯化水温29℃下, 叶状泥蟹(Eurypanopeus abbreviatus)的CTM随温升速率的升高而上升, 而古巴石蟹(Menippe hodifrons)的CTM则随温升速率的升高呈先上升后降低趋势[19]; 在28℃~29℃驯化水温下, 斑马鱼(Danio rerio)和孔雀鱼(Poecilia reticulata)的CTM随温升速率的升高而上升, 而云纹副鳚(Parablennius marmoreus)的CTM则随温升速率升高而降低[19, 35]。温升速率影响海洋动物的热耐受性的另一个特征是明显受驯化水温影响。许氏平鲉(Sebastes schlegeli)和大泷六线鱼(Hexagrammos otakii)的CTM在低驯化水温(5℃和13℃)下随温升速率升高呈下降趋势, 而在高驯化水温(17℃和26℃)下随温升速率增大而显著上升[36]。即使是同一温升速率下, 海洋动物在不同驯化水温下也通常会表现出显著差异。例如, 在1℃/h的温升速率下, 热带厚纹蟹(Pachygrapsus transversus)的CTM值随水温从26℃升高到32℃而增大约1℃[24]; 在2℃/h温升速率下, 四川裂腹鱼(Schizothorax kozlovi)的CTM值在驯化水温从10℃升高到26℃的背景下从32.86℃增大到34.54℃[28]。在低驯化水温条件下, 海洋动物难以迅速适应水温的快速升高, 生命活动被抑制, 易达到临界高温致死状态, 导致其CTM随温升速率的升高而降低。而在高驯化水温下, 海洋动物本身对高温产生了一定适应性, 在实验过程中又可通过机体自身的新陈代谢等生理活动进一步适应水温升高, 其CTM值随温升速率升高而增大。目前关于温升速率对海洋动物热耐受性影响的研究中所采用的温升速率尚未建立统一标准[37], 其中在80余种北美地区的鱼类的热耐受性研究中, 0.027~60℃/h均有采用[38]。海洋动物不同的遗传特性及表型可塑性使得它们对温升速率的响应不尽相同[39], 制定统一规范的温升速率标准对不同实验动物来说并不可行。因此, 在热耐受性研究过程中, 不仅要考虑自然状态下温排水所引起的温升速率差异, 也应充分考虑不同物种的遗传特性、地理区系差异以及实验可行性等因素, 在合理温升速率下开展其热耐受性研究会更好地评估温排水对海洋生物产生的不利作用。
本研究为评估和监测电厂温排水对三门湾水域资源环境的热效应提供科学参考。据对三门湾核电站温排水扩散特征的初步模型分析预测, 当核电站正常运行时, 温排水引起的4℃温升最大包络面积小于4 km2, 1℃温升最大包络面积小于23 km2, 对生物群落的影响范围及程度相对较小。考虑到核电站实际运行中可能出现的极端状况和温排水的热累积效应, 本研究发现日本
