紫球藻两种藻红蛋白特性的比较研究——γ亚基在稳定藻红蛋白结构方面的作用

摘要:
从紫球藻中分离纯化两种藻红蛋白(B-藻红蛋白和b-藻红蛋白),对它们的吸收光谱和电泳图谱进行比较研究。结果表明,尽管电泳图谱与文献报道的相同,但b-藻红蛋白的吸收光谱与文献报道的有较大的差别,表现在本研究发现b-藻红蛋白在545nm处有一吸收峰,在565nm处仅有一个吸收肩峰,而不是文献中报道的在545nm和565nm均有独立的吸收峰。通过比较B-藻红蛋白和b-藻红蛋白在不完全变性条件下的电泳图谱,发现B-藻红蛋白比b-藻红蛋白要稳定的多,进一步分析认为这主要是因为B-藻红蛋白含有γ亚基,而b-藻红蛋白不含有γ亚基,因此认为γ亚基有稳定分子的作用。

COMPARATIVE STUDY ON THE PROPERTIES OF TWO DIFFERENT KINDS OF PHYCOERYTHRIN FROM PORPHYRIDIUM CRUENTUM

Abstract:
Two kinds of phycoerythrin, B-phycoerythrin and b-phycoerythrin, were isolated and purified from Porphyridium cruentum, and the characterization of the two kinds of the phycoerythrin was studied using both spectral analysis and electrophoresis. In comparison with the result reported by Glazer et al, the electrophoresis is very similar, while spectral properties of b-phycoerythrin had some difference, which means that besides a peak at 545nm just a should not an absolute peak at 565nm was showed in the absorption spectrum of b-PE. Through comparing the results of electrophoresis at the none-completely denatured condition between B-PE and b-PE, it was found that B-PE is much more stable than b-PE. Further studying indicates that γ subunit has the function to stabilize the structure of phycoerythrin.
