山东日照和福建厦门沿海花鲈(Lateolabrax japonicus)遗传多样性的RAPD研究

摘要:
采用RAPD技术, 分析了日照、厦门沿海花鲈两群体的遗传多样性水平。结果表明, 31条随机引物共扩增出两地花鲈455条RAPD标记带, 其中日照群体占263条, 厦门群体为192条;455条中有184条带为两群体共有。日照群体内遗传相似度I为0.8928(差异度为0.1072), 厦门群体I为0.9225(差异度为0·0775), 而两者群体间的遗传相似度I为0.8338(遗传距离D=0.1662)。两地花鲈在基因组DNA水平上存在较大的遗传差异。利用随机引物S11、S42、S45、S52和S203, 在花鲈两群体间扩增出8条稳定、明显的群体间特异性标记带。

COMPARATIVE ANALYSIS OF RAPD GENETIC VARIATION BETWEEN TWO STOCKS OF SEABASS (LATEOLABRAX JAPONICUS) FROM SHANDONG RIZHAO AND FUJIAN XIAMEN SEA AREAS

Abstract:
RAPD method was used in analyzing the genetic diversity of two stocks of L. japonicus, which were collected respectively from the sea area of Rizhao Shandong and Xiamen Fujian Provinces, inJ une 2001 and February 2002. The muscle of the 80 L. japonicus was used to extract the genetic DNA. The best reaction system concluded 100ng genetic DNA, 200μmol/L dNTP, 2mmol/LMg2+, 1.5μmol/L primers and 1.0UTaq DNA polymerase. Samples were subjected to PCR in a PE9600 Gene Cycler using 40 cycles of 1 min at 94℃, 1 min at 38℃, 2 min at 72℃. The inter-stock genetic similarity, intra-stock genetic similarity were analyzing. Sixty random primers were used for the analysis of nuclear DNA polymorphism of two stocks. The result showed that thirty-one primers were effective. 455 bands were obtained by the experiment. Among them, 263 bands were acquired from Rizhao stock, while 192 bands from Xiamen stock. Two stocks shared 184 bands. Using the primers of S11, S42, S45, S52 and S203, the eight distinctive bands were characterized by Rizhao stock, which could be used as fingerprinting marker to identify Rizhao and Xiamen stock. According to the pattern, the inter-stock genetic similarity of Rizhao stock is 0.8928, while Xiamen stock 0.9225. The intra-stock genetic similarity between two stocks is 0.8338. The percentage of polymorphic loci (P) of Rizhao stock was 31.18%, obviously higher than the average percentage of the vertebrate (24.7%). The percentage of polymorphic loci of Fujian stock is 24.08%, 7.1% lower than Rizhao stock and vertebrate. These results suggest that there is a significant genetic variation between two stocks and there is high genetic diversity of L. japonicus. However, the genetic diversity of the stock of Xiamen is not good. So the measures must be used to protect L. japonicus resource.
