合浦珠母贝β-肌动蛋白基因的克隆和序列分析

摘要:
为进一步研究珍珠质基质蛋白外排机制,同时为今后合浦珠母贝(Pinctada fucata)不同组织的基因表达研究提供阳性对照,根据大鼠(Rattus norvegicus)胞质型肌动蛋白β-actin设计引物,通过RT-PCR方法克隆鉴定了合浦珠母贝肌动蛋白同源基因片段。序列分析表明,该片段编码的氨基酸序列与相应同源片段具有较高的保守性。合浦珠母贝β-肌动蛋白有多个特有的氨基酸残基。合浦珠母贝可能是在系统进化上较早分出的一支。

Cloning and analysis of the gene fragment of β-actin from Pinctada fucata
Abstract:
Nacre is the internal lustrous "mother of pearl" layer, and pearls are composed entirely of nacre. Nacre is formed by the outer epithelia of the mantle, incorporating secret matric proteins with calcium carbonate. So the endocytosis and exocytosis of the outer epithelia are very reactive. β-actin is an important component of cytoskeleton, which involved in a variety of cellular events including endocytosis and exocytosis. It is very important to elucidate the nacreous protein’s exocytosis mechanism. The identification of β-actin in Pinctada fucata may also supply the positive comparison for gene expression in different tissues. In this paper we identified a fragment of β-actin gene by PT-PCR from P. fucata. Analysis shows a very high degree of homology between β-actin of P. fucata and β-actin from other animals. However the β-actin from P. fucata has several special characters in its amino acids and system evolution.
