以从海洋微藻中分离到的206株共栖细菌为材料, 采用生物传感器法筛选具有群体感应系统的细菌, 根据16S rRNA基因对它们进行了分子生物学鉴定和系统发育学分析。利用气相质谱联用法(GC-MS)检测群体感应信号分子酰基高丝氨酸内酯类化合物(AHLs)的种类; 采用细胞活体荧光测定方法, 研究了这类共栖细菌对微藻生长的抑制作用。结果表明, 在分离到的206株微藻共栖细菌中有11株具有群体感应系统, 它们分属于3类海洋细菌的系统分支: α-Proteobacteria、γ-Proteobacteria和Gram+细菌, 通过GC-MS检测确定11株共栖细菌都含有信号分子C8-HSL, 部分含有C6-HSL和C14-HSL, 其中9株细菌具有抑藻活性, 其代谢产物分别对东海原甲藻、棕囊藻、海链藻和赤潮异湾藻的生长具有不同程度的抑制作用。 |

SCREENING OF MICROALGAE ASSOCIATED BACTERIA WITH QUORUM SENSING SYSTEM AND THEIR ALGICIDAL ACTIVITY |

Two hundred and six associated bacteria strains were isolated from marine microalgae. The bacteria with quorum sensing system were screened by biosensor method. And the identification and phylogenetic analysis of these bacteria were carried out by molecular method based on their 16S rRNA gene. Acylhomoserine lactones (AHLs), as the quorum sensing signals, were detected in the extracts of bacteria strains with quorum sensing system by gas chromatography mass spectrometry (GC-MS), as well as the algicidal activities were analyzed by viable cell fluorescence detection method. Results showed that 11 among the 206 strains had quorum sensing system and they belonged to three phylogenetic branches: α-Proteobacteria, γ-Proteobacteria and Gram+ bacteria. GC-MS analysis showed that all of the 11 screened strains contained the quorum sensing signal of C8-HSL, and some of them also contained C6-HSL and C14-HSL. The algicidal activity study showed that 9 strains had inhibition effects in different degrees against four red tide microalgae including Prorocentrum donghaiense,Phaeocystis globosa,Thalassiosira sp. and Heterosigma akashiwo. |
