大黄鱼(Pseudosciaena crocea)源美人鱼发光杆菌(Photobacterium damselae)的分离鉴定及致病性研究

摘要:
于2010年从宁德患病大黄鱼中分离得到一株优势菌NPL1006, 经鉴定该菌株为美人鱼发光杆菌。人工感染实验结果显示该菌对大黄鱼的半致死浓度(LD50)为7.96×103CFU/g体重。药敏性实验结果显示NPL1006对头孢唑啉、头孢噻吩和诺氟沙星等9种药物高度敏感。采用纸片法检测得到NPL1006的胞外产物具有蛋白酶、明胶酶和淀粉酶活性。胞外产物感染斑马鱼研究结果显示其LD50为1.20mg蛋白/kg体重。以上结果表明大黄鱼源美人鱼发光杆菌NPL1006胞外产物含有多种毒力因子, 与该菌对大黄鱼的致病性密切相关。

STUDIES ON THE ISOLATION, IDENTIFICATION AND VIRULENCE OF PHOTOBACTERIUM DAMSELAE ISOLATED FROM PSEUDOSCIAENA CROCEA

Abstract:
A dominant strain (NPL1006) was isolated from diseased Pseudosciaena crocea in 2010. The results of artificial infection experiment showed that the LD50 of NPL1006 to P. crocea was 7.96×103 CFU/g body weight. NPL1006 was identified as Photobacterium damselae by Time of Flight Mass Spectrometer and the 16S rDNA sequencing analysis. Susceptibility experiments showed that NPL1006 is highly sensitiveto 9 drugs such as Cefazolin, Cefalotin and Norfloxacin. The extracellular products of NPL1006 were found to exhibit protease, gelatinase and amylase activities. The LD50 of extracellular products to zebrafish was 1.20mg protein/kg body weight. These results suggested thatextracellular products of P. damselae NPL1006 separated from P. crocea contained a variety of virulent factors, and it’s closely related to the pathogenicity of the bacteria P. crocea.
