ISSR 标记在牙鲆(Paralichthys olivaceus)与石鲽(Kareius bicoloratus)种间杂交一代的分离方式分析

摘要:
利用ISSR 标记对牙鲆♀与石鲽♂单对杂交亲本及其子一代(全同胞家系)的分离方式进行了研究。结果表明, 8 个ISSR 引物共扩增出181 个标记, 其中116 个为多态性, 占总数的64.09%; 标记在F1 代呈三种分离方式:符合孟德尔分离比例、偏离孟德尔分离比例和异常分离。符合孟德尔分离比例的情况包括:不分离(代表了亲本基因型的5 种组合:AA×AA、AA×Aa、Aa×AA、AA×aa、aa×AA); 1:1 分离(代表了亲本基因型的2 种组合:Aa×aa、aa×Aa); 3:

THE SEGREGATION OF ISSR MARKERS IN THE F1 PROGENY OF INTERSPECIES CROSSBREED BETWEEN PARALICHTHYS OLIVACEUS AND KAREIUS BICOLORATUS

Abstract:
The segregation pattern of ISSR markers in the F1 of interspecies cross between Paralichthys olivaceus and Kareius bicoloratus was studied. Eight primers yielded 181 markers, of which 116 (64.09%) were polymorphic. The segregation showed three patterns: Mendelian, deviated Mendelian, and abnormal. The Mendelian segregation included three types: (1) non-segregation that reflects five genotypic combinations of the parents: AA×AA, AA×Aa, Aa×AA, AA×aa, and aa×AA; (2) 1︰1 segregation, reflecting two genotypic combination of the parents, Aa×aa and aa×Aa; (3) 3︰1 segregation, reflecting one genotypic combination of the parents, Aa×Aa. The deviated pattern includes two types: polymorphic in the parents and deviated from 1︰1 ratio in the progenies and the appeared in the parents and deviated from 3︰1 ratio in progenies. The abnormal pattern includes two types: absent in the parents but present in the progenies, and present in the parents but absent in the progenies. The percentages and amounts of the three patterns were 82.87%, 150; 11.05%, 20; and 6.08%, 11; respectively. In addition, 85.33% of the markers presenting in both the parents did not segregate and 28.57%—35.85% of the markers presenting in only one parent showed normal segregation. The above results could provide certain theoretical basis for further work to the construction of linkage map of P. olivaceus and K. bicoloratus.
