长江中华鲟遗传多样性变化

摘要:
采用随机扩增多态性DNA(RAPD)技术对2000年共 30尾长江中华鲟(Acipenser sinensis)幼鲟遗传多样性进行了分析。共用40个10bp长的随机引物,在25个可分析的引物中,只有 OPK01,OPK02,OPK03,OPK09和OPK14RAPD-PCR产物有多态现象,多态引物占20%。25个引物共扩增出101条稳定的 DNA带。其中12条为多态带,多态座位比例为9.9%。香农遗传多样性指数(Hsub>0)为2.6332,遗传多样性指数(H)为0.0261,遗传相似性指 数平均为0.9824,遗传距离平均为0.0176。其遗传多样性明显低于葛洲坝截流以前。

The variation of genetic diversity of Chinese sturgeon (Acipenser sinensis) in the Yangtz River
Abstract:
Totally 30 samples collected in 2000 were analyzed by random amplification of polymorphic DNA(RAPD). 40 decamer primers were applied for analysis and only 25 primers gave stable results. Among 25 primers studied only 5 primers(20%) such as OPK01, OPK02, OPK03, OPK09 and OPK14 produced polymorphic bands. Total 101 bands(locus) were obtained using 25 primers. Out of the 101 bands(locus), 10 bands were polymorphic. The percentage of polymorphic loci was about 9.9%. The Shannon’s index of phenotypic diversity Hsub>0=2.633 2. The genetic diversity H=0.026 1. Genetic similarity and genetic distance between individuals were 0.982 4, 0.017 6 respectively. The genetic diversity of the samples in this study was lower than that before the construction of Gezhouba dam.
