从患腹水症养殖牙鲆鱼苗分离到一株细菌B17,人工感染实验证实其具有致病性。用Biolog-GN细菌鉴定系统对B17进行测试,发现其与参考菌株的相似性低,不能进行鉴定。常规生理生化测试表明,B17具有弧菌属的特征,其表型特征与Vibrio splendidus非常相似。为了进一步明确菌株B17的分类学地位,测定了其16S rRNA基因序列,分析了相关细菌相应序列的同源性,构建了系统发生树,结果表明菌株B17与V. splendidus的亲缘关系最近。综合上述结果,菌株B17可鉴定为V. splendidus。

Bacterial strain B17, which was isolated from the Japanese flounder (Paralichthys olivaceus) larvae with swollen abdomen, was proved to be a pathogenic organism by oral infection experiments. Biolog-GN (gram negative) identification system were applied in the determination of strain B17, showing 65.7% similarity with reference strain of Vibiro vulnificus. The similarity was low and could not give identification of strain B17. The traditional physiological and biochemical methods showed that most of the phenotypes of strain B17 were similar to Vibiro splendidus, according to the description of Paul et al(1984). In order to investigate the phylogenetic position of this pathogen, a 1477bp of 16S rRNA gene was sequenced and compared with that of related strains. Molecular phylogenetic dendrogram was constructed based on the genetic distance analysis, which revealed that strain B17 exhibited the highest levels of 99.3% similarity to Vibiro splendidus. Based on the results obtained from phenotypes and 16S rRNA gene analysis, strain B17 was identified as Vibiro splendidus.
