厚壳贻贝(Mytilus coruscus)足丝蛋白 mcofp3 和 mcofp6 的 cDNA 克隆及序列分析

摘要:
通过设计特异性引物, 采用 PCR 扩增的方法对厚壳贻贝足腺细胞 cDNA 文库进行筛选, 共克隆得到 14条厚壳贻贝 mcofp3的 cDNA序列和 8条 mcofp6的 cDNA序列, 并对其基因序列及推导的氨基酸序列进行了序列比对和变异位点初步分析。结果表明, 14 条 mcofp3 cDNA 序列分属于厚壳贻贝mcofp3家族的两个亚家族, 其成熟肽序列变异相对活跃; 而8条mcofp6 cDNA序列并无明显的亚家族划分, 序列相对保守。本次实验所获得的 mcofp cDNA 序列初步显示了厚壳贻贝足丝蛋白家族基因的分子多样性, 为将来深入了解厚壳贻贝足丝蛋白的分子多样性机制以及在此基础上开发相关的新型生物黏附剂奠定了基础。

cDNA CLONING AND SEQUENCE ANALYSIS OF mcofp3 AND mcofp6 FROM MYTILUS CORUSCUS

Abstract:
Hard-shell mussels Mytilus coruscus attach securely to solid surfaces in the presence of moisture through a holdfast known as the byssus formed by various kinds of foot proteins secreted from the foot glander. Mfp3, the most abundant mfp family to date, locates mainly in the plaques, together with a newly found mfp family member, mfp6, and they both play key roles in underwater adhesion between the byssus and surrounding materials. Even though being one of the chief shellfish species cultivated in China’s eastern seaboard, M. coruscus had rarely been studied for its foot proteins. For better understanding of the gene composition and diversity mechanism of mfp3 and mfp6 from M. coruscus, PCR amplifications were executed with specific primers designed according to high conserved gene regions and the cDNA library of the foot glander cells as the template. Fourteen full-length cDNA sequences of mcofp3 variants and 8 of mcofp6 variants were obtained followed by sequence alignment and variation sites analysis. Notwithstanding its limitation, this study did build the foundation for further research on the adhesion mechanism of the foot proteins from M. coruscus and the development of related new adhesive.
