海藻糖类的气相色谱分析 Ⅲ.红藻糖苷的提取与测定

摘要:
以气相色谱法分析红藻中游离低分子碳水化合物红藻糖苷。结果表明,红藻糖苷可以用80%的乙醇提取,然后用除色素除盐,经硅醚化后用气相色谱进行定性与定量分析;对于影响提取与硅醚化的各种因素进行了研讨,并与传统的苯酚硫酸法进行了比较。

GAS CHROMATOGRAPHIC ANALYSIS OF SUGARS IN SEAWEEDS Ⅲ. EXTRACTION AND DETERMINATION OF FLORIDOSIDE IN RED SEAWEEDS
Abstract:
Floridosides were extracted from the powder of smashed algae samples with 80% EtOH in 15 min(×3). The EtOH soloble fractions were combined, evaporated to dryness at 50℃. The lipids and pigments were removed with petrolium ether, then the insoluble fractions were dissolved in H2O and deionized by using ion exchange resins. The neutral fractions were filtered and evaporated to dryness and kept in vacuo over P2O5. Floridosides were treated with Trimethylsilylation reagent (Sily1-8), heated at 40℃ for 1 hour and allowed to stand for 4hours at room temperature peior to Gas Chromatography in 3m × 3m glass column (packed with 15% SE-30) with N2 as carrier gas (2.5kPa/mm2). Column temperature was 210℃, injector and FID detector temperatures were 270℃. Heneicosane was used as the inner standard. The effects of temperature, time and reagent amount on the extraction and trimethylsilylation of floridoside were investigated. The optimal conditions were selected.
