COMPARATIVE STUDY ON THE SEPARATION OF NUCLEIC ACIDS FROM THE NUCLEI OF RAPANA THOMASIANA LIVER |

Abstract: |
Two different procedures for the separation of nucleic acids from cell nuclei of Rapana thomasiana liver have been studied. Differences were found between the results chromatograp-hed through Sepharose 2B and Hydroxylapate. The nucleic acids extracted with Sodium dodecyl sulfate can only be separated into two peaks of DNA and RNAs by chromatography on Sepharose 2B. In ease of using Hydroxyapatite columns, a good separation of polynucleotides, tRNA, rRNA and DNA can be obtained, the recovery rate of DNA and RNAs obtained on Hydroxyapatite column is higher than that on Sepharose 2B. Absorption spectrum analysis study showed that the quality of the products eluted from Hydroxyapatite column is better than that from Sepharose 2B. So Hydroxyapatite column chromatography is a useful method for the separation of DNA and various classes of RNA. |
