蜈蚣藻和管形藻的RAPD分析

摘要:
于1997年9月在大连石庙海区分株采集蜈蚣藻、管形藻和舌状蜈蚣藻,采用RAPD技术在分子水平上对属于红藻门的两种藻类—蜈蚣藻和管形藻进行了分析。所试60个引物中,有21个经扩增得到了120个多态片段。应用PHYLIP软件,按照UPGMA和Neighbor-Joining(N-J)法分别构建聚类图。分析结果表明,RAPD多态性提示蜈蚣藻和管形藻差异较大,应为两个不同的属。

RAPD ANALYSIS OF GRATELOUPIA FILICINA AND SINOYUBIMORPHA PORRACEA

Abstract:
Randomly amplified polymorphic DNA (RAPD) was used to study Grateloupia filicina and Sinoyubimorpha porracea (Rhodophyta), in terms of the molecular systematics. Amplification with 60 random primers was made under predetermined optimal reaction (samples were first heated at 95°C for 10 min., and followed by 45 cycles of 60 sec at 94°C, 100 sec at 36°C, and 160 sec at 72°C, then held at 72°C for 10 min after 45 cycles). 21of the 60 random primers tested were positive, generating a total of 120 prominent amplification products. The amplified fragments were scored as present (1) or absent (0) for each DNA sample and an index of degree of band sharing (S) was calculated by using Nei and Li’s matching coefficient method. The value of (1–S) was used to evaluate genetic distances between species. The dendrograms were built based on UPG-MA and Neighbor-Joining analysis of genetic distance matrix data. The result suggests that RAPD is useful in distinguishing G. filicina from S. porracea and that G. filicina and S. porracea may belong to different genera.
