织锦巴非蛤总类胡萝卜素两种提取方法比较分析

摘要:
以织锦巴非蛤(Paphia textzle)为研究对象,分别采用湿重法与干重法(冷冻干燥法)提取织锦巴非蛤斧足、外套膜、闭壳肌及鳃4种组织的总类胡萝卜素,并比较两种方法提取效果。结果表明:两种提取方法的变异系数均较高,干重法的得率要显著高于湿重法(P<0.05)。湿重法提取总类胡萝卜素含量(TCC)从高到低分别是:斧足、鳃、外套膜、闭壳肌,干重法提取的结果从高到低分别是鳃、斧足、外套膜、闭壳肌。由于变异系数均较大,导致两种方法测定总类胡萝卜素排序结果存在差异。建议以后在对贝类中的总类胡萝卜素提取时,如果实验要求高或组织富含类胡萝卜素,宜选用冷冻干燥法;若实验要求低或者组织所含类胡萝卜素含量少时,建议选择湿重法。

Comparative analysis of two extracting methods for total carotenoids from Paphia textzle

Abstract:
The total carotenoids from four tissues of Paphia textzle including axe foot, mantle, adductor muscle and gill were extracted by wet weight method and dry weight method (freeze-drying method) respectively, and the extraction effects of the two methods were compared. The results showed that the coefficient of variation of the two extraction methods were both higher, and the yield of freeze-drying method was significantly higher than that of wet weight method (P < 0.05). The total carotenoid content (TCC) extracted by wet weight method from high to low were axe foot, gill, mantle, and adductor muscle while the results extracted by freeze-drying method from high to low were gill, axe foot, mantle, and adductor muscle. The different detect results of total carotenoid content by using two extracted methods due to the larger coefficient of variation. Therefore, it is suggested the freeze-drying method should be adopted if the experimental requirements are high or the tissues are rich in carotenoids in the future. Wet weight method is recommended if the experimental requirement is not strict or the carotenoid content in tissues is not so much.
