彩虹明樱蛤基因组DNA提取及RAPD扩增的初步研究

摘要:
对采自浙江温岭的彩虹明樱蛤(Moerella iridescens)养殖群体的DNA提取和RAPD扩增条件进行了研究。从38种随机引物中筛选到重复性好的6种引物分别对11个个体的DNA进行扩增,共产生247个DNA条带(300~1 600 bp),总共检测到43个位点,其中多态位点32个,多态位点比例为74.42%。11个个体间的遗传相似系数在0.553~0.884之间,平均为0.691,个体间的平均遗传距离为0.309。表明该群体的遗传多样性比较丰富,种质资源处于良好状态。

Genomic DNA isolation and RAPD analysis on Moerella iridescens
Abstract:
Studies of DNA extraction and RAPD(Random Amplified Polymorphic DNA) analysis were performed on the cultured population of Moerella iridescens in Wenling, Zhejiang Province. Among 11 individuals, six screened from thirty eight primers, which were reproducible, amplified a total of 247 fragments ranging from 300 bp to 1 600 bp. 43 loci were detected and the polymorphic loci(P) were 74.42%.Genetic similarity index(S) among individuals were 0.691 and the average genetic distance(D) were 0.309.The result indicates that the degree of genetic polymorphism in the analyzed population is high.
