海洋源生防细菌 LHB02 的筛选、鉴定及其抑菌谱检测

摘要:
从高度污染的海滩污泥中, 分离得到一株对部分水产病原菌有很强拮抗作用的生防细菌LHB02。 采用细菌的通用引物进行 LHB02 的 16S rRNA 基因片段扩增, 得到 l511bp 的 DNA 片段, 其序列与枯草芽孢杆菌(Bacillus subtilis)在进化位置上最为接近, 结合传统的形态学、生理生化鉴定手段, 鉴定该菌株为枯草芽孢杆菌(Bacillus subtilis)。LHB02 的无菌发酵液经硫酸铵沉淀, 沉淀物透析除盐后所得的活性物质初提物初步鉴定为一种抗菌蛋白质。与水产主要病原菌进行拮抗试验, 结果表明, LHB02 对三种弧菌包括哈维氏弧菌(Vibrio harveyi)、溶藻弧菌(V. alginolyticus)和鳗弧菌(V. anguillarum)有很强的抑制作用。

Abstract:
The bacterium strain LHB02, which exhibited high inhibitory effects against a number of aquatic pathogens, was separated from marine sediments heavily polluted by aquaculture. Using a pair of universal primers, a fragment of LHB02’s 16S rRNA gene was amplified and a 1511bp of its DNA sequence was obtained. Based on its morphological, physiological and biochemical characteristics, LHB02 was identified as Bacillus subtilis. The active substance purified using ammonium sulfate precipitation and subsequent dialysis was identified as a crude antibacterial protein. In the following antimicrobial test, LHB02 showed strong antagonistic effects against three types of vibrios: Vibrio harveyi, V. alginolyticus and V. anguillarum.
