通过抑菌及抗肿瘤活性实验, 明确海洋真菌Neosartorya fischeri 1008F1次级代谢产物中的活性有效组分。应用对峙培养法和生长速率法, 对菌株的抑菌活性进行测定。应用MTT 法, 对菌株的抗肿瘤活性进行测定。应用凝胶(Sephadex LH-20)柱层析的方法, 对菌株发酵产物的提取物进行分离。结果发现, 海洋真菌Neosartorya fischeri 1008F1的水溶性提取物对番茄早疫菌(Alternaria solani)具有一定的抑制活性。菌株的粗提物浸膏经Sephadex LH-20 柱梯度洗脱后, 得到的组分B、C、D、E 和F。在0.5 g/L 的供试浓度下, 组分B、C 和D 对胃癌细胞SGC-7901 的抑制率均高于80%, 而组分B、C、D、E 和F 对肝癌细胞BEL-7404 的抑制率均高于70%。 |

Antimicrobial and antitumor activities of secondary metabolites from a marine fungus Neosartorya fischeri 1008F1 |

Abstract: |
In this study, we determined antimicrobial and antitumor activity of secondary metabolites obtained from a marine fungus Neosartorya fischeri 1008F1. The antimicrobial activity was evaluated by confront culture and growth rate methods. The antitumor activity was evaluated by MTT assays. The extract of fermentation product was isolated through gel (Sephadex LH-20) column chromatography. The results show that the water soluble extract of marine fungus N. fischeri 1008F1 had inhibitory activity against Alternaria solani. The extract of the strain was isolated through gel (Sephadex LH-20) column chromatography and fractions B, C, D, E and F were obtained. At 0.5 mg/mL, the inhibition rates of fractions B, C and D against SGC-7901 cell line were higher than 80%. At 0.5 mg/mL, the inhibition rates of fractions B, C, D, E and F against BEL-7404 cell line were higher than 70%. |
