连续降温对大菱鲆(Scophthalmus maximus)成鱼血清生化指标及Wap65-1基因表达的影响

摘要:
将大菱鲆(Scophthalmus maximus)从正常养殖水温18℃ 快速连续降温至1℃, 并在18、13、8、5、3 和1℃ 共计6 个温度点取血采样, 分别测定血清中的谷丙转氨酶(ALT)、肌酐(CR)、乳酸脱氢酶(LDH)、血糖(GLU)、皮质醇以及不同组织中Wap65-1 基因的表达量。结果发现, 随着温度降低,大菱鲆的血清ALT 活性呈升高的趋势, 且在8—1℃ 区间内显著高于18—13℃ 区间(P<0.05); 血清CR 浓度呈升高的趋势, 在3℃ 时最高, 且显著高于除1℃ 之外的其它实验(P<0.05); 血清中LDH 活性呈先升高后降低的趋势, 在13—5℃ 区间内显著高于其它实验组(P<0.05); 血清GLU 浓度呈先升高后降低的趋势, 在8—3℃ 区间内显著高于其它实验组(P<0.05); 血清皮质醇浓度呈升高的趋势,在1℃ 时最高, 且显著高于其它实验组(P<0.05)。连续降温过程中, 大菱鲆所有组织Wap65-1 基因在特定温度下表达量都有上调(脑、胃、肝脏、头肾、肾脏、脾脏、肌肉、肠道、心脏)。其中, 脑、胃、头肾、肾脏和心脏组织中的Wap65-1 基因在正常温度下基本不表达, 只在降温过程中表达; 肝脏、脾脏、肠道和肌肉这4 种组织中的Wap65-1 基因在正常温度下有所表达, 且在降温过程中表达量显著升高(P<0.05)。

EFFECT OF CONTINUOUS COOLING ON SERUM BIOCHEMICAL PARAMETERS AND Wap65-1 EXPRESSION OF ADULT TURBOT SCOPHTHALMUS MAXIMUS

Abstract:
Adult turbots were subject to continuous cooling from 18 to 1℃, and five individuals were collected at each specific temperature including 18, 13, 8, 5, 3, and 1℃. Contents of five serum biochemical indices were measured consisting of ALT, CR, LDH, GLU, cortisol and the expression of Wap65-1 gene in different tissues was tested. The results indicate that as temperature decreased, the serum ALT activity increased and groups from 8℃ to 1℃ were significantly higher than 18℃ to 13℃ (P<0.05). The concentration of serum CR increased as temperature decreased, and was the highest at 3℃, which was significantly higher than those of other groups except for 1℃ group (P<0.05). Serum LDH activity increased and then decreased, and in the range from 13℃ to 5℃ it was significantly higher than those of other groups. Serum GLU concentration increased and then decreased, and in the range from 8℃ to 3℃ it was significantly higher than the other groups. Serum cortisol concentration increased as the temperature decreased, and was the highest at 1℃ which was significantly higher than those of other groups (P<0.05). The expression of Wap65-1 showed that the continuous cooling had an effect on all tissues (brain, stomach, liver, head kidney, kidney, spleen, muscle, intestine, and heart at specific temperature). The expression of Wap65-1 in them increased significantly during cooling. Wap65-1 of the brain tissue, stomach tissue, head kidney, kidney and heart tissue at normal temperatures had almost no expression, but expressed significantly during the cooling process. Wap65-1 of liver, spleen, intestine, and muscle tissues expressed at normal temperature and expression levels elevated significantly in the cooling process.
