基于响应面法的鮟鱇活性肽制备工艺

摘要:
鮟鱇是一种低脂肪高蛋白的优质海洋生物资源, 其氨基酸含量丰富, 蛋白质含量高达75.30%。本实验选用胰蛋白酶, 胃蛋白酶, 木瓜蛋白酶和菠萝蛋白酶水解鮟鱇鱼糜, 比较各酶解产物对1, 1-二苯基-2-三硝基苯肼(DPPH)自由基的清除率, 确定木瓜蛋白酶为最佳用酶。在单因素实验基础上进行响应面优化实验, 确定酶解反应的最佳条件为: 加酶量829.51 U/g, pH值7.37, 酶解温度47.09 ℃, 酶解时间5.86 h, 液固比5.79∶1。在最优条件下验证其DPPH自由基清除率为80.22%±0.63%, 接近预测值80.55%。经响应面法实验制备的鮟鱇活性肽粉末呈淡黄色, 具有较好的抗氧化活性, 其羟自由基清除能力的IC50值为1.69 mg/mL, 可作为潜在的抗氧化剂。

Optimization of monkfish active peptide using response surface methodology

Abstract:
Minced monkfish fillet is an excellent marine biological resource with a diverse range of amino acids, a high protein content of up to 75.30%, and low fat content. In this experiment, four kinds of proteases, i.e., trypsin, pepsin, papain, and bromelain, were used for hydrolysis to produce the monkfish protein hydrolysate. Papain was chosen as the best enzyme of the four after considering the 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of various enzymatic hydrolysates. Based on a single factor experiment, the study used response surface methodology to obtain the optimal conditions. The parameters of the experiment were as follows: enzyme concentration of 829.51 U/g, pH of 7.37, hydrolysis temperature of 47.09 ℃, hydrolysis time of 5.586 h, and liquid- to-solid ratio of 5.79:1. The investigation found that the maximum DPPH scavenging activity was 80.22%±0.63%, which was comparable to the experimental predicted value of 80.55%. The monkfish peptide prepared using response surface methodology was a light-yellow powder with high antioxidant activity. Its IC50 of hydroxyl radical scavenging ability was 1.69 mg/mL, showing its potential as an antioxidant.
