东南沿海小孔蛸(Cistopus sp.)线粒体序列的比较研究

摘要:
采用线粒体DNA 16S rRNA 和COⅠ基因序列对福建、浙江沿海分布的5 个小孔蛸(Cistopus sp.)群体进行了遗传多样性研究。结果表明, 序列总长度分别为497—501bp (16S rRNA)和652bp (COⅠ); 小孔蛸5 个群体的序列碱基组成均显示出较高的A+T 含量(16S rRNA 基因72.1%, COⅠ基因65.9%)。16S rRNA 基因片段在种内和群体间个体差异均较小, 5 个群体共检测到8 个变异位点(其中7 个插入/缺失位点); COⅠ基因序列不存在

GENETIC DIVERSITY IN POPULATIONS OF CISTOPUS SP. BASED ON THE SEQUENCES OF MITOCHONDRIAL 16S rRNA AND COⅠ GENE FRAGMENTS

Abstract:
Sequences of 16S rRNA and COⅠ gene fragments in mtDNA of Cistopus sp. were studied. The samples were collected from five populations in the East China Sea. The sequences were compared by PCR. Nucleotide sequences of 497—501bp for 16S rRNA gene and 652bp for COⅠ gene were obtained. A high percentage of A+T base composition in the two sequences was observed. The alignment indicated that the variation level of intra-stocks was very low in both genes. Eight variable sites were detected for 16S rRNA gene fragment (of which seven were indels) across the five stocks. 21 polymorphic sites were detected in COⅠ gene fragments among the five stocks, with no indels. The NJ and UPGMA phylogenetic trees showed that no remarkable genetic difference was observed among those populations.
