基于麦麸培养的藻栖深绿木霉RR-dl-3-9特征性代谢产物研究

摘要:
本文以藻栖深绿木霉Trichoderma atroviride RR-dl-3-9为对象, 以麦麸为培养基, 对其次生代谢产物进行了研究。运用正向硅胶柱层析、反相硅胶柱层析、凝胶柱层析、制备薄层层析以及高效液相色谱等方法, 从其在麦麸的发酵产物中分离得到了10个化合物, 分别为dehydrovomifoliol (1)、blumenol A (2) cucumegastigmane I (3)、9, 10-dihydroxy-4-megastigmen-3-one (4)、stigmastan-3-one (5)、stigmast-4-en-3-one (6)、6β-hydroxystigmast-4-en-3-one (7)、stigmastan-3β, 6α-diol (8)、stigmastan-3β, 5α, 6β-triol (9)和β-sitosterol (10)。生物活性测试结果表明化合物1和2在浓度为200 µg/mL时对禾谷镰刀菌4A1F和ACCC 39334的生长具有抑制能力; 而化合物4在同浓度下仅对菌株4A1F表现出抑制能力。

Characteristic metabolites of algicolous Trichoderma atroviride RR-dl-3-9 cultured on wheat bran

Abstract:
This study investigated the secondary metabolites of the algicolous fungus Trichoderma atroviride RR-dl-3-9 cultured on wheat bran. Ten compounds were isolated from the wheat bran fermentation using normal-phase silica gel column chromatography, RP-18 reversed-phase silica gel column chromatography, Sephadex LH-20 gel column chromatography, preparative thin-layer chromatography, and semi-preparative high-performance liquid chromatography. They were identified as dehydrovomifoliol (1), blumenol A (2), cucumegastigmane I (3), 9, 10-dihydroxy-4-megastigmen-3-one (4), stigmastan-3-one (5), stigmast-4-en-3-one (6), 6β-hydroxystigmast-4-en-3- one (7), stigmastan-3β, 6α-diol (8), stigmastan-3β, 5α, 6β-triol (9), and β-sitosterol (10). Bioassay results indicated that Compounds 1 and 2 exhibited inhibitory effects on the growth of Fusarium graminearum 4A1F and strain ACCC 39334 at a concentration of 200 µg/mL, while Compound 4 showed inhibitory activity only against strain 4A1F at the same concentration.
