松江鲈幼鱼性腺分化的组织学观察 |

摘要: |
为探究松江鲈(Trachidermus fasciatus Heckel)性腺分化和发育的规律,以孵化后10~120日龄的松江鲈幼鱼为研究对象,对其性腺进行常规石蜡切片和H-E组织染色法观察。结果表明:20日龄幼鱼原始性腺形成;40日龄幼鱼出现卵巢腔的雏形,标志着卵巢解剖学分化的开始;45日龄幼鱼出现初级卵母细胞,表明卵巢开始进行细胞学分化;60日龄幼鱼卵巢开始进入Ⅱ期,卵母细胞基本进入第II时相;65日龄幼鱼精巢处于Ⅰ期,输精小管的出现表明精巢开始进行解剖学分化;70日龄幼鱼形成囊状的精原细胞团,表明精巢开始细胞学上的分化;90日龄幼鱼精巢进入Ⅱ期,初级精母细胞形成,细胞排列成束状。松江鲈鱼早期性腺分化属于分化型雌雄异体型,卵母细胞为部分同步型发育,精巢的发育晚于卵巢,且其性腺分化方向受生殖细胞数量的影响。松江鲈鱼的卵母细胞对脂质的积累能力相对较强。研究结果明确了松江鲈早期性腺分化过程及性腺分化关键时间节点,为其性别决定和分化机制研究提供了重要基础资料,也将为松江鲈人工繁育和人工性别控制诱导敏感期等提供理论参考。 |

Histological observation on gonadal sex differentiation in Trachidermus fasciatus |

To elucidate the gonadal differentiation and development process of Trachidermus fasciatus, larvae aged 10 to 120 days post-hatching (dph) were examined using conventional paraffin sections and H-E tissue staining. The results revealed several key milestones: primordial gonadal formation at 20 dph, appearance of the embryonic ovarian cavity at 40 dph, initiation of ovarian cytological differentiation marked by primary oocytes at 45 dph, entry into stage II at 60 dph, and the appearance of fissure seminiferous tubules at 65 dph indicating the onset of anatomical differentiation of the testis (stage I). Furthermore, at 70 dph, the formation of a vesicle of spermatogonium signaled the cytological differentiation of spermatogonia. By 90 dph, the presence of stage II spermatocytes, forming primary spermatocytes arranged in bundles, marked a significant stage in testicular development. T. fasciatus exhibits heteromorphic differentiation into males and females. The oocyte development is partially synchronous. The differentiation time of the testis is later than that of the ovaries, and the differentiation direction of the gonads is affected by the number of germ cells. In terms of lipid accumulation ability, T. fasciatus oocytes exhibit relatively strong. T. fasciatus displays distinct reproductive characteristics, providing valuable insights into its gonadal development and differentiation. The results of this study clarified the gonadal differentiation process and key time nodes of gonadal differentiation, provided important foundational data for the study of its sex determination mechanism, and also provided theoretical reference for artificial breeding and artificial sex control induction sensitive periods of T. fasciatus. |
